It has been suggested that this protein may be involved in the induction of apoptosis as it preferentially recognizes poly (A) homopolymers and induces DNA fragmentation in CTL targets. The major granule-associated species is a 15-kDa protein that is thought to be derived from the carboxyl terminus of the 40-kDa product by proteolytic processing. UCHL1 is a thiol protease that recognizes and hydrolyzes a peptide bond at the C-terminal glycine of ubiquitin. UCHL1 also binds to free monoubiquitin and may prevent its degradation in lysosomes. Mouse Alx3 is a homeobox gene that is related to the Drosophila aristaless gene and to a group of vertebrate genes including Prx1, Prx2, Cart1, and Alx4. Ubiquitin Carboxyl-Terminal Hydrolase 14 (USP14) belongs to the ubiquitin-specific processing (USP) family which is a deubiquitinating enzyme (DUB) with His and Cys domains. USP14 located in the cytoplasm is a proteasome-associated deubiquitinase which releases ubiquitin from the proteasome targeted ubiquitinated proteins. USP14 acts also as a physiological inhibitor of endoplasmic reticulum-associated degradation (ERAD) under the non-stressed condition by inhibiting the degradation of unfolded endoplasmic reticulum proteins via interaction with ERN1. Carboxypeptidase A1 (CPA1) is secreted as a pancreatic peptidase that comes from the precursor form of inactive procarboxypeptidase. CPA1 comprises a signal peptide, a pro region and a mature chain, and can be activated after cleavage of the pro peptide. It has a free C-terminal carboxyl group, with the preference of residues with aromatic or branched aliphatic side chains. CPA1 cleaves the C-terminal amide or ester bond of peptides and involves in zymogen inhibition. Three different forms of human pancreatic procarboxypeptidase A have been isolated. In contrast to procarboxypeptidase B which was always secreted by the pancreas as a monomer, procarboxypeptidase A occurs as a monomer and/or associated to one or two functionally different proteins, such as zymogen E.

Apoptosis Regulator BAX (BAX) belongs to the Bcl-2 family. BAX exists as a homodimer and is expressed in a wide variety of tissues. The Bax gene encodes different isoforms including Bax alpha (21 kDa) and Bax beta (24 kDa). Although both isoforms contain BH1, BH2 and BH3 domains, Bax beta has a unique carboxyl terminus and does not contain a hydrophobic transmembrane domain. BAX accelerates programmed cell death by binding to, and antagonizing the apoptosis repressor BCL2 or its adenovirus homolog E1B 19k protein. BAX also promotes activation of CASP3, and thereby apoptosis.

Chymotrypsin C (CTRC) is a member of the peptidase S1 family. CTRC is a serum calcium-decreasing factor that has chymotrypsin-like protease activity. CTRC has broad substrate specificity, but prefers tocleave on the carboxyl side of hydrophobic residues. CTRC is expressed primarily in the pancreas, and is secreted into the digestive tract. CTRC plays a protective role in the pancreas by mitigating premature trypsinogen activation through degradation. It has been proposed that CTRC is a key regulator of digestive zymogen activation and is a physiological coactivator of digestive carboxypeptidases proCPA1 and proCPA2. The mutation of CTRC gene encodes the digestive enzyme CTRC contribute to the development of pancreatitis.
