Myotubularin-related protein 14 (MTMR14), also known as Jumpy, is a myotubularin-related phosphoinositol-3-phosphate (PI3P) phosphatase. Mutations in the MTMR14 gene have been associated with centronuclear myopathy. MTMR14 deficiency in mice leads to altered calcium homeostasis and muscle disorders. MTMR14 has also been shown to play a role in autophagy, a process that is highly regulated by phosphatidylinositides through the type III PI3K, Vps34. MTMR14 was localised to autophagic isolation membranes and early autophagosomes. In these studies, MTMR14 inhibited autophagy and mutations of MTMR14 associated with centronuclear myopathy were also defective in autophagy inhibition. In zebrafish, MTMR14 knockdown was shown to increase the number of autophagosomes, suggesting that its activity is associated with an inhibition of autophagy.

This gene encodes a transmembrane protein containing six epidermal growth factor repeats. The protein is involved in the differentiation of several cell types, including adipocytes; it is also thought to be a tumor suppressor. It is one of several imprinted genes located in a region of on chr 14q32. Certain mutations in this imprinted region can cause phenotypes similar to maternal and paternal uniparental disomy of chromosome 14 (UPD14). This gene is expressed from the paternal allele. A polymorphism within this gene has been associated with child and adolescent obesity. The mode of inheritance for this polymorphism is polar overdominance; this non-Mendelian inheritance pattern was first described in sheep with the callipyge phenotype, which is characterised by muscle hypertrophy and decreased fat mass.

Rab proteins, a family of Ras-related small GTP-binding proteins, play a key role in regulating intracellular vesicle trafficking (1?). Rab GDP dissociation inhibitor (Rab GDI or GDI2) forms a soluble complex with Rab proteins and thereby prevents the exchange of GDP for GTP (1?). In mammals, there exist two major isoforms, Rab GDI alpha (also known as XAP-4) and Rab GDI beta (1,4). While the mammalian Rab GDI beta-genes are ubiquitously expressed, the Rab GDI ?genes are predominantly brain-specific (1). Since it is expressed predominantly in neural and sensory tissues, Rab GDI a may serve a specific function in neural signal transmission (5). The gene sequences for the Rab GDI proteins are extremely conserved in evolution, with substantial homology preserved across three eukaryotic kingdoms (5).

Rad and Gem related GTP binding protein (REM) is a member of the RGK subfamily of Ras-like GTPases that also includes Rad, REM2 and Gem/Kir. REM is a phosphorylated protein that is highly expressed in cardiac muscle and moderately expressed in lung, kidney and skeletal muscle. REM associates with several 14-3-3 isoforms as well as with calmodulin in a calcium-dependent manner. REM mediates two distinct signal transduction pathways that regulate both cytoskeletal reorganization and voltage-gated calcium channel activity. REM decreases the current that passes through cardiac voltage-gated L-type Ca channels (Ca(V)). Overexpression of REM may result in the development of cytoplasmic processes, reorganization of the Actin cytoskeleton, reduction in focal adhesion size and an elongated or dendritic-like cell morphology.

Modulation of the chromatin structure plays an important role in the regulation of transcription in eukaryotes. The nucleosome, made up of four core histone proteins (H2A, H2B, H3 and H4), is the primary building block of chromatin. The N-terminal tail of core histones undergoes different posttranslational modifications including acetylation, phosphorylation and methylation. These modifications occur in response to cell signal stimuli and have a direct effect on gene expression. In most species, the histone H2B is primarily acetylated at lysines 5, 12, 15 and 20. Histone H3 is primarily acetylated at lysines 9, 14, 18 and 23. Acetylation at lysine 9 appears to have a dominant role in histone deposition and chromatin assembly in some organisms. Phosphorylation at Ser10 of histone H3 is tightly correlated with chromosome condensation during both mitosis and meiosis.

Semaphorins are a family of cell surface and secreted proteins involved in neural development that are conserved from insects to humans. Members of this family are approximately 750 amino acids in length (including signal sequences) and are defined by a conserved extracellular “semaphorin†domain of approximately 500 amino acids containing 14-16 cysteines, blocks of conserved sequences and no obvious repeats. The transmembrane semaphorins are characterized by an additional 80 amino acid transmembrane domain and an 80-110 amino acid cytoplasmic domain. SEMA6C, also known as SEMA Y, is a transmembrane protein expressed in fetal brain and adult skeletal muscle. Three isoforms of this semaphorin exist due to alternative splicing: SEMA6C 1, SEMA6C 2 and SEMA6C 3. The extracellular domain of SEMA6C induces growth cone collapse of dorsal root ganglion and plays a role in generation or stability of entorhino-hippocampal synapses.

Cytochrome c oxidase, the terminal component of the mitochondrial respiratory chain, catalyzes the electron transfer from reduced cytochrome c to oxygen. This component is a heteromeric complex consisting of three catalytic subunits encoded by mitochondrial genes, and multiple structural subunits encoded by nuclear genes. The mitochondrially-encoded subunits function in electron transfer, while the nuclear-encoded subunits may function in the regulation and assembly of the complex. This nuclear gene encodes polypeptide 2 (liver isoform) of subunit VIIa, with this polypeptide being present in both muscle and non-muscle tissues. In addition to polypeptide 2, subunit VIIa includes polypeptide 1(muscle isoform), which is present only in muscle tissues, and a related protein, which is present in all tissues. CNTF is a survival promoting factor for different types of neurons in vitro and in vivo. This antibody recognizes a protein of ~35 kDa, identified as CD74. It is a type II transmembrane protein which binds to the peptide binding groove of newly synthesized MHC class II alpha/beta heterodimers and prevents their premature association with endogenous polypeptides. CD74 is expressed primarily by antigen presenting cells, such as B-lymphocytes (from before the pre-B cell stage to before the plasma cell stage), macrophages, and monocytes, and many epithelial cells. Anti-CD74 stains predominantly germinal center lymphocytes and B-cell lymphomas, but rarely T-cell lymphomas. Anti-CD74 has been shown to be useful in differentiating atypical fibroxanthoma (-) from malignant fibrous histiocytoma ( ).
