Green fluorescence protein (GFP) is a 27 kDa protein derived from the jellyfish Aequorea victoria, which emits green light (emission peak at a wavelenth of 509 nm) when excited by blue light (excitation peak at a wavelenth of 395 nm). Green Fluorescent Protein (GFP) has become an invaluable tool in cell biology research, since its intrinsic fluorescence can be visualized in living cells. GFP fluorescence is stable under fixation conditions and suitable for a variety of applications. GFP has been widely used as a reporter for gene expression, enabling researchers to visualize and localize GFP-tagged proteins within living cells without the need for chemical staining. Other applications of GFP include assessment of protein protein interactions through the yeast two hybrid system and measurement of distance between proteins through fluorescence energy transfer (FRET) protocols. GFP technnology has considerably contributed to a greater understanding of cellular physiology.

Nocturnin is a 431 amino acid circadian deadenylase protein that is expressed in a broad range of tissues with greatest abundance in the liver, kidney and testis. Nocturnin plays a role in circadian regulation as well as diet-induced obesity. The mRNA abundance of Nocturnin exhibits circadian rhythmicity, peaking after dusk in photoreceptors, spleen, heart, kidney and liver. Nocturnin is thought to be responisble for turning off genes that are involved in circadian regulation. In Xenopus retinal photoreceptor cells, the rhythmic regulation of Nocturnin is thought to be controlled by phosphorylated CREB. Mice lacking Nocturnin remain lean on high fat diets with a reduction in visceral fat, which suggests that this protein may also be responsible for lipid metabolism and fat storage.

Cyclin dependent kinases are positively regulated by association with cyclins and negatively regulated by binding to inhibitory subunits. The activity of cyclin dependent kinases is also regulated by the phosphorylation status, which is controlled by the antagonistic action of Wee1 kinase and CDC25 phosphatases. Three CDC25 genes are present in human cells: CDC25A, CDC25B, and CDC25C. These three genes function at different phases of the cell cycle. Whereas CDC25A and CDC25B are expressed throughout the cell cycle, with peak expression in G1 for CDC25A and in both G1 S phase and G2 for CDC25B, CDC25C is predominantly expressed in G2.

The mini-chromosome maintenance (MCM) family of proteins, including MCM2, MCM3, MCM4 (Cdc21), MCM5 (Cdc46), MCM6 (Mis5) and MCM7 (Cdc47), are regulators of DNA replication that act to ensure replication occurs only once in the cell cycle. Expression of MCM proteins increases during cell growth, peaking at G1 to S phase. The MCM proteins each contain an ATP-binding motif, which is predicted to mediate ATP-dependent opening of double-stranded DNA. MCM proteins are regulated by E2F transcription factors, which induce MCM expression, and by protein kinases, which interact with MCM proteins to maintain the postreplicative state of the cell. MCM2/MCM4 complexes function as substrates for Cdc2/cyclin B in vitro. Cleavage of MCM3, which can be prevented by caspase inhibitors, results in the inactivation of the MCM complex (composed of at least MCM proteins 2-6) during apoptosis.

Recognizes a protein of 70 kDa, which is identified as CD86 (HLDA V; WS Code BP BP072. HLDA V; WS Code A A109. HLDA VI; WS Code BP 95. HLDA VI; WS Code B CD86.9). CD86 is a type I transmembrane glycoprotein and a member of the immunoglobulin superfamily of cell surface receptors. It is expressed at high levels on resting peripheral monocytes and dendritic cells and at very low density on resting B and T lymphocytes. CD86 expression is rapidly upregulated by B cell specific stimuli with peak expression at 18 to 42 hours after stimulation. CD86, along with CD80/B71, is an important accessory molecule in T cell co-stimulation via its interaction with CD28 and CD152/CTLA4. Since CD86 has rapid kinetics of induction, it is believed to be the major CD28 ligand expressed early in the immune response. It is also found on malignant Hodgkin and Reed Sternberg (HRS) cells in Hodgkin's disease.

CDCA7 is a nuclear protein without a known function, although its high homology with the transcription factor JPO2 or RAM2 suggests that it is also a transcription factor. CDCA7 is one of many target genes regulated by the c-Myc transcription factor. Overexpression of CDCA7 occurs in a significant proportion of cancers and it may play an important role in tumorigenesis. Normally it is periodically expressed in the cell cycle, peaking at the G1 to S phase transition.

Cyclin G2 may play a role in growth regulation and in negative regulation of cell cycle progesssion. Cyclin G2 expression levels increase through the cell cycle to peak in the mid/late-S phase and decrease during G2/M phase. It may also contribute in maintaining the quiescent state of differentiated cells. Cyclin G2 is similar to cyclin A in the cyclin box, although no kinase activity is detected.

Beschreibung:
Centrosomes are dynamic organelles involved in many aspects of cell function and growth. Centrosomes act as microtubule organizing centers, and provide a site for concerted regulation of cell cycle progression. Duplication of centrosomes occurs once during each cell cycle and requires proper mitotic spindle formation and chromosome segregation. Defects in centrosome duplication or function are linked to many human diseases, including various forms of cancer. The centrosome and spindle pole-associated protein 1 (CSPP1) interacts with centrosomes and microtubules and may play a role in the regulation of G(1)/S-phase progression and spindle assembly. Two isoforms of CSPP1 exist as a result of alternative splicing events. Isoform 1 expression increases throughout the cell cycle and peaks in G2/M phase, whereas isoform 2 expression is highest in G1 phase and decreases thereafter.
