iASPP (inhibitor of apoptosis stimulating protein of p53, RelA-associated inhibitor (RAI), NFKap B-interacting protein 1, protein phosphatase 1 regulatory subunit 13-like, PPP1R13L) is the third member of the ASPP family of proteins. Unlike ASPP1 and ASPP2, which interact with p53 and enhance its ability to induce apoptosis, iASPP inhibits apoptosis induced by p53 overexpression. iASPP has been identified in a yeast-two hybrid screen as an interacting protein of the p65 subunit of NFKap B (RelA),3 interacts with NFKap B in vivo, and acts as an efficient inhibitor of HIV-1 gene expression in which both NFKap b and Sp1 play major roles. iASPP expression is upregulated in human breast carcinomas expressing wild-type p53, and gene overexpression may play an important role in the leukemogenesis and/or progression of acute leukemia. Playing a key role in the development of the limb bud, Radical Fringe transfers a beta-D-GlcNAc residue from UDP-D-GlcNAc to the fucose residue of a fucosylated protein acceptor. Lunatic Fringe is required for normal somite segmentation and patterning and is thought to be a target of the molecular clock. Manic Fringe, also involved in somatic development, has been shown to render mouse NIH/3T3 cells tumorigenic in SCID mice.

Aldehyde dehydrogenases (ALDHs) mediate NADP+-dependent oxidation of aldehydes into acids during the detoxification of alcohol-derived acetaldehyde; metabolism of corticosteroids, biogenic amines and neurotransmitters; and lipid peroxidation. ALDH1A1, also designated retinal dehydrogenase 1 (RalDH1 or RALDH1), aldehyde dehydrogenase family 1 member A1, aldehyde dehydrogenase cytosolic, ALDHII, ALDH-E1 or ALDH E1, is a retinal dehydrogenase that participates in the biosynthesis of retinoic acid (RA). There are two major liver isoforms of ALDH1 that can localize to cytosolic or mitochondrial space. The ALDH1A2 (RALDH2, RALDH2-T) gene produces three different transcripts and also catalyzes the synthesis of RA from retinaldehyde. ALDH1A3 (ALDH6, RALDH3, ALDH1A6) is a 37 kb gene that consists of 13 exons and produces a major transcript of approximately 3.5 kb most abundant in salivary gland, stomach and kidney. ALDH3A1 (stomach type, ALDH3, ALDHIII) forms a cytoplasmic homodimer that preferentially oxidizes aromatic aldehyde substrates. ALDH genes upregulate as a part of the oxidative stress response, and appear to be abundant in certain tumors that have an accelerated metabolism toward chemotherapy agents.

Heterotrimeric G proteins function to relay information from cell surface receptors to intracellular effectors (1). Each of a very broad range of receptors specifically detects an extracellular stimulus (a photon, pheromone, odorant, hormone or neurotransmitter) while the effectors (i.e., adenylyl cyclase), which act to generate one or more intracellular messengers, are less numerous. In mammals, G protein alpha, Beta and Gamma polypeptides are encoded by at least 16, 4 and 7 genes, respectively (2-5). Most interest in G proteins has been focused on their a subunits, since these proteins bind and hydrolyze GTP and most obviously regulate the activity of the best studied effectors. Four distinct classes of G alpha subunits have been identified; these include Gs, Gi, Gq and Ga 12/13 (3,4). The Gi class comprises all the known a subunits that are susceptible to pertussis toxin modifications, including Ga i-1, Ga i-2, Ga i-3, Ga o, Ga t1, Ga t2, Ga z and Ga gust (4). Of these, the three Ga i subtypes function to open atrial potassium channels (6). Ga 16 is a member of the Gq subfamily and is expressed specifically in hematopoietic cells (7).
