Listeria monocytogenes ist eines der am weitesten verbreiteten Lebensmittelpathogene der Welt und verantwortlich für schwere Infektionen bei immungeschwächten Personen, schwangeren Frauen und Neugeborenen. In Folge ist die Mortalitätsrate bei infizierten Personen hoch, trotz der relativ geringen Anzahl von Fällen weltweit. Listeria-Infektionen (Listeriose) können zu einer milden Gastroenteritis bis hin zu schweren Fällen von Sepsis, Meningitis, Enzephalitis sowie zu Fehlgeburten führen. Aufgrund der ubiquitären Verteilung von Listeria und ihrer Fähigkeit, sich auch bei Kühlschranktemperaturen (2 bis 8 °C) zu vermehren, bilden Lebensmittel eine der wichtigsten Infektionsquellen. Listerien sind eine Gattung von Gram-positiven, nicht sporenbildenden, stabförmigen Bakterien. Von den sechs bekannten Spezies der Gattung Listeria ist Listeria monocytogenes als Human- und Tierpathogen besonders hervorzuheben; L. ivanovii ist nur tierpathogen, während L. innocua, L. seeligeri, L. grayi und L. welshimeri als unbedenkliche Umweltbakterien gelten. Folglich schreibt das Lebensmittelgesetz in vielen Ländern strenge Beschränkungen für Listeria spp. oder Listeria monocytogenes vor. Bei der Durchführung von Risiko-bezogenen Qualitätskontrollen in Lebensmitteln sollten die Tests für L. monocytogenes und für die Gattung Listeria im Allgemeinen durchgeführt werden. Das Vorhandensein von Listerien – insbesondere von L. innocua – ist ein Indikator für kritische hygienische Bedingungen im Produktionsprozess. Innovative Schnelltests erleichtern das Screening auf pathogene Keime. Das Ein-Schritt-Format reduziert die Möglichkeit von Fehlern bei der Durchführung.

Recognizes a protein of 33-37 kDa, identified as CD20 (Workshop V; Code CD20.12). B9E9 recognizes extracellular domain of CD20. The epitope is similar to or identical to that recognized by other CD20 antibodies including Leu-16 and B1. This MAb can be used for immunophenotyping of leukemia and malignant cells, B lymphocyte detection in peripheral blood, B cell localization in tissues and B lymphocyte purification by immunosorbent methods. CD20 is a non-Ig differentiation antigen of B-cells and its expression is restricted to normal and neoplastic B-cells, being absent from all other leukocytes and tissues. CD20 is expressed by pre B-cells and persists during all stages of B-cell maturation but is lost upon terminal differentiation into plasma cells. Protein passes through the membrane 4 times with both ends in cytoplasm and exposes one short and one longer loop to the external environment. CD20 is not glycosylated in resting B cells and its cytoplasmic domains are differentially phosphorylated upon activation. It acts as a calcium channel involved in B-cell activation and cell cycle progression.

Voltage-dependent calcium channels are essential for the release of neurotransmitters. L-type (long lasting current) voltage-dependent calcium channels are composed of four subunits: an Alpha1 subunit, a Beta subunit, a Beta subunit and an Alpha2 Gamma subunit. The Beta subunit is encoded by four genes, designated Beta1-Beta 4, all of which contribute to the diversity of calcium currents and are involved in membrane trafficking of the Beta subunit. L-type Ca++ CP Beta 4, also known as CACNB4 (Calcium channel voltage-dependent subunit beta 4), CACNLB4 or CAB4, is a 484 amino acid protein that contains one SH3 domain and is expressed in ovary, brain and smooth muscle. Functioning as one of the four components of the Beta subunit, L-type Ca++ CP Beta 4 increases the peak calcium current in voltage-dependent calcium channels, thereby shifting the voltage dependencies of activation and inactivation and controlling G protein inhibition and Beta membrane targeting. Two isoforms of L-type Ca++ CP Beta4 exist due to alternative splicing events.

Protocadherins are a large family of cadherin-like cell adhesion proteins that are involved in the establishment and maintenance of neuronal connections in the brain. There are three protocadherin gene clusters, designated alpha, beta and gamma, all of which contain multiple tandemly arranged genes. PCDHB6 (Protocadherin beta-6) is a 794 amino acid single pass transmembrane protein that is one of 16 proteins in the protocadherin beta cluster. Unlike the alpha and gamma gene clusters whose genes are spliced to downstream constant region exons during transcription, members of the beta cluster (such as PCDHB6) do not use constant-region exons to produce mRNAs. As a result, each protocadherin beta gene encodes the transmembrane, extracellular and short cytoplasmic domains of the protein. PCDHB6 is likely a calcium-dependent cell adhesion protein that is involved in the maintenance of neural connections in the brain. Unlike most protocadherin-beta proteins, PCDHB6 has not one but two PXXP motifs within its cytoplasmic domain, suggesting a role in signal transduction cascade events.

The RA3-6B2 antibody reacts with the human and mouse CD45 isoform known as CD45R, or B220, a protein tyrosine phosphatase of 220 kDa. CD45 is one of the most abundant hematopoietic markers, and is expressed on all leukocytes (the Leukocyte Common Antigen, LCA). Various isoforms are generated and expressed in cell-specific patterns, all critical for leukocyte function. In mouse, the CD45R/B220 isoform is predominantly found on B cells, at varying levels on all stages from pro-B cells to activated B cells, and may also be detected on certain T cell and NK cell subsets. It is of note that B220 is not similarly expressed on human B cells, where it appears to be differentiation-specific and therefore expressed on only some B cell subsets. Other forms of CD45 with restricted cellular expression include CD45RA, CD45RB, CD45RO and several others. The RA3-6B2 antibody is one of the most consistently used leukocyte markers for B cells, T cell subsets and NK cell subsets in human and mouse. This antibody has also been reported as cross-reactive with feline CD45R/B220.

Guanylate cyclases belong to the adenylyl cyclase class-4/guanylyl cyclase family. There are two forms of guanylate cyclase. The soluble forms, known as GCS or sGC, act as receptors for nitric oxide. The membrane-bound receptor forms, known as GC, are peptide hormone receptors. GCS, a cGMP-synthesizing enzyme, is the major receptor for the neurotransmitter nitric oxide (NO). It plays a crucial role in smooth muscle contractility, platelet reactivity and neurotransmission. GCS is a heme containing heterodimer, consisting of one alpha subunit, designated GCS-alpha-1, and one beta subunit. The heme moeity mediates NO activation, and this heme group also binds carbon monoxide, which weakly stimulates the enzyme. Both NO and CO stimulation are enhanced by the allosteric activator 3-(5'-hydroxymethyl-2'furyl)-benzyl-indazole, YC-1. YC-1 can also stimulate GCS in a NO-independent manner. Both the alpha and beta subunits are required for cGMP generation, and at least two isoforms exist for each subunit. Heterodimers consisting of alpha-1/beta-1 and alpha-2/beta-1 have been identified, and both display similar enzymatic activity.

Localized to both the cytoplasm and the nucleus, EXOSC10 is part of the post-splicing exosome complex and is involved in mRNA surveillance, mRNA nuclear export and nonsense-mediated decay of mRNAs containing premature stop codons. against EXOSC10 have been found in patients with scleroderma and/or polymyositis (chronic diseases of the skin and muscle, respectively), suggesting that EXOSC10 may be involved in the pathogenesis of these diseases. Two isoforms of EXOSC10 exist due to alternative splicing events.
