JPH1 Antibody: Junctional complexes between the plasma membrane (PM) and endoplasmic/sarcoplasmic reticulum (ER/SR) are a common feature of all excitable cell types and mediate cross talk between cell surface and intracellular ion channels. Junctophilins (JPs) are important components of the junctional complexes. JPs are composed of a carboxy-terminal hydrophobic segment spanning the ER/SR membrane and a remaining cytoplasmic domain that shows specific affinity for the PM. Four JPs have been identified as tissue-specific subtypes derived from different genes: JPH1 is expressed in skeletal muscle, JPH2 is detected throughout all muscle cell types, and JPH3 and JPH4 are predominantly expressed in the brain and contribute to the subsurface cistern formation in neurons. JPH1 is essential for stabilizing the T-tubule and SR membranes to form junctions and provide an environment for the assembly of receptors such as the ryanodine receptor type 1 (RyR1).

Rkhd1 Antibody: Rkhd1, also known as TINO or MEX3D is a member of a novel family of four homologous human MEX3 proteins each containing two heterogeneous nuclear ribonucleoprotein K homology (KH) domains and one carboxy-terminal RING finger module. MEX3 proteins, including Rkhd1, are phosphoproteins that bind RNA through their KH domains and shuttle between the nucleus and the cytoplasm via the CRM1 export pathway. These proteins are a novel family of evolutionarily conserved RNA-binding proteins, differentially recruited to P bodies and potentially involved in post-transcriptional regulatory mechanisms. Rkhd1 binds to the AU-rich element in the 3'-untranslated region of Bcl-2 mRNA and is thought to be a negative regulator of Bcl-2 expression. Rkhd3 and Rkhd4, but not Rkhd1, co-localize with both the hDcp1a decapping factor and Argonaute (Ago) proteins in processing bodies (P bodies), recently characterized as centers of mRNA turnover.

Cyclin-T1 is a protein that in humans is encoded by the CCNT1 gene. This gene encodes a member of the highly conserved cyclin C subfamily. The encoded protein tightly associates with cyclin-dependent kinase 9, and is a major subunit of positive transcription elongation factor b (p-TEFb). In humans, there are multiple forms of positive transcription elongation factor b, which may include one of several different cyclins along with cyclin-dependent kinase 9. The complex containing the encoded cyclin and cyclin-dependent kinase 9 acts as a cofactor of human immunodeficiency virus type 1 (HIV-1) Tat protein, and is both necessary and sufficient for full activation of viral transcription. This cyclin and its kinase partner are also involved in triggering transcript elongation through phosphorylation of the carboxy-terminal domain of the largest RNA polymerase II subunit. Overexpression of this gene is implicated in tumor growth. Alternative splicing results in multiple transcript variants.

MYST1 (MYST histone acetyltransferase 1,MOF) belongs to the MYST family of histone acetyltransferases, which are employed in the cell to bring about transcriptional regulation. The MYST family includes MYST1, is named for the founding members MOZ, yeast YBF2 and SAS2, and TIP60. All members of this family contain a MYST region of about 240 amino acids with a canonical acetyl-CoA-binding site and a C2HC-type zinc finger motif. Most MYST proteins also have a chromodomain involved in protein- protein interactions and targeting transcriptional regulators to chromatin. Although MOF is expressed in both males and females, it associates with the X chromosome only in males. MOF contains a zinc-finger domain that is used to contact the globular part of the nucleosome and histone H4. The carboxy terminal domain of human MOF also has histone acetyltransferase activity directed against histones H3 and H2A, a characteristic shared with other MYST family histone.

Pax genes contain paired domains with strong homology to genes in Drosophila, which are involved in programming early development. Lesions in the Pax-6 gene account for most cases of aniridia, a congenital malformation of the eye, chiefly characterised by iris hypoplasia, which can cause blindness. Pax-6 is involved in other anterior segment malformations besides aniridia, such as Peters anomaly, a major error in the embryonic development of the eye with corneal clouding with variable iridolenticulocorneal adhesions. The Pax-6 gene encodes a transcriptional regulator that recognises target genes through its paired-type DNA-binding domain. The paired domain is composed of two distinct DNA-binding subdomains, the amino-terminal subdomain and the carboxy-terminal subdomain, which bind respective consensus DNA sequences. The human Pax-6 gene produces two alternatively spliced isoforms that have the distinct structure of the paired domain.

The mucins are a family of highly glycosylated, secreted proteins with a basic structure consisting of a variable number of tandem repeats (VNTRs). Membrane-associated and secretory mucins are high molecular weight glycoproteins of the glycocalyx (polysaccharide biofilm) that protects mucosal epithelium from particulate matter and microorganisms. Epithelial mucins are large, secreted and cell surface glycoproteins crucial for adhesion modulation, signaling and epithelial cell protection. The number of repeats is highly polymorphic and varies among different alleles. The Mucin family consists of Mucins 1-4, Mucin 5 (AC and B), 6-8, 11-13 and 15-17. The Mucin 16 protein (also commonly referred to as CA125), encoded for by the gene MUC16, is a very high molecular weight tumor antigen consisting of three domains: a carboxy terminal domain, an extracellular domain and an amino terminal domain. Mucin 16, an ovarian cancer-associated antigen, is used as a marker to monitor the progress of epithelial ovarian cancer. It is a hydrophilic membrane-associated protein that may be involved in vitamin A functions.

The protein encoded by FGA gene is the alpha component of fibrinogen, a blood-borne glycoprotein comprised of three pairs of nonidentical polypeptide chains. Following vascular injury, fibrinogen is cleaved by thrombin to form fibrin which is the most abundant component of blood clots. In addition, various cleavage products of fibrinogen and fibrin regulate cell adhesion and spreading, display vasoconstrictor and chemotactic activities, and are mitogens for several cell types. Mutations in this gene lead to several disorders, including dysfibrinogenemia, hypofibrinogenemia, afibrinogenemia and renal amyloidosis. Alternative splicing results in two isoforms which vary in the carboxy-terminus.
The total protein is measured using the Biuret procedure with bovine albumin as the standard. To ensure that the product titer falls within the required range, antibody titer is standardized by microtiter plate ELISA with human fibrinogen. The product is tested for purity and specificity at final concentration by immunoelectrophoresis. The antibody is predominantly goat IgG; no trace of albumin is detected.
