Dorfin is a multi-pass membrane, RING-IBR type, E3 ubiquitin-protein ligase. It is widely expressed with highest levels found in heart and ubiquitous expression throughout the central nervous system. Dorfin functions by accepting ubiquitin in the form of a thioester from UBCH7 and UBC8 and then transferring it to the targeted substrates. Dorfin is responsible for ubiquitylating synphilin-1, CaSR and mutant variants of SOD-1, a protein at fault for familial ALS (amyotrophic lateral sclerosis). Dorfin physically interacts with VCP (Valosin-containing protein) via its C-terminus. Together these two proteins are associated with the formation of ubiquitylated inclusions (UBIs) that characterize many neurodegenerative disorders, such as Parkinson's disease and ALS. This association with UBIs suggests that Dorfin plays an important role in the disease process.

The protein encoded by this gene shares strong similarity with Saccharomyces cerevisiae protein Cdc27, and the gene product of Schizosaccharomyces pombe nuc 2. This protein is a component of anaphase-promoting complex (APC), which is composed of eight protein subunits and highly conserved in eucaryotic cells. APC catalyses the formation of cyclin B-ubiquitin conjugate that is responsible for the ubiquitin-mediated proteolysis of B-type cyclins. This protein and 3 other members of the APC complex contain the TPR (tetratricopeptide repeat), a protein domain important for protein-protein interaction. This protein was shown to interact with mitotic checkpoint proteins including Mad2, p55CDC and BUBR1, and thus may be involved in controlling the timing of mitosis. Alternatively spliced transcript variants encoding different isoforms have been found for this gene.

ZPI, also known as SERPINA10 (serpin peptidase inhibitor, clade A (alpha-1 antiproteinase, antitrypsin), member 10) or PZI, is a 444 amino acid secreted protein that functions as a Protein Z-dependent protease inhibitor. Expressed by the liver, ZPI is secreted into the plasma where, in the presence of calcium, Protein Z and phospholipids, it inhibits the activated pro-coagulation factors X and XI (Factor X and Factor XI). This inhibition helps properly regulate intravenous blood clotting. ZPI, a member of the serpin protein family, contains five potential N-linked glycosylation sites and a tyrosine at position 387 which, when disrupted, renders ZPI inactive. Defects in the gene encoding ZPI may increase susceptibility to venous thrombosis, the formation of blood clots within a vein.

The amphoterin-induced gene and ORF (AMIGO) family of proteins consists of AMIGO-1, AMIGO-2 and AMIGO-3. All three members are single pass type I membrane proteins that contain several leucine-rich repeats, one IgG domain, and a transmembrane domain. The AMIGO proteins are specifically expressed on fiber tracts of neuronal tissues and participate in their formation. The AMIGO proteins can form complexes with each other, but can also bind itself. AMIGO-1, also designated Alivin-2, promotes growth and fasciculation of neurites and plays a role in myelination and fasciculation of developing neural axons. In cerebellar neurons, AMIGO-2 (Alivin-1) is crucial for depolarization-dependent survival. Similar to AMIGO-1 and AMIGO-2, AMIGO-3 (Alivin-3) plays a role in homophilic and/or heterophilic cell-cell interaction and signal transduction.
