G protein-coupled receptors (GPRs or GPCRs), also known as seven transmembrane receptors, heptahelical receptors, or 7TM receptors, are members of the largest protein family and play a role in many different stimulus-response pathways. G-protein coupled receptors mediate extracellular signals into intracellular signals (G-protein activation). They respond to a great variety of signaling molecules, including hormones, neurotransmitters and other proteins and peptides. GPR proteins are integral seven-pass membrane proteins with some conserved amino acid regions. GPR19, an orphan receptor, shows elevated expression during embyronic development of the nervous sytem as well as in specific regions of adult mouse brain, including the olfactory bulb, the hippocampus, hypothalamic nuclei and the cerebellum. The GPR19 gene maps to a location on chromosome 12, which is a frequent target for rearrangement in cancer cells and involved in childhood acute lymphoblastic leukemia (ALL).

Highly expressed in brain tissue and present in lower amounts in placenta and spinal cord, ANKK1 functions to catalyze the ATP-dependent phosphorylation of target proteins and is thought to play a role in alcohol and nicotine dependence. The gene encoding ANKK1 maps to human chromosome 11, which houses over 1,400 genes and comprises nearly 4% of the human genome.

The Major facilitator superfamily consists of presumed carbohydrate transporters with 10-12 membrane-spanning domains. Belonging to the facilitator superfamily, HIAT1 is a 490 amino acid multi-pass membrane protein that may function as a sugar transporter and is expressed in adult and embryonic brain. The HIAT1 gene was first observed while analyzing for active genes in neonatal mouse hippocampus. The gene encoding HIAT1 maps to human chromosome 1, the largest human chromosome spanning about 260 million base pairs and making up 8% of the human genome. There are about 3,000 genes on chromosome 1, and considering the great number of genes there are also a large number of diseases associated with chromosome 1. Notably, the rare aging disease Hutchinson-Gilford progeria is associated with the LMNA gene which encodes lamin A. Stickler syndrome, Parkinsons, schizophrenia, familial adenomatous polyposis, Gaucher disease and Usher syndrome are also associated with chromosome 1.

The LIN-12/notch family of transmembrane receptors is believed to play a central role in development by regulating cell fate decisions. To date, four notch homologs have been identified in mammals and have been designated Notch 1, Notch 2, Notch 3 and Notch 4. The notch genes are expressed in a variety of tissues in both the embryonic and adult organism, suggesting that the genes are involved in multiple signaling pathways. The notch proteins have been found to be overexpressed or rearranged in human tumors. Ligands for notch include Jagged1, Jagged2 and Delta. Jagged can activate notch and prevent myoblast differentiation by inhibiting the expression of muscle regulatory and structural genes. Jagged2 is thought to be involved in the development of various tissues whose development is dependent upon epithelial-mesenchymal interactions. Normal Delta expression is restricted to the adrenal gland and placenta. Delta expression has also been found in neuroendocrine tumors such as neuroblastomas and pheochromocytomas.

Dyrk (for dual specificity tyrosine phosphorylation regulated kinase) is the homolog of the Drosophila mnb (minibrain) gene, which is required for neurogenesis. Dyrk is a dual-specificity tyrosine kinase and serine/threonine kinase, which is itself regulated by tyrosine phosphorylation. Several mammalian Dyrk related proteins have been identified and are thought to compose a family of dual specificity protein kinases. Dyrk family members, including Dyrk1A (originally Dyrk), Dyrk1B, Dyrk1C, Dyrk2, Dyrk3, Dyrk4A and Dyrk4B, are thought to be involved in diverse cellular functions. Two isoforms of human fetal brain Dyrk2 exist: a deduced 528-amino acid protein and a protein containing 73 additional amino acids at the amino terminus. Dyrk3 is strongly expressed in testis, only after the onset of spermatogenesis, and very weakly expressed in spleen and adrenal gland. The genes which encode Dyrk2 and Dyrk3 map to human chromosomes 12 and 1q32, respectively.
