The cytochrome P450 proteins are monooxygenases which catalyze many reactions involved in drug metabolism and synthesis of cholesterol, steroids and other lipids. This protein localizes to the endoplasmic reticulum and its expression is induced by some polycyclic aromatic hydrocarbons (PAHs), some of which are found in cigarette smoke. The enzyme's endogenous substrate is unknown; however, it is able to metabolize some PAHs to carcinogenic intermediates. CYP1A1 has been associated with lung cancer risk. This gene, CYP1A1, encodes a member of the cytochrome P450 superfamily of enzymes. Receptors for the Fc region of immunoglobin G (Fc gamma R) are divided into three classes and Fc gamma RIII is a multifunctional, low/intermediate affinity receptor. In humans, Fc gamma RIII is expressed as two distinct forms (Fc gamma RIIIA and Fc gamma RIIIB) that are encoded by two different but highly homologous genes in a cell type-specific manner. Fc gamma RIIIB is a low-affinity, GPI-linked receptor expressed by neutrophils and eosinophils, whereas Fc gamma RIIIA is an intermediate affinity polypeptide-anchored transmembrane glycoprotein expressed by a subset of T lymphocytes, natural killer (NK) cells, monocytes, and macrophages. The Fc gamma RIIIA receptor is involved in phagocytosis, secretion of enzymes, inflammatory mediators, antibody-dependent cellular cytotoxicity (ADCC), mast cell degranulation, and clearance of immune complexes. Fc gamma RIIIA has an immunoreceptor tyrosine-based activation motif (ITAM) in its cytoplasmic domain and delivers an activation signal in the immune responses. Aberrant expression or mutations in this gene is implicated in susceptibility to recurrent viral infections, systemic lupus erythematosus, and alloimmune neonatal neutropenia. In humans, it is a 50 -70 kD type I transmembrane activating receptor. The Fc gamma RIIIA cDNA encodes 254 amino acid including a 16aa signal sequence, 191 amino acid ECD with two C2-type Ig-like domains, five potential N-glycosylation sites, a 22 amino acid transmembrane sequence and a 25 amino acid cytoplasmic domain.

Recognises a protein of 165 kDa, identified as Desmoglein-2 (DSG2). This monoclonal antibody Recognises the extracellular domain of human desmoglein-2. Desmoglein-2 is a member of the desmosomal cadherin family. Desmosomes are intercellular adhering junctions that represent cell surface attachment sites for intermediate filament. Desmocollins and desmogleins are the main desmosomal transmembrane proteins. Desmogleins consist of Dsg1, Dsg2, Dsg3, and Dsg4 isoforms. Within the desmosome, the extracellular domain of desmoglein is essential for calcium dependent heterophilic binding to the desmocollins, whereas the intracellular domain is essential for binding to the desmosomal plaque protein, plakoglobin. Human Desmoglein-2 is a type I transmembrane glycoprotein of 1117 amino acid (aa) residues with a 23 aa signal peptide and a 25 aa propeptide. It differs from other classic cadherins by having four instead of five cadherin repeat domains in its extracellular region, and a much larger cytoplasmic region containing five desmoglein repeat domains which share homology with the cadherin repeats. Instead of having the HAV adhesion motif found in type I cadherins, desmogleins have R/YAL as the adhesion motif on its amino-terminal cadherin repeat. The cytoplasmic tails of desmogleins interact with desmoplakins, plakoglobin and plakophilins. In turn, these proteins link the desmogleins with the intermediate filaments. Desmoglein-2 has been shown to be important in establishing cell-cell adhesion and function in epithelial cells. Desmoglein2 was originally identified in colon carcinoma and colon, and was named HDGC (human desmoglein colon).

Folic acid, also known as folate, is a B vitamin that can be found in a variety of fruits and vegetables. It can also be chemically synthesized. Folate, a watersoluble vitamin, helps the body form red blood cells and aids in the formation of genetic material within every body cell. This product exhibits metal binding properties. Hematopoietic vitamin present, free or combined with one or more additional molecules of L-(+)-glutamic acid, in liver, kidney, mushrooms, spinach, yeast, green leaves, and grasses.
Folic acid (Vitamin B9 and folate) is essential to numerous bodily functions. The human body needs folate to synthesize DNA, repair DNA, and methylate DNA as well as to act as a cofactor in certain biological reactions. It is especially important in aiding rapid cell division and growth, such as in infancy and pregnancy. Folic acid (FA) and dihydrofolic acid (FAH2) are substrates of dihydrofolate reductase(s) which reduce them to tetrahydrofolate (THF), which in turn supports ‘one carbon’ transfer. Tetrahydrofolates are required for de novo synthesis of purines, thymidylic acid and various amino acids and for post-translational methylation (epigenetics). Very slightly soluble in cold water (0.0016 mg/mL at 25 °C), soluble to about 1% in boiling water. Slightly soluble in methanol, appreciably less soluble in ethanol and butanol. Insoluble in acetone, chloroform, ether, benzene. Relatively soluble in acetic acid, phenol, pyridine, solutions of alkali hydroxides and carbonates. Soluble in hot dilute HCl and H2SO4.

Ethidium bromide(EtBr) has been used extensively to reduce mitochondrial DNA copy number in proliferating cells and most commonly used in nucleic acid stain for PAGE or agarose gel electrophoresis. The fluorescence of EtBr increases 21-fold upon binding to double-stranded RNA and 25-fold on binding double-stranded DNA so that destaining the background is not necessary with a low stain concentration (10 μg/mL). Ethidium bromide has been used in a number of fluorimetric assays for nucleic acids. It has been shown to bind to single-stranded DNA (although not as strongly) and triple-stranded DNA.4 Because of its ability to bind to DNA, EtBr is an inhibitor of DNA polymerase.
A 10mg/mL easy-to-use solution of ethidium bromide in specially filtered, deionized water which is excellent for nucleic acid electrophoresis and purification applications, eliminates the dust hazard associated with powdered ethidium bromide and saves time spent on weighing and mixing. Used in the fluorometric detection of double stranded nucleic acids in gel electrophoresis. Also acts as an RNA polymerase inhibitor, and in separation of high molecular weight DNA's. It confers a deep red stain to DNA by acting as an intercalating agent between the starch bases. Subsequent fluorescence techniques enable the extraction and characterization of DNA bands.
Biochem/physiol Actions:
Ethidium bromide intercalates double-stranded DNA and RNA and acts as a frameshift mutagen. It can also be used in conjunction with acridine orange to differentiate between viable, apoptotic and necrotic cells.
