Beschreibung:   AROS-29 is suggested to have a functional role in protection against oxidative stress. The gene encoding AROS-29 is located on human chromosome 19, which consists of over 63 million bases, houses approximately 1,400 genes and is recognized for having the greatest gene density of the human chromosomes. It is the genetic home for a number of immunoglobulin superfamily members, including the killer cell and leukocyte Ig-like receptors, a number of ICAMs, the CEACAM and PSG family and Fc receptors (FcRs). Key genes for eye color and hair color also map to chromosome 19.

Beschreibung:   ELAC2 is a zinc phosphodiesterase which displays some tRNA 3' processing endonuclease activity. It is thought to be involved in tRNA maturation, acting by removing a 3' trailer from precursor tRNA. ELAC2 also interacts with activated Smad family member 2 (Smad2) and its nuclear partner forkhead box H1 (also known as FAST-1), and reduced expression can suppress transforming growth factor-beta induced growth arrest. Mutations in this gene result in an increased risk of prostate cancer. A number of allelic variants have been investigated in hereditary prostate cancer.

Beschreibung:   GNL2 is a nucleolar guanasine-triphosphate binding protein that is ubiquitously expressed at low levels in almost all tissues. GNL2 is involved in the crucial process of trafficking proteins out of the nucleus. Specifically, it is a GTPase that interacts with the 60s preribosomal subunit in the nucleus and facilitates export of the subunit into the cytoplasm. GTPases are responsible for the hydrolysis of GTP by way of a protein region dubbed the G domain. GTPases are often involved in the translocating proteins through membranes gleaning energy for the activity by hydrolizing GTP. GNL2 shares G domain homology and some functionality with nucleostemin (GNL3), another nuclear GTPase. Highest expression of GNL2 is found in testis.

Beschreibung:   Syntaxins were originally thought to be docking proteins, but have more recently been categorized as anchoring proteins that anchor themselves to the cytoplasmic surfaces of cellular membranes. Syntaxins have been shown to bind to various proteins involved in exocytosis, including VAMPs (vesicle-associated membrane proteins), NSF (N-ethylmaleimide-sensitive factor), SNAP 25 (synaptosomal-associated protein of 25kDa), SNAPs (soluble NSF attachment proteins) and synaptotagmin. VAMPs (also designated synaptobrevins), including VAMP-1 and VAMP-2, and synaptotagmin, a protein that may function as an inhibitor of exocytosis, are vesicular proteins.

Beschreibung:   This gene encodes a member of the vaccinia-related kinase (VRK) family of serine/threonine protein kinases. This gene is widely expressed in human tissues and has increased expression in actively dividing cells, such as those in testis, thymus, fetal liver, and carcinomas. Its protein localises to the nucleus and has been shown to promote the stability and nuclear accumulation of a transcriptionally active p53 molecule and, <i>in vitro</i>, to phosphorylate Thr18 of p53 and reduce p53 ubiquitination. This gene, therefore, may regulate cell proliferation. This protein also phosphorylates histone, casein, and the transcription factors ATF2 (activating transcription factor 2) and c-JUN.

Beschreibung:   Probable E3 ubiquitin-protein ligase. The BRCA1-BARD1 heterodimer specifically mediates the formation of 'Lys-6'-linked polyubiquitin chains and coordinates a diverse range of cellular pathways such as DNA damage repair, ubiquitination and transcriptional regulation to maintain genomic stability. Plays a central role in the control of the cell cycle in response to DNA damage. Acts by mediating ubiquitin E3 ligase activity that is required for its tumor suppressor function. Also forms a heterodimer with CSTF1/CSTF-50 to modulate mRNA processing and RNAP II stability by inhibiting pre-mRNA 3' cleavage.

Beschreibung:   BIVM (for basic, immunoglobulin like variable motif containing) refers to a recently identified gene product that maps to human chromosome 13q32-q33 and is predicted to encode a 503 amino acid protein. BIVM shows ubiquitous expression in normal human tissue and the presence of a 5' CpG island suggests it is a housekeeping gene. BIVM is likely essential for some aspect of basic cellular function. BIVM is highly charged and localizes to the cytoplasm and nucleus where it may bind to either DNA or RNA or associate with other cellular proteins. Significant sequence homology exists with many organisms.

Beschreibung:   MP21 is a member of the EMP24/GP25L/p24 cargo family of proteins that regulates vesicular trafficking in the early secretory pathway. TMP21 is a ubiquitously expressed single-pass type I membrane protein localizing to the Golgi cisternae and the plasma membrane. It contains one GOLD (Golgi dynamics) domain and participates in protein transport and quality control between the endoplasmic reticulum (ER) and the Golgi complex. In addition, TMP21 is a component of the heteromeric secretase complex (or Presenilin complex) and functions to regulate the ?secretase activity. Two isoforms, namely TMP21-I and TMP21-II, exist for this protein.

Beschreibung:   Tuberin, or TSC2 (Tuberous sclerosis complex), is implicated as a tumor suppressor. It may function in vesicular transport, and may also play a role in the regulation of cell growth arrest and in the regulation of transcription mediated by steroid receptors. Interaction between hamartin (TSC1) and tuberin may facilitate vesicular docking. It specifically stimulates the intrinsic GTPase activity of the Ras related protein RAP1A and RAB5, suggesting a possible mechanism for its role in regulating cellular growth. Mutations in tuberin lead to constitutive activation of RAP1A in tumors. At least three isoforms of Tuberin exist.

Beschreibung:   Chondroitinase is a 522 amino acid protein that localizes to the lysosome and functions as an exohydrolase that is essential for the degradation of glycosaminoglycans, keratan sulfate and chondroitin 6-sulfate. Using calcium as a cofactor, Chondroitinase, which exists as a disulfide linked oligomer, catalyzes the hydrolysis of the 6-sulfate group on target substrates. Defects in the gene encoding Chondroitinase are the cause of mucopolysaccharidosis type 4A (MPS4A), an autosomal recessive lysosomal storage disease that is characterized by the intracellular accumulation of keratan sulfate and chondroitin-6-sulfate and is associated with dental anomalies, short stature and, in some cases, death in the second or third decade of life.
