The MEL-14 monoclonal antibody specifically reacts with L- selectin (CD62L), a receptor with lectin-like and Epidermal Growth Factor-like domains. The weight of the CD62L molecules depend on their origin: 74 kDa (on lymphocytes) or 95 kDa (on neutrophils).In the Mouseorganism, CD62L can be expressed by most thymocytes, on subsets of B and T lymphocytes, monocytes, eosinophils, and neutrophils. The L-selectin binds sulfated, fucosylated, and glycosylated glycoproteins (MadCAM-1, GLYCAM-1, and CD 34). It mediates the migration of lymphocytes to the site of inflammation and their return to the peripheral lymphoid tissues and to the HEV (high endothelial venules). In vitro, L-selectin inhibits this binding and the lymphocyte extravasation into peripheral lymph nodes.

Recognizes the L1 or Calprotectin molecule, an intra-cytoplasmic antigen comprising of a 12 kDa alpha chain and a 14 kDa beta chain expressed by granulocytes, monocytes and by tissue macrophages. Macrophages usually arise from hematopoietic stem cells in the bone marrow. Under migration into tissues, the monocytes undergo further differentiation to become multifunctional tissue macrophages. They are classified into normal and inflammatory macrophages. Normal macrophages include macrophages in connective tissue (histiocytes), liver (Kupffer's cells), lung (alveolar macrophages), lymph nodes (free and fixed macrophages), spleen (free and fixed macrophages), bone marrow (fixed macrophages), serous fluids (pleural and peritoneal macrophages), skin (histiocytes, Langerhans's cell) and in other tissues. Inflammatory macrophages are present in various exudates. Macrophages are part of the innate immune system, recognizing, engulfing and destroying many potential pathogens including bacteria, pathogenic protozoa, fungi and helminthes. C19orf28, also known as PP3501, is a multi-pass membrane protein that belongs to the major facilitator superfamily. The gene encoding C19orf28 localizes to chromosome 19 and, due to alternative splicing events, C19orf28 exists as two isoforms. Consisting of around 63 million bases with over 1,400 genes, chromosome 19 makes up over 2% of human genomic DNA. Chromosome 19 includes a diversity of interesting genes and is recognized for having the greatest gene density of the human chromosomes. It is the genetic home for a number of immunoglobulin superfamily members including the killer cell and leukocyte Ig-like receptors, a number of ICAMs, the CEACAM and PSG family, and Fc receptors. Key genes for eye color and hair color also map to chromosome 19. Peutz-Jeghers syndrome, spinocerebellar ataxia type 6, the stroke disorder CADASIL, hypercholesterolemia and insulin-dependent diabetes have been linked to chromosome 19. Translocations with chromosome 19 and chromosome 14 can be seen in some lymphoproliferative disorders and typically involve the proto-oncogene BCL3. The C19orf28 gene product has been provisionally designated C19orf28 pending further characterization.

HAS1, HAS2 and HAS3 are HA (hyaluronan or hyaluronic acid) synthase proteins. The extracellular matrix in most vertebrates express HA, which is a high molecular weight linear polysaccharide composed of alternating glucuronic acid and N-acetylglucosamine residues linked by b-1,3 and b-1,4 glycosidic bonds. The three HAS genes show distinct patterns of expression during development and their protein products play significantly different roles in the formation of the HA matrix. Both HAS1 and HAS2 synthesize high molecular-weight HA, whereas HAS3 produces lower molecular weight HA. The expression of the three HAS isoforms is more prominent in growing cells than in resting cells and is differentially regulated by various stimuli, suggesting distinct functional roles of the three proteins. HAS3 produces both secreted and cell-associated forms of hyaluronan and is the most active of the three isoforms of this enzyme in adults. HAS3 gene expression plays a crucial role in the regulation of hyaluronan synthesis in the epidermis. Specifically, IFN-g markedly upregulates HAS3 mRNA, whereas TGF Beta downregulates HAS3 transcript levels. The human HAS3 gene maps to chromosome 16q22.1.
