Human serum albumin (HSA), the most abundant protein in human blood plasma, is essential for maintaining osmotic pressure. It is produced in the liver, consists of a single polypeptide chain, with 585 amino acids with 17 tyrosil residues and one tryptophan located in position 214. HSA is organised in three domains, I, II and III, each consisting of two subdomains, A and B. In the physiological states, HSA occurs in two forms – the non-modified HSA with a free thiol group of Cys-34, and the modified (oxidised) form (oHSA), also called human mercaptoalbumin (HMA) and human nonmercaptoalbumin (HNA), respectively. HMA and HNA are in equilibrium, depending on the redox state of Cys-34, and their ratio also varies depending on age and the diseased state. HSA functions primarily as a carrier protein for drugs, steroids, fatty acids, and thyroid hormones, and plays a role in stabilising extracellular fluid volume.

Matrix Metalloproteinase-1 (MMP-1) is expressed by fibroblasts, keratinocytes, endothelial cells, monocytes and macrophages. MMP1 contains several distinct domains: a prodomain that is cleaved upon activation, a catalytic domain containing the zinc binding site, a short hinge region, and a carboxyl terminal (hemopexin like) domain. MMP-1 can degrade a broad range of substrates including types I, II, III, VII, VIII, and X collagens as well as casein, gelatin, alpha1 antitrypsin, myelin basic protein, L-Selectin, pro-TNF, IL1, IGFBP3, IGFBP5, pro-MMP2, and pro-MMP9. A significant role of MMP1 is the degradation of fibrillar collagens in extracellular matrix remodeling, characterized by the cleavage of the interstitial collagen triple helix into 3/4, 1/4 fragments. MMP1 may also be involved in enzyme cascades, cytokine regulation and cell surface molecule modulation.

These receptors share a characteristic molecular structure of two immunoglobulin-like and two fibronectin type III repeats and have been best Characterised for their roles in immune regulation, fertility, thrombosis and phagocytosis. Gas6 and protein S have been identified as ligands for these receptors. Gas6 binding induces tyrosine phosphorylation and downstream signallling pathways that can lead to cell proliferation, migration, or the prevention of apoptosis. Tyro3 and Axl play important regulatory roles in a variety of tissues, including the central nervous, reproductive, immune, and vascular systems. Tyro3 is widely expressed during embryonic development and preferentially expressed during neurogenesis in the central nervous system.

NIPSNAP2 Antibody: The NIPSNAP proteins comprise a family of evolutionarily well-conserved proteins with strong sequence similarity to the central portion of a protein encoded by C. elegans chromosome III between a 4-nitrophenylphosphatase (NIP) domain and non-neuronal SNAP25-like protein. NIPSNAP2, a novel gene encoding a protein with tyrosine phosphorylation sites and a transmembrane domain, is co-amplified with EGFR in approximately 40% of glioblastomas, the most common and malignant form of central nervous system tumors. It is widely expressed and most abundant in heart and skeletal muscle. NIPSNAP proteins have been suggested to be important in vesicular transport. NIPSNAP2 antibody is predicted to not cross-react with any other members of the NIPSNAP protein family.

Human Chemokine (C-X-C motif) Ligand 7 (CXCL7), also known as neutrophil activating peptide 2 (NAP-2), is a member of the CXC chemokines containing an ELR domain (Glu-Leu-Arg tripeptide motif). Similar to other ELR domain containing CXC chemokines, such as IL-8 and the GRO proteins, CXCL7 binds CXCR2, chemoattracts and activates neutrophils. CXCL7, Connective Tissue Activating Protein III (CTAPIII) and beta thrombogulin ( beta TG), are proteolytically processed carboxylterminal fragments of platelet basic protein (PBP) which is found in the alphagranules of human platelets. Although CTAPIII, beta TG, and PBP represent amino-terminal extended variants of NAP2 and possess the same CXC chemokine domains, these proteins do not exhibit CXCL7/NAP2 activity. CXCL7 induces cell migration through the G-protein-linked receptor CXCR-2.

Members of this family are involved in a number of cellular functions including initiation of translation, RNA splicing, and ribosome assembly. A cluster of genes, BAT1-BAT5, has been localized in the vicinity of the genes for TNF alpha and TNF beta. These genes are all within the human major histocompatibility complex class III region. This protein is a negative regulator of inflammation. It is also thought to be a translation initiation factor. This gene is a strong candidate gene for rheumatoid arthritis. There are multiple alternatively spliced transcript variants known for this gene but only two have been fully described. Both of these variants encode the same isoform. This gene has been found to have multiple polyadenylation sites.

Glial Fibrillary Acidic Protein (GFAP) was discovered by Amico Bignami and co-workers as a major fibrous protein of multiple sclerosis plaques. It was subsequently found to be a member of the 10 nm or intermediate filament (IF) family, specifically the IF family Class III, which also includes peripherin, desmin and vimentin. GFAP is strongly and specifically expressed in astrocytes and certain other astroglia in the CNS, in satellite cells, peripheral ganglia, and in non-myelinating Schwann cells in peripheral nerves. In many damage and disease states GFAP expression is heavily upregulated in astrocytes. In addition, neural stem cells frequently strongly express GFAP. Point mutations in the protein coding region of the GFAP gene lead to Alexander disease which is characterized by the presence of abnormal astrocytes containing GFAP protein aggregates known as Rosenthal fibers.

The neural adhesion molecule Contactin-6, also known as NB-3, is a contactin/F3 subgroup member of immunoglobulin superfamily. It is expressed exclusively in the nervous system and mainly upregulated at the early postnatal stage during mouse brain development. Employing Northern blot analysis Kamei et al found that amongst different regions of the adult human nervous system cerebellum expressed highest level of NB-3 mRNA. The expression of NB-3 in the cerebellum increases until adulthood. In contrast, the expression in the cerebrum declines to a low level after postnatal day 7. NB-3 like other neural recognition molecules plays a vitally important role in axonal guidance during development, plasticity, and maintenance of synaptic connections in the adult brain. Cui et al recently showed that NB-3 acts as a novel Notch ligand to participate in oligodendrocyte generation. Furthermore, NB-3 triggers nuclear translocation of the Notch intracellular domain and promotes oligodendrogliogenesis from progenitor cells and differentiation of oligodendrocyte precursor cells via Deltex1. In primary oligodendrocytes, NB-3 increases myelin-associated glycoprotein transcripts. Hence, the NB-3/Notch signaling pathway may be worthwhile a closer examination for its potential for the treatment of demyelinating diseases. Human NB-3 shares with rat NB-3 86% identity in nucleotide sequences and 90% identity in amino acid sequences. FUNCTION: Contactins mediate cell surface interactions during nervous system development. Participates in oligodendrocytes generation by acting as a ligand of NOTCH1. Its association with NOTCH1 promotes NOTCH1 activation through the released notch intracellular domain (NICD) and subsequent translocation to the nucleus. Involved in motor coordination. SUBCELLULAR LOCATION: Cell membrane; lipid-anchor; GPI-anchor. ALTERNATIVE PRODUCTS: 2 named isoforms produced by alternative splicing. TISSUE SPECIFICITY: Expressed in brain. In brain, it is preferentially expressed in the accessory olfactory bulb, layers II/III and V of the cerebral cortex, piriform cortex, anterior thalamic nuclei, locus coeruleus of the pons and mesencephalic trigeminal nucleus and in Purkinje cells of the cerebellum. DEVELOPMENTAL STAGE: Highly expressed after birth, reaching a maximum at the postnatal day 7, and declines thereafter in the cerebrum, whereas it increases in the cerebellum to adulthood.
