Beschreibung:   The Alpha-, Beta-, Gamma-, and Delta -catenins are proteins that bind to the highly conserved, intracellular cytoplasmic tail of E-cadherin. Together, the catenin/cadherin complexes play an important role mediating cellular adhesion. Alpha-catenin interacts with E-cadherin associated protein and also associates with other members of the cadherin family, such as N-cadherin and P-cadherin. Beta-catenin associates with the cytoplasmic portion of E-cadherin, which is necessary for the function of E-cadherin as an adhesion molecule. Beta-catenin also complexes with the tumor suppressor protein APC. Gamma-catenin, also known as plakoglobin, is a protein that binds ?catenin and N-cadherin. Gamma-catenin interacts with presenilin 1 and is expressed in the brain. The gene encoding Gamma-catenin maps to human chromosome 5p15.2. A hemizygous loss of the gene encoding Gamma-catenin leads to the mental retardation associated with Cri-du-Chat syndrome. In addition, the transmembrane phosphatase PTPm associates with catenin/ cadherin complexes and may regulate complex signaling.

Beschreibung:   Guanine nucleotide-binding proteins (G proteins) are involved as a modulator or transducer in various transmembrane signaling systems. The beta and gamma chains are required for the GTPase activity, for replacement of GDP by GTP, and for G protein-effector interaction. Plays a role in the regulation of adenylyl cyclase signaling in certain regions of the brain. Plays a role in the formation or stabilzation of a G protein heterotrimer (G(olf) subunit alpha-beta-gamma-7) that is required for adenylyl cyclase activity in the striatum.

Beschreibung:   Protocadherins are a large family of cadherin-like cell adhesion proteins that are involved in the establishment and maintenance of neuronal connections in the brain. There are three protocadherin gene clusters, designated alpha, beta and gamma, all of which contain multiple tandemly arranged genes. PCDHB14 (Protocadherin beta 14) is a 798 amino acid protein that is one of 16 proteins in the protocadherin beta cluster. Unlike the alpha and gamma gene clusters whos genes are spliced to downstream constant region exons during transcription, members of the beta cluster (such as PCDHB14) do not use constant-region exons to produce mRNAs. As a result, each protocadherin beta gene encodes the transmembrane, extracellular and short cytoplasmic domains of the protein. Localized to the cell membrane, PCDHB14 is a single-pass type I membrane protein that contains six cadherin domains.

Beschreibung:   Enzymes of the phospholipase C family catalyze the hydrolysis of phospholipids to yield diacylglycerols and water soluble phosphorylated derivatives of the lipid head groups. A number of these enzymes have specificity for phosphoinositides. Of the phosphoinositide specific phospholipase C enzymes, C beta is regulated by heterotrimeric G protein coupled receptors, while the closely related C gamma 1 and C gamma 2 enzymes are controlled by receptor tyrosine kinases. The C gamma 1 and C gamma 2 enzymes are composed of phospholipase domains that flank regions of homology to noncatalytic domains of the SRC oncogene product, SH2 and SH3.

Beschreibung:   Mitochondrial ATP synthases (ATPases) transduce the energy contained in membrane electrochemical proton gradients into the energy required for synthesis of high-energy phosphate bonds. ATPases contain two linked complexes: F1, the hydrophilic catalytic core; and F0, the membrane-embedded protein channel. F1 consists of three Alpha chains and three Beta chains, which are weakly homologous, as well as one Gamma chain, one Delta chain and one Gamma chain. F0 consists of three subunits: a, b and c. A mitochondrial F1-ATPase inhibitor protein, ATPIF1 (ATPase inhibitory factor 1), also known as IP, IF1, ATPI or ATPIP (ATPase inhibitor protein), binds to the C-terminal region of a Beta subunit of the F1-ATPase at low pH values and, via interference of the Beta and Gamma subunit interaction, ATPIF1 regulates the activity of the F1F0-ATPase. This reversible ATPIF1 binding to F1F0-ATPase also occurs on the surface of endothelial cells.
