The mucins are a family of highly glycosylated, secreted proteins with a basic structure consisting of a variable number of tandem repeats (VNTRs). Membrane-associated and secretory mucins are high molecular weight glycoproteins of the glycocalyx (polysaccharide biofilm) that protects mucosal epithelium from particulate matter and microorganisms. Epithelial mucins are large, secreted and cell surface glycoproteins crucial for adhesion modulation, signaling and epithelial cell protection. The number of repeats is highly polymorphic and varies among different alleles. The Mucin family consists of Mucins 1-4, Mucin 5 (AC and B), 6-8, 11-13 and 15-17. The Mucin 16 protein (also commonly referred to as CA125), encoded for by the gene MUC16, is a very high molecular weight tumor antigen consisting of three domains: a carboxy terminal domain, an extracellular domain and an amino terminal domain. Mucin 16, an ovarian cancer-associated antigen, is used as a marker to monitor the progress of epithelial ovarian cancer. It is a hydrophilic membrane-associated protein that may be involved in vitamin A functions.

Twenty human keratins are resolved with two-dimensional gel electrophoresis into acidic (pI 6.0) subfamilies. This antibody cocktail recognises acidic (Type I or LMW) and basic (Type II or HMW) cytokeratins, which include Keratins 1, 3, 4, 5, 6, 8, 10, 14, 15, 16, and 19. Many studies have shown the usefulness of keratins as markers in cancer research and tumor diagnosis. KRTL/KRTH is a broad spectrum anti pan-cytokeratin antibody cocktail, which differentiates epithelial tumors from non-epithelial tumors e.g. squamous vs. adenocarcinoma of the lung, liver carcinoma, breast cancer, and esophageal cancer. It has been used to characterize the source of various neoplasms and to study the distribution of cytokeratin containing cells in epithelia during normal development and during the development of epithelial neoplasms. This antibody stains cytokeratins present in normal and abnormal human tissues and has shown high sensitivity in the recognition of epithelial cells and carcinomas.

T-Cell Intracellular Antigen-1 is a cytoplasmic granule-associated protein expressed in lymphocytes processing cytolytic potential. It is a member of an RNA-binding protein family and possesses nucleolytic activity against cytotoxic lymphocyte (CTL) target cells. It has been suggested that this protein may be involved in the induction of apoptosis as it preferentially recognises poly(A) homopolymers and induces DNA fragmentation in CTL targets. The major granule-associated species is a 15 kDa protein thought to be derived from the carboxyl terminus of the 40 kDa product by proteolytic processing. TIA-1 antibody labels cytotoxic T cells and natural killer cells (NK cells). It is also expressed in T-cell lymphoma, large granular lymphocyte (LGL) leukemia and hairy cell leukemia. TIA1 expression in T-cell malignancies may help in differentiating LGL leukemia (high expression) from T-cell lymphocytosis and other T-cell diseases (low expression). TIA1 may also be used to label tumor-infiltrating lymphocytes in the study of immune response to malignancies.

Alpha-1 Antitrypsin or alpha1-antitrypsin (A1AT) is a protease inhibitor belonging to the serpin superfamily. It is generally known as serum trypsin inhibitor. Alpha 1-antitrypsin is also referred to as alpha-1 proteinase inhibitor (A1PI) because it inhibits a wide variety of proteases. It protects tissues from enzymes of inflammatory cells, especially neutrophil elastase, and has a reference range in blood of 1.5 - 3.5 gram/liter (in US the reference range is generally expressed as mg/dL or micromoles), but the concentration can rise manyfold upon acute inflammation. In its absence (such as in alpha 1-antitrypsin deficiency), neutrophil elastase is free to break down elastin, which contributes to the elasticity of the lungs, resulting in respiratory complications such as emphysema, or COPD (chronic obstructive pulmonary disease) in adults and cirrhosis in adults or children.<p>Like all serine protease inhibitors, A1AT has a characteristic secondary structure of beta sheets and alpha helices. Tweens® are a series of nonionic surfactants derived from sorbitan esters. They are soluble or dispersible in water but differ widely in organic and oil solubilities. Used as oil-in-water emulsifiers in pharmaceuticals, cosmetics, cleaning compounds, etc.
Tween® 80 has been widely used in biochemical applications including: solubilizing proteins, isolating nuclei from cells in culture selective protein extraction growing of tubercule bacilli, and emulsifying and dispersing substances in medicinal and food products. It has little or no activity as an anti-bacterial agent. It has been shown to have an adverse effect on the antibacterial effect of methyl paraben and related compounds. Non-ionic detergent used for selective protein extraction and isolation of nuclei from mammalian cell lines. Soluble/miscible in water to give a clear yellow solution; miscible with alcohol, dioxane, and ethyl acetate; practically insoluble in liquid paraffin and fixed oils (such as mineral oil). Autoclaving of solutions is not recommended. Sterile filtering is suggested with a 0.22 micron filter. Tween® may need to be warmed to about 40 °C and alternated with portions of hot distilled water while being poured through the filter. Store at Room Temperature (15-30 °C).

Antibodies are produced by B lymphocytes, each expressing only one class of light chain. Once set, light chain class remains fixed for the life of the B lymphocyte. In a healthy individual, the total kappa to lambda ratio is roughly 3:1 in serum (measuring intact whole antibodies) or 1:1.5 if measuring free light chains, with a highly divergent ratio indicative of neoplasm.<p>Individual B-cells in lymphoid tissue possess either kappa or lambda light chains, but never both together. Specific rearrangement of lambda light chain of immunoglobulins can lead to loss of some protein coding genes, which does not seem to be functionally relevant (while functionally relevant miR-650 can be overexpressed). Using immunohistochemistry, it is possible to determine the relative abundance of B-cells expressing kappa and lambda light chains. If the lymph node or similar tissue is reactive, or otherwise benign, it should possess a mixture of kappa positive and lambda positive cells. If, however, one type of light chain is significantly more common than the other, the cells are likely all derived from a small clonal population, which may indicate a malignant condition, such as B-cell lymphoma.
