The RING-type zinc finger motif is present in a number of viral and eukaryotic proteins and is made of a conserved cysteine-rich domain that is able to bind two zinc atoms. Proteins that contain this conserved domain are generally involved in the ubiquitination pathway of protein degradation. RNF32 (RING finger protein 32), also known as HSD15 or FKSG33, is a 362 amino acid cytoplasmic protein that contains one IQ domain and two RING-type zinc fingers. Highly expressed in testis with lower expression levels in ovary tissue, RNF32 is thought to play a role in spermatogenesis, specifically contributing to the growth and maturation of round spermatids. Six isoforms of RNF32 exist due to alternative splicing events.

BZW2, also known as HSPC028 or MSTP017, is a 419 amino acid protein that contains one W2 domain and is thought to be involved in neuronal differentiation. The gene encoding BZW2 maps to human chromosome 7. Chromosome 7 houses over 1,000 genes and comprises nearly 5% of the human genome. Defects in some of the genes localized to chromosome 7 have been linked to Osteogenesis imperfecta, Pendred syndrome, Lissencephaly, Citrullinemia and Shwachman-Diamond syndrome. The deletion of a portion of the q arm of chromosome 7 is associated with Williams-Beuren syndrome, a condition characterized by mild mental retardation, an unusual comfort and friendliness with strangers and an elfin appearance. Deletions of portions of the q arm of chromosome 7 are also seen in a number of myeloid disorders, including cases of acute myelogenous leukemia and myelodysplasia.

Prickle1 is an 831 amino acid protein that contains one PET domain and three LIM zinc-binding domains and localizes to the cytoplasm, as well as to the nuclear membrane. Expressed at higher levels in placenta and at lower levels in liver, brain, kidney, lung and pancreas, Prickle1 is thought to function as a nuclear receptor that interacts with NRSF, a silencer protein that binds the DNA sequence element NRSE (neuron-restrictive silencer element). Defects in the gene encoding Prickle1 are associated with autosomal recessive progressive myoclonic epilepsy-1B, which is characterized by quick jerks of the arms, shoulders or legs. The gene encoding Prickle1 maps to human chromosome 12, which encodes over 1,100 genes and comprises approximately 4.5% of the human genome.

SPPL2b is a 592 amino acid multi-pass membrane protein that contains one protease associated domain and belongs to the GXGD family of aspartic proteases. Localized to the membrane of the Golgi apparatus and to lysosomes, endosomes and the plasma membrane, SPPL2b functions as an intramembrane protease that specifically cleaves the transmembrane domain of TNF? Once cleaved, TNF?releases its intracellular domain, thus triggering the immunity-related expression of cytokines (also known as interleukins) which are used in various pathways throughout the body. SPPL2b also interacts with and catalyzes the intramembrane proteolysis of ITM2B (integral membrane protein 2B), a protein that, when mutated, is associated with dementia. Four isoforms of SPPL2b exist due to alternative splicing events.

Mono-ADP-ribosylation is one of the posttranslational protein modifications regulating cellular metabolism (e.g. nitrogen fixation) in prokaryotes. Mono-ADP-ribosylation is a process in which the ADP-ribose moiety of nicotinamide adenine dinucleotide is transferred to an acceptor amino acid. Five mammalian ADP-ribosyltransferases (ART1-ART5) have been cloned, and expression is restricted to tissues such as cardiac and skeletal muscle, leukocytes, brain and testis. ART3 (ADP-ribosyltransferase 3), also known as Ecto-ADP-ribosyltransferase 3, is a testis specific membrane protein that does not appear to have ADP-ribosyltransferase activity. It lacks the R-S-EXE active site motif and is therefore unable to catalyze the reaction. ART3 is predominantly found in spermatocytes and may play a role in spermatogenesis.

MYBPC3 encodes the cardiac isoform of the thick-filament myosin-binding protein C. It is found in the crossbridge-bearing zone (C region) of A bands in vertebrate striated muscle. Regulatory phosphorylation of MYBPC3 by cAMP-dependent protein kinase (PKA) upon adrenergic stimulation may be linked to modulation of cardiac contraction. MYBPC3 binds F-Actin, MHC and native thin filaments, and modifies the activity of Actin-activated myosin ATPase. Mutations in the MYBPC3 gene lead mainly to truncation of the protein, which results in one cause of familial hypertrophic cardiomyopathy type 4 (CMH4), a heart disorder characterised by ventricular hypertrophy, which often involves the interventricular septum and is usually asymmetric. The MYBPC3 gene maps to chromosome 11p11.2.

DHRS1 (dehydrogenase/reductase (SDR family) member 1), also known as SDR19C1, is a 313 amino acid protein that belongs to the short-chain dehydrogenases/reductases (SDR) family and likely functions as an oxidoreductase. Abundantly expressed in heart and liver, DHRS1 contains an SDR motif and is encoded by a gene that maps to human chromosome 14q12. Human chromosome 14 houses over 700 genes and comprises nearly 3.5% of the human genome. Chromosome 14 encodes the presinilin 1 (PSEN1) gene, which is one of the three key genes associated with the development of Alzheimer's disease (AD). The SERPINA1 gene is also located on chromosome 14 and, when defective, leads to the genetic disorder ?-antitrypsin deficiency, which is characterized by severe lung complications and liver dysfunction.

One of the major pre-mRNA-binding proteins. Binds tenaciously to poly(C) sequences. Likely to play a role in the nuclear metabolism of hnRNAs, particularly for pre-mRNAs that contain cytidine-rich sequences. Can also bind poly(C) single-stranded DNA. Plays an important role in p53/TP53 response to DNA damage, acting at the level of both transcription activation and repression. When sumoylated, acts as a transcriptional coactivator of p53/TP53, playing a role in p21/CDKN1A and 14-3-3 sigma/SFN induction (By similarity). As far as transcription repression is concerned, acts by interacting with long intergenic RNA p21 (lincRNA-p21), a non-coding RNA induced by p53/TP53. This interaction is necessary for the induction of apoptosis, but not cell cycle arrest.

Belonging to the F-box family of proteins, FBXO45 is a 286 amino acid protein that contains one C-terminal F-box domain. F-box proteins are critical components of the SCF (Skp1-CUL-1-F-box protein)-type E3 ubiquitin ligase complex and are involved in substrate recognition and protein recruitment for ubiquitination. They are members of a larger family of proteins that are involved in the regulation of a wide variety of cellular mechanisms, including the cell cycle, the immune response, signaling cascades and developmental processes. They function by targeting proteins, such as cyclins, cyclin-dependent kinase inhibitors, I˚B-å and ∫-catenin, for degradation by the proteasome after ubiquitination. Via its F-box domain, FBXO45 can directly interact with Skp1 p19 and CUL-1. FBXO45 has been shown to be an estrogen-induced gene.

ADCK2 (aarF domain containing kinase 2), also known as AARF, is a 626 amino acid single-pass membrane protein belonging to the protein kinase superfamily and the ADCK protein kinase family. The ADCK family consists of five paralogs in human (ADCK1-5). Encoded by a gene that maps to human chromosome 7q34, ADCK2 contains one protein kinase domain. ADCK2 participates in ATP and nucleotide binding, transferase functions and protein serine/threonine kinase activities. Expression of ADCK2 inversely correlates with cellular viability, suggesting elevated expression of ADCK2 may be essential for tumour survival. ADCK2 is necessary for cell proliferation of glioblastoma multiforme (GBM), a fatal primary brain tumor containing countless genetic and epigenetic alterations.
