E3 ubiquitin-protein ligase which accepts ubiquitin from E2 ubiquitin-conjugating enzymes UBE2L3 and UBE2L6 in the form of a thioester and then directly transfers the ubiquitin to targeted substrates such as LCMT2, thereby promoting their degradation. Induces apoptosis via a p53/TP53-dependent but caspase-independent mechanism. GDE3, also known as glycerophosphodiester phosphodiesterase 3, glycerophosphodiester phosphodiesterase domain containing 2 (GDPD2) or osteoblast differentiation promoting factor (OBDPF), is a 539 amino acid protein belonging to the glycerophosphoryl diester phosphodiesterase family. Possessing glycerophosphoinositol inositolphosphodiesterase activity, GDE3 hydrolyzes glycerophosphoinositol to form inositol 1-phosphate and glycerol, and is suggested to play a role in Actin cytoskeleton remodeling and osteoblast differentiation and growth. A multi-pass membrane protein, GDE3 localizes to cell membrane and cytoplasm, and colocalizes with actin in the cytoskeleton. GDE3 contains one GDPD domain, binds calcium as a cofactor and is encoded by a gene mapping to human chromosome Xq13.1.

This complex plays an important role in coupling antigen recognition to several intracellular signal-transduction pathways. The genes encoding the epsilon, gamma and delta polypeptides are located in the same cluster on chromosome 11. The epsilon polypeptide plays an essential role in T-cell development. Emp is a 396 amino acid ubiquitously expressed adhesion protein. Expressed as 5 alternatively spliced isoforms, Emp contains one CTLH domain and one LisH domain. Emp can form a complex with F-actin, which is involved regulating actin distribution in erythroblasts and macrophages. Considered to assist with cell division and nuclear architecture, Emp is localized with condensed chromatin at prophase, nuclear spindle poles at metaphase and in the contractile ring during telophase and cytokinesis. Although the exact function of Emp is unknown, Emp is suggested to be involvement in erythroblast-macrophage cell attachment, terminal maturation and enucleation of erythroid cells, and inhibiting apoptosis of erythroblasts.

Bmf (Bcl-2-modifying factor) belongs to the Bcl-2 protein family. Bcl-2 family members form hetero- or homo-dimers and act as anti- or pro-apoptotic regulators that are involved in a wide variety of cellular activities. Bmf contains a single Bcl-2 homology domain 3 (BH3), and has been shown to interact with other Bcl-2 proteins and functions as an apoptotic activator. Two different start sites can give rise to two proteins of 25 or 30 kDa. It is found to be sequestered to myosin V motors by its association with dynein light chain 2 (DLC2), which may be important for sensing intracellular damage and triggering apoptosis.

This gene encodes a member of the Wiskott-Aldrich syndrome protein family. The gene product is a protein that forms a multiprotein complex that links receptor kinases and actin. Binding to actin occurs through a C-terminal verprolin homology domain in all family members. The multiprotein complex serves to tranduce signals that involve changes in cell shape, motility or function. The published map location (PMID:10381382) has been changed based on recent genomic sequence comparisons, which indicate that the expressed gene is located on chromosome 1, and a pseudogene may be located on chromosome X. Two transcript variants encoding different isoforms have been found for this gene.
