Beschreibung:   Mouse SDC4 is a ubiquitous transmembrane proteoglycan which belongs to the syndecan proteoglycan family. SDC4 is a cell surface proteoglycan that bears heparan sulphate. The four vertebrate syndecans, Syndecan-1 through -4, have similar short cytoplasmic domains and extracellular portions that diverge, except for HS attachment sites. Structurally diverse side chains add considerably to the size of the core proteins and serve as binding sites for growth factors, cytokines, and extracellular matrix proteins. Syndecans are present as homodimers or multimers, and are often expressed in developmental and cell type-specific patterns. It is expressed highly in liver, kidney and lung. SDC4 localises to the focal adhesions of adherent cells and binds to a range of extracellular ligands, including growth factors and extracellular-matrix proteins. Through its extracellular domain, syndecan-4 cooperates with adhesion molecules and binds matrix components relevant for cell migration. As a heparan sulphate proteoglycan, SDC4 works as a coreceptor for various growth factors. Syn4 deficiency limits neointimal formation after vascular injury by regulating vascular smooth muscle cells (VSMCs) proliferation and vascular progenitor cells (VPCs) mobilisation. SDC4 have an array of functions including regulating cell growth, differentiation, and adhesion.

Beschreibung:   Cytotoxic and Regulatory T-Cell Molecule (CRTAM) is a member of Nectin family under the immunoglobulin superfamily that is expressed by activated CD8+ and NK T cells. CRTAM is found in spleen, thymus, small intestine, peripheral blood, and it is highly expressed by Purkinje cells of the cerebellum. CRTAM is a type I transmembrane glycoprotein containing one Ig-like C2-type domain and one Ig-like V-type domain in its extracellular domain, while its cytoplasmic region shows a potential class I PDZ domain. CRTAM is expressed as a homodimer on the cell surface but does not show homotypic binding in trans. The high affinity of CRTAM/IGSF4 adhesion allows CRTAM to disrupt IGSF4 homotypic interactions. IGSF4 and T cell receptor coengagement of CD8+ cells expressiong CRTAM induces increased IFN gamma or IL-22 production.

Beschreibung:   bHLH proteins are essential for neurogenesis, myogenesis, hematopoiesis and sex determination. HES1 (Hairy and Enhancer of Split 1) is a basic Helix-Loop-Helix (bHLH) transcriptional repressor regulated by the Notch pathway and expressed in various tissues. HES1 is well known in development of different tissues, particularly endoderm derived endocrine organs like pancreas and neuroendocrine cells of lung. HES1 controls normal development by regulating proliferation and differentiation and so cell fate. Notch signalling plays both oncogenic and tumour suppressor roles depending on cell type. In contrast to T cell acute lymphoblastic leukaemia (ALL), where Notch activation promotes leukemogenesis, induction of Notch signalling in B cell ALL (B-ALL) leads to growth arrest and apoptosis. The Notch target HES1 is sufficient to reproduce this tumour suppressor phenotype in B-ALL by interacting and activating PARP-1 and regulating proapoptotic signals.

Beschreibung:   Endothelial Protein C Receptor (EPCR) is a Vitamin K-dependent Serine Protease that plays a major role in blood coagulation. Binding of Protein C to EPCR leads to the proteolytic activation of PAR1 (Protease-Activated Receptor 1) on endothelial cells and subsequent up-regulation of Protein C-induced genes. EPCR is a type I transmembrane glycoprotein in the CD1/MHC family. It is expressed most strongly in the endothelial cells of arteries and veins in heart and lung. Membrane bound EPCR is released by metalloproteolytic cleavage to generate the soluble receptor. The extracellular domain of human and mouse EPCR shares approximately 61% amino acid sequence homology. EPCR plays an important role in augmenting Protein C activation by the Thrombin-Thrombomodulin complex and in regulating blood coagulation and inflammation. EPCR is found primarily on endothelial cells. Deletion of EPCR function results in embryonic death, at least in part due to placental thrombosis.

Beschreibung:   Interleukin-27 (IL-27) is a heterodimeric group 2 receptor ligand molecule that belongs to the IL-6/IL-12 family of long type I cytokines. It is composed of EBI3 (EBV-induced gene 3), a 34 kDa glycoprotein that is related to the p40 subunit of IL-12 and IL-23, and p28, the cloned 28 kDa glycoprotein that is related to the p35 chain of IL-12. IL-27 is expressed by monocytes, endothelial cells and dendritic cells. IL-27 binds to and signals through a heterodimeric receptor complex composed of WSX1 (TCCR) and gp130. Evidence suggests IL-27 interacts only with WSX-1. IL-27 has both anti- and proinflammatory properties. As an antiinflammatory, IL-27 seems to induce a general negative feedback program that limits T and NK-T cell activity. At the onset of infection, IL-27 induces an IL-12 receptor on naie CD4+ T cells, making them susceptible to subsequent IL-12 activity (and possible Th1 development). Notably, IL-12 family cytokines are both induced and inhibited by bacterial products. Microbes promote IL-27 secretion through TLR4, and also block IL-27 production via C5a induction.

IL-36 alpha has been detected in both neuronal and synovial tissue, whereas IL-36 beta and IL-36 gamma are expressed in both cutaneous and mucosal epithelial cells, including the respiratory tract. IL-36 beta and IL-36 gamma stimulate proliferation, maturation and/or cytokine expression by innate immune cells (such as keratinocytes and dendritic cells), and adaptive immune cells (neutrophils and T-cells) in both humans and mice. The activity of IL-36 alpha is mediated by interleukin 1 receptor-like 2 (IL1RL2/IL1R-rp2), and is specifically inhibited by interleukin 1 family, member 5 (IL1F5/IL-1 delta). IL-36 gamma plays an important role in communicating the cell death to surrounding cells.
