FGF-2 (Basic fibroblast growth factor) belongs to the FGF superfamily that is made up of 22 different fibroblast growth factor genes. FGF-2 and FGF-1 (acidic FGF) are unique in that they do not follow the conventional signal sequence for secretion. FGF-2 is found in five isoforms via alternative initiation of translation. Signalling of FGF-2 occurs through the high-affinity tyrosine kinase receptors FGFR1-4. FGF-2 is a potent wide-spectrum mitogen whose overexpression is associated with immortalisation and unregulated cell proliferation in many tumors. FGF-2 functions in angiogenesis, cell survival, tissue repair, embryonic development and inflammation. FGF-2 is widely used to maintain the self-renewal of human embryonic stem cells (ESCs) and to induce pluripotent stem cells. FGF-2 is an established neurogenic factor for proliferation and differentiation of multipotent neural stem cells both during development and in the adult mouse brain as well.

The NMDAR plays an essential role in memory, neuronal development and it has also been implicated in several disorders of the central nervous system including Alzheimer’s disease, epilepsy and ischemic neuronal cell death. Increased membrane surface expression of the NR1 subunit of the receptor has been associated with synaptic plasticity. There are a number of different splice variants of the NR1. Differential splicing of three exons in the NR1 subunit generates up to eight NR1 splice variants and 7 of these have been identified in cDNA libraries. These exons encode a 21 amino acid N-terminal domain (N1) and adjacent sequences in the C-terminus (C1 and C2). Splicing out the C2 cassette eliminates the first stop codon and produces a new reading frame that generates a new sequence of 22 amino acids (C2'). Considerable attention has been focused on the distribution and expression of these splice variants that may affect the functional properties and regulation of the NMDAR.

The Urokinase Type Plasminogen Activator (uPA) receptor (uPAR) is a widely expressed receptor for urokinase plasminogen activator (uPA) and pro-uPA. uPAR / CD87 is a highly glycosylated, 55-60kDa integral membrane protein linked to the plasma membrane by a glycosylphosphatidylinositol (GPI) anchor. uPAR is expressed by T-cells, NK cells, monocytes, and neutrophils as well as non-hematopoietic cells that include vascular endothelial cells, fibroblasts, smooth muscle cells, keratinocytes, placental trophoblasts, hepatocytes, and a wide variety of tumor cells (including breast, colon, and prostate carcinoma, melanoma). It plays a critical role in the regulation of cell-surface plasminogen activation in physiological and pathological conditions, and it is also involved in cellular adhesion, the transmission of extracellular signals across the plasma membrane and the subsequent regulation of gene expression. uPAR has been implicated in several biological processes including angiogenesis, monocyte migration, cancer metastasis, trophoblast implantation, and wound healing. Human uPAR is encoded as a 313 amino acid residue polypeptide, excluding a 22 residue signal peptide and shows 60-70% similarity with the murine uPAR amino acid sequence although binding of uPA to uPAR shows strong species specificity.

D-Biotin is a growth factor present in small amounts in every living cell. It is involved in naturally occurring carboxylation reactions. It occurs mainly bound to proteins or polypeptides. It is more abundant in the liver, kidney, pancreas, yeast and milk. Biotin levels are higher in cancerous tumors than in normal tissues. It is inactivated by binding to avidin. D-Biotin may be used to elute proteins from avidin/streptavidin resins. It is widely used for dietary supplements and fortified foods. It is also used for tablets and hard-shell capsule preparation due to its pharmaceutical properties. Essential vitamin that is important for amino acid and energy metabolism, and fatty acid synthesis. It is a prosthetic group in four mammalian carboxylase families and facilitates the binding and transfer of carbon dioxide. Soluble in water (22 mg/100 mL), ethanol (80 mg/100 mL), more soluble in hot water and in dilute alkalies; insoluble in other common organic solvents. Soluble in 2 M Ammonium hydroxide (50 mg/mL - clear, colorless solution), dimethylformamide (1.7 mg/mL).

Applications:
Urea is used for the denaturation of proteins and as a mild solubilization agent for insoluble or denatured proteins. Useful for renaturing proteins from samples already denatured with 6 M guanidine chloride such as inclusion bodies and in the extraction of the mitochondrial complex. It is commonly used to solubilize and denature proteins for denaturing isoelectric focusing and two-dimensional electrophoresis and in acetic acid-urea PAGE gels. May be used with guanidine hydrochloride and dithiothreitrol (DTT) in the refolding of denatured proteins into their native or active form. Urea is used in cell or tissue culture media to increase the osmolality. Urea has also been used as fertilizer because of the easy availability of nitrogen; in animal feeds; it is reacted with aldehydes to make resins and plastics; condensed with malonic ester to form barbituric acid; used in the paper industry to soften cellulose. Biochem/physiol Actions: Urea has been shown to act as an aldosterone antagonist in the development of peanut agglutinin binding in cultured embryonic renal collecting duct epithelial cells. Urea is the principal end product of nitrogen metabolism in most mammals, formed by the enzymatic reactions of the Kreb's cycle. Typical Working Concentration: The use of 2 g/L urea in the culture of Kluyveromyces marxianus to produce a thermostable extracellular lipase has been described. Urea is typically used at a concentration of 8 M for protein denaturation or solubilization. A final concentration of 5 M urea is commonly used in molecular biology for sequencing gels. To prevent carbamylation, do not heat urea containing buffers above 37 °C

Receptors for the Fc region of immunoglobin G (Fc gamma R) are divided into three classes and Fc gamma RIII is a multifunctional, low/intermediate affinity receptor. In humans, Fc gamma RIII is expressed as two distinct forms (Fc gamma RIIIA and Fc gamma RIIIB) that are encoded by two different but highly homologous genes in a cell type-specific manner. Fc gamma RIIIB is a low-affinity, GPI-linked receptor expressed by neutrophils and eosinophils, whereas Fc gamma RIIIA is an intermediate affinity polypeptide-anchored transmembrane glycoprotein expressed by a subset of T lymphocytes, natural killer (NK) cells, monocytes, and macrophages. The Fc gamma RIIIA receptor is involved in phagocytosis, secretion of enzymes, inflammatory mediators, antibody-dependent cellular cytotoxicity (ADCC), mast cell degranulation, and clearance of immune complexes. Fc gamma RIIIA has an immunoreceptor tyrosine-based activation motif (ITAM) in its cytoplasmic domain and delivers an activation signal in the immune responses. Aberrant expression or mutations in this gene is implicated in susceptibility to recurrent viral infections, systemic lupus erythematosus, and alloimmune neonatal neutropenia. In humans, it is a 50 -70 kD type I transmembrane activating receptor.
