The 2H7 antibody reacts with human CD20, expressed by pre-B cells, resting and activated B cells, follicular dendritic cells and a small subset of T cells, but not on plasma cells. CD20 is a 33-37 kDa transmembrane protein which is differentially phosphorylated in resting and active B cells. The CD20 molecule plays a role in regulating B-cell activation. The 2H7 antibody is reported to be cross-reactive with several non-human species including Baboon, Chimpanzee, Cynomolgus and Rhesus. Please choose the appropriate format for each application.

The stain uses an enhanced Periodic Acid-Schiff (PAS) method for detection of glycoprotein sugars. The supplied agent first oxidises the cis-diol sugar groups to aldehydes. The aldehyde groups react with the sensitive glycoprotein stain solution forming Schiff bonds and producing strong magenta colour bands. RAPIDstain™ can be used after glycoprotein staining to detect non-glycosylated proteins and the use of the stain enhances glycoprotein staining. The retinal pigment epithelium (RPE) is a monolayer simple epithelium in proximity to the outer surface of the retinal photoreceptor cells. Retinal pigment epithelium-specific protein (RPE65) is a 65 kDa protein belonging to the β-carotene dioxygenase family. This protein is important in 11-cis retinal production as well as in visual pigment regeneration. RPE65 is attached to the membrane by a lipid anchor when palmitoylated (membrane form) and soluble when unpalmitoylated. The soluble form of the protein binds vitamin A. Defects in RPE65 causes autosomal dominant retinitis pigmentosa and/or Leber congenital amaurosis type 2.

B lymphocytes marker CD20 is a non glycosylated protein with a molecular weight of 35 or 37 kDa depending on the degree of phosphorylation. Although not a member of the tetraspanin superfamily of cell surface receptors, it crosses the cell membrane four times. The CD20 antigen is present on human pre B lymphocytes and on B lymphocytes at all stages of maturation, except on plasma cells. Low level expression of the CD20 antigen has been detected on normal T lymphocytes. The CD20 molecule is involved in regulation of B cell differentiation, presumably via its reported function as a Ca++ channel subunit.

SHP1 is a member of the non receptor protein tyrosine phosphatase subfamily. The PTP family comprises of at least 37 proteins, characterized by a catalytic phosphatase domain of approximately 240 amino acids, and includes both transmembrane and cytosolic enzymes. PTP1B is cytosolic. The PTPs have high substrate specificity for phosphotyrosyl proteins, at the primary sequence level sharing little similarity with the protein serine phosphatases, protein threonine phosphatases, or the acid and alkaline phosphatases. SHP1 is implicated in the control of tyrosine kinase signalling pathways in cellular proliferation, with a potential role in cancer.
