IL-16 was initially identified as a chemotactic cytokine (1), but is now known to possess a wide range of activities. Later studies have more fully characterized IL-16 as an immunomodulatory cytokine that contributes to the regulatory process of CD4+ T cell recruitment and activation at sites of inflammation in association with asthma and several autoimmune diseases (2). The precursor of IL-16 (pro-IL-16) is thought to be cleaved towards the C-terminal region by Caspase-3, releasing a 20 kDa active form that binds to and signals through CD4 (3,4). Besides acting as a chemotactic cytokine, IL-16 is thought to also be involved in the regulation of T cell proliferation (5) and multiple infectious, immune-mediated, and autoimmune inflammatory disorders including irritable bowel syndrome, systemic lupus erythematosus, and neurodegenerative disorders (6). At least two isoforms of IL-16 are known to exist; the longer isoform (also known as NIL-16) is detected only in neurons of the cerebellum and hippocampus (7). This recombinant protein fragment is derrived from this longer isoform.

TGF beta actions are directed by ligand induced activation of TGF beta receptors. Complexes formed move into the nucleus, where they act as components of a transcriptional complex. Lefty, a novel member of the TGF beta superfamily, inhibits TGF beta signaling. Lefty acts to inhibit phosphorylation of Smad2 following activation of the TGF beta receptor. Lefty also inhibits events downstream from R Smad phosphorylation. Lefty provides a repressed state of TGF beta responsive genes. The Lefty family is comprised of Lefty 1 and Lefty 2 in mouse, and Lefty A and Lefty B in humans. Members of the TGF beta superfamily require processing for their activation. Cleavage is therefore an essential step for Lefty activation. Lefty is synthesized as a large inactive precursor (41 Kda) that must be endoproteolytically processed to release the bioactive polypeptide (28 kDa and 34 kDa forms). The 28kDa form induces MAPK activity.

EMAP II (Endothelial monocyte-activating polypeptide 2) is a pro-inflammatory cytokine and participant in apoptotic cell death. It is synthesised as a precursor (pro-EMAP II), lacking a secretion signal peptide. In apoptotic cells, the ~34 kDa pro-form of EMAP II is cleaved by Caspase-7 (and Caspase-3 to a lesser extent) at aspartate 144 to its mature, ~20 kDa form. The mature form of EMAP II has been found to have many different functions. In vivo, it recruits leukocytes to the site of apoptotic cell death. In vitro it has been shown to activate endothelial cells (and the release of von Willebrand factor, E-selectin and P-selectin), neutrophils, and induces macrophage expression of TNFa and TF (tissue factor). EMAP II is also a possible biomarker for brain injury, having been found to be differentially expressed in two types: traumatic injury shows an increase and ischemic injury shows a decrease in EMAP II expression (Dave, et al, 2014).
