GAP43 is very abundant protein which is found concentrated in neurons. One group discovered it as one of three proteins which becomes unregulated during the regeneration of the toad optic nerve (1). Three GAPs (Growth associated proteins) were discovered, and the number 43 comes from the apparent SDS-PAGE molecular weight of the one named GAP43. The HGNC name for this protein is, not surprisingly, GAP43. Later work showed that GAP43 does not run on SDS-PAGE in a fashion which accurately reflects its molecular weight, and that GAP43 proteins from different species may run at different apparent molecular weights. Partly due to these features GAP43 were independently discovered by several different groups and therefore has several alternate names, such as protein F1, pp46, neuromodulin, neural phosphoprotein B-50 and calmodulin-binding protein P-57. In each case the number reflects the apparent SDS-PAGE molecular weight, and underlines the unusual properties of this molecule. Mammalian GAP43 proteins contains only 226-243 amino acids, and so the real molecular weight is 23.61-25.14 kDa. GAP43 has been extensively studied and is known to be a major protein kinase C substrate and to bind calmodulin avidly. GAP43 is anchored to the plasma membrane by palmitoylation modifications.

C16orf44 is believed to play a role in protein ubiquitination and may function as a substrate-specific adapter of an E3 ubiquitin-protein ligase complex. E3 ligases accept a ubiquitin residue from an E2 ubiquitin-conjugating enzyme and immediately transfer that residue to a protein that is targeted for degradation. Specifically, C16orf44 interacts with CUL-3, a member of the cullin family of mediators that participate in the selective targeting of proteins for ubiquitin-mediated proteolysis. Due to alternative splicing events, two isoforms of C16orf44 are expressed.

Hereditary hemorrhagic telangiectasia, which is characterized by harmful vascular defects, and Familial dysautonomia, are both associated with chromosome 9. Notably, chromosome 9 encompasses the largest interferon family gene cluster.

It exists as two alternatively spliced isoforms that are abundantly expressed in brain, lung, and testis as well as in embryonic hippocampal and cortical neurons. FARP2 functions as a Rho-guanine nucleotide exchange factor that activates RAC1 and is thought to regulate neurite remodeling of embryonic neurons. Sema3A binding to neuropilin-1 induces the dissociation of FARP2 from plexin-A1, thereby activating FARP2's Rac GEF activity which is critical for repulsion of outgrowing axons and suppression of neuronal adhesion. Downregulation of the FARP2 gene has been implicated in autism.

ILVBL is a 632 amino acid single-pass membrane protein that belongs to the TPP enzyme family. Expressed in the majority of tissues, ILVBL has the highest level of expression in heart, pancreas and placenta. ILVBL is highly homologous to several bacterial enzymes, including the B isozyme of the large catalytic subunit of E. coli acetohydroxy-acid synthase (AHAS) and the oxalyl-coA decarboxylase of O. formigenes, that utilize thiamine pyrophosphate as a cofactor. ILVBL binds one magnesium ion and one thiamine pyrophosphate per subunit, and may catalyze the initial step in branched-chain amino acid biosynthesis. RGS14 is a member of the regulator of G-protein signaling family. This protein contains one RGS domain, two Raf-like Ras-binding domains (RBDs), and one GoLoco domain. The protein attenuates the signaling activity of G-proteins by binding, through its GoLoco domain, to specific types of activated, GTP-bound G alpha subunits. Acting as a GTPase activating protein (GAP), the protein increases the rate of conversion of the GTP to GDP. This hydrolysis allows the G alpha subunits to bind G beta/gamma subunit heterodimers, forming inactive G-protein heterotrimers, thereby terminating the signal. Alternate transcriptional splice variants of this gene have been observed but have not been thoroughly characterised.

Glycosylphosphatidylinositol anchor attachment 1 protein (GPAA1), also designated GPI anchor attachment protein 1 or GAA1 protein homolog, is a membrane protein localized to the endoplasmic reticulum which is involved in GPI-anchor biosynthesis. GPAA1 is crucial for GPI-anchoring of precursor proteins and catalyzes the attachment of GPI to proteins containing a C-terminal GPR attachment signal. GAA1 contains an N-terminal signal sequence, one cAMP- and cGMP-dependent protein kinase phosphorylation site, two potential N-glycosylation sites, one leucine zipper pattern and eight putative transmembrane domains. GPAA1 is ubiquitously expressed and shows higher levels of expression in fetal tissues than in adult tissues.

Localised to the cytoplasm, ASC2 interacts with ASC to modulate NF-B and pro-caspase-1 regulation. ASC2 is predominantly expressed in monocytes, macrophages and granulocytes.
