Recognizes proteins of 33, 35 and 41 kDa, which are identified as various isoforms of CD74. Its epitope is localized in the extracellular domain of CD74. CD74 is a type II transmembrane protein which binds to the peptide binding groove of newly synthesized MHC class II alpha/beta heterodimers and prevents their premature association with endogenous polypeptides. The CD74 molecule plays a critical role in the presentation of peptides, by the MHC class II antigens, to CD4 positive lymphocytes. CD74 is expressed on MHC class II positive cells including B cells, a subset of activated T cells, monocytes, and dendritic cells and by various types of carcinomas. RTF1 is the the major histocompatibility complex (MHC) in the rat which contains genes that code for two class II histocompatibility antigens. RT1-B is an antigen of the RT1 complex. It is a protein dimer consisting of an alpha and beta glycoprotein chain and is homologous to I-A and I-E genes, respectively, in the H-2 complex of the mouse. MHC Class II antigens are useful in studying T helper cell interaction with class II positive antigen presenting cells (dendritic cells, B cells, macrophages) and offer new possibilities for studying the development of T helper cells since these also stain stromal cells in the thymus.

It reportedly reacts with B- & non-T, non-B cell lines but not with T- and myeloid cell lines and leukemias. Differential expression of MHC class II antigens on fetal and adult lymphocytes, malignant B cells appears to reflect the stage of cell differentiation which may be useful in the study of lymphoproliferative disorders.
