CHRDL2 is a novel chordin like protein that can act as a BMP antagonist. Amember of the chordin family of proteins, it contains a signal peptide andthree CR (cysteine-rich repeat) domains. When expressed as a recombinantprotein it is secreted and binds to activin A, but not to BMP-2, -4, -6. Differential expression has been detected in developing chondrocytes, myoblasts, osteoblasts, and osteoarthritic joints. Complex alternative splicing of CHRDL2 potentially results in distinct isoforms that differ at their C termini, in the expression of signal peptide, and in the content of CR domains. CHRDL2 was originally characterized as a novel protein exclusively expressed in breast, lung, and colon tumors.

WD-repeat proteins are a large family of eukaryotic proteins coordinating multi-protein complex assemblies. Their role has been implicated in multiple cellular processes including signal transduction, transcriptional regulation, cell cycle control and apoptosis. NRIP is a novel 860a.a nuclear protein consisting of seven conserved WD40 domains and one NLS motif. It binds to androgen and glucocorticoid receptors and up-regulates their transcriptional activity, thereby functioning as a nuclear receptor co-activator. Role of NRIP has been implicated in cell growth and also in cervical and prostrate cancer, thus indicating a potential therapeutic intervention. Northern Blot analysis detects a high expression of NRIP in skeletal muscle and testis and low expression in heart, prostrate and adrenal gland.

Nudel is important for normal cortical development. It is invovled in microtubule organization, nuclear translocation, and neuronal positioning in concert with various other factors (including Lis1, Pafah1b1, Pahfah1b2, dynein, dynorphin A and cdk5). Western blot analysis of mouse tissues shows abundant expression of Nudel in brain and testis, and much lower expression in heart, liver, kidney, and skeletal muscle. In fractionated rat brain, Nudel and Lis1 are both found in fractions enriched for postsynaptic density proteins. Immunostaining of embryonic day 18 mouse brain sections revealed staining of migrating neurons and thalamocortical axons of the intermediate zone of the developing cerebral cortex, as well as several other developing brain regions. The deduced protein contains 345 amino acids and has a calculated molecular mass of 38.4 kDa. It has a coiled coil motif (residues 19 to 201), followed by several potential phosphorylation sites for casein kinase II, protein kinase C or CDK5. Nudel shares about 50% identity with mouse and human NUDE proteins.

The mature form of Factor X (Factor X A) is generated by Factor IX A- or Factor VII A-mediated cleavage at the tripeptide sequence, Arg-Lys-Arg, to yield a disulfide linked dimer. Together with the cofactor Factor V A and Ca2+ on the surface of platelets or endothelial cells, Factor X A coordinates as part of the prothrombinase complex, which mediates proteolysis of Prothrombin into active Thrombin. Mutations at the Factor X locus resulting in Factor X deficiencies can contribute to hemorrhagic diathesis.
