FBXO24 is a member of the F-box protein family which is characterized by an approximately 40 amino acid motif, the F-box. The F-box proteins constitute one of the four subunits of the ubiquitin protein ligase complex called SCFs (SKP1-cullin-F-box), which function in phosphorylation-dependent ubiquitination. The F-box proteins are divided into 3 classes: Fbws containing WD-40 domains, Fbls containing leucine-rich repeats, and Fbxs containing either different protein-protein interaction modules or no recognizable motifs. The protein belongs to the Fbxs class. Alternative splicing of this gene generates two transcript variants. SCUBE1 Antibody: SCUBE1 is an EGF-related protein with a CUB (C1s-like) domain that is expressed primarily in the developing gonad, nervous system, somites, surface ectoderm and limb buds. The N-terminal EGF-like repeats of SCUBE1 participate in the formation of adhesions in a Ca2+-dependent fashion. In contrast, the C-terminal cysteine-rich motifs and CUB domain can directly bind and antagonize bone morphogenic protein activity; genetic ablation of this region resulted in brain malformation in mice embryos, suggesting SCUBE1 plays a role in brain morphogenesis. SCUBE1 has also been implicated in early craniofacial development in mice.

PION Antibody: Accumulation of the amyloid-beta peptide (Abeta) in the cerebral cortex is a critical event in the pathogenesis of Alzheimer's disease. The beta-amyloid protein precursor (APP) is cleaved by one of two beta-secretases (BACE and BACE2), producing a soluble derivative of the protein and a membrane anchored 99 -amino acid carboxy-terminal fragment (C99). The C99 fragment serves as substrate for gamma-secretase to generate the 4 kDa amyloid-beta peptide (Abeta), which is deposited in the Alzheimer's disease patient's brains. PION, or GSAP, selectively increases amyloid-beta production through a mechanism involving its interaction with both gamma-secretase and the APP C-terminal fragment, suggesting that PION may be a potential therapeutic target for the treatment of Alzheimer's disease.

Differential screening-selected gene aberrative in neuroblastoma (DAN) is a member of the DAN family of secreted glycoproteins. DAN family antagonists are Characterised by a DAN domain that contains a cystine knot motif which is essential for binding to BMP ligands. Members of this family include DAN, gremlin, protein related to DAN and cerberus (PRDC), cerberus, sclerostin (SOST) and uterine sensitisation-associated gene 1 protein, and control diverse processes in growth, development and the cell cycle. It has also been reported that DAN family plays crucial role in early mouse embryo development by inhibiting the action of bone morphogenic proteins and modulating the action of transforming growth factor- beta superfamily members. DAN is Synthesised by small-to intermediate-sised DRG neurons and transported to the sensory nerve terminals in the skin or to the sensory nerve terminals in the dorsal horn. It has been reported that DAN is ubiquitously expressed in adult rat and human tissues. Morphological studies have revealed that, in adult rat, DAN mRNA is expressed ubiquitously in lung and brain, but not in liver.

Neuroserpin is a member of the serpin superfamily and is predominantly expressed by neurons of the developing and adult brain. This serpin is secreted from axonal growth cones of the CNS and PNS where it inhibits the enzyme tissue plasminogen activator. The expression pattern of neuroserpin and its in vivo inhibitory activity implicate this serpin in regulating axonal growth, reducing seizure activity, controlling damage in cerebral infarction and regulating emotional behaviour and memory. Four different mutations have been described in the human neuroserpin gene that promote neuroserpin abnormal polymerization, giving rise to intraneuronal inclusion bodies and a new form of dementia called Familial Encephalopathy with Neuroserpin Inclusion Bodies.

It and the related proteins AMIGO2 and AMIGO3 are thought to be cell adhesion molecules expressed on fiber tracts of neuronal tissues and participate in their formation. AMIGO1 has also been suggested to play important roles in dendritic outgrowth during development and could modulate the survival of developing and adult neurons.
