G protein-coupled receptors (GPRs), also known as seven transmembrane receptors, heptahelical receptors or 7TM receptors, comprise a superfamily of proteins that play a role in many different stimulus-response pathways. G protein coupled receptors translate extracellular signals into intracellular signals (G protein activation) and they respond to a variety of signaling molecules, such as hormones and neurotransmitters. GPR110 (G protein-coupled receptor 110), also known as PGR19, is a 911 amino acid protein that belongs to the G-protein coupled receptor 2 family and LN-TM7 subfamily. Characterized as an adhesion GPCR, GPR110 is a multipass membrane-bound protein with a long amino-terminus that contains multiple domains. One of these domains is the GPCR proteolytic site (GPS), which is essential for proteolytic cleavage of the amino-terminus and for cell surface expression.

GUP1 is a 504 amino acid multipass membrane protein of the endoplasmic reticulum that functions as a membrane bound O-acyltransferase. With specific expression in heart, GUP1 negatively regulates amino-terminal palmitoylation of Shh by HHAT, a protein that is required for Shh signaling. Deletion of the gene encoding GUP1 results in higher sensibility to specific sphinogolipid biosynthesis inhibitors and resistance to ergosterol biosynthesis inhibitors, indicating that GUP1 is an essential component in lipid metabolism. Also, GUP1 also seems to be important for cell wall assembly and stability due to evidence in Saccharomyces cerevisiae GUP1 mutants, which exhibit altered plasma membrane lipid composition and membrane potential.

The NGG1 interacting factor 3-like 1 (NIF3L1) is a 377 amino acid protein expressed mainly in the cytoplasm of cells in several different tissues. It has been highly conserved throughout evolution, from bacteria to mammals. NIF3L participates in retinoic acid-primed neural differentiation of P19 embryonic carcinoma cells by cooperating with Trip15/CSN2, a transcriptional corepressor/component of COP9 signalosome. NIF3L1 interacts with itself and with the NIF3L1 binding protein 1 (NIF3L1 BP1), which is a novel protein presumed to contain a leucine zipper domain.

This reference sequence was derived from multiple replicate ESTs and validated by similar human genomic sequence. This gene encodes a member of a family of small membrane proteins that share a 35-amino acid signature sequence domain, beginning with the sequence PFXYD and containing 7 invariant and 6 highly conserved amino acids. The approved human gene nomenclature for the family is FXYD-domain containing ion transport regulator. Transmembrane topology has been established for two family members (FXYD1 and FXYD2), with the N-terminus extracellular and the C-terminus on the cytoplasmic side of the membrane. FXYD2, also known as the gamma subunit of the Na,K-ATPase, regulates the properties of that enzyme. FXYD1 (phospholemman), FXYD2 (gamma), FXYD3 (MAT-8), FXYD4 (CHIF), and FXYD5 (RIC) have been shown to induce channel activity in experimental expression systems. This gene product, FXYD7, is novel and has not been characterized as a protein. [RefSeq curation by Kathleen J. Sweadner, Ph.D., sweadner@helix.mgh.harvard.edu., Dec 2000].

The leucine-rich (LRR) repeat is a 20-30 amino acid motif that forms a hydrophobic å/∫ horseshoe fold, allowing it to accommodate several leucine residues within a tightly packed core. All LRR repeats contain a variable segment and a highly conserved segment, the latter of which accounts for 11 or 12 residues of the entire LRR motif. SLITRK3 (SLIT and NTRK-like family, member 3) is a 977 amino acid single-pass type I membrane protein that contains 20 LRR repeats and belongs to the SLITRK family. Expressed at highest levels in cerebral cortex, SLITRK3 is also found in adult and fetal neural tissues and some astrocytic brain tumors. SLITRK3 functions to suppress neurite outgrowth and plays a role in the regulation of neuronal function. SLITRK3 is encoded by a gene that maps to human chromosome 3, which houses over 1,100 genes, including a chemokine receptor (CKR) gene cluster and a variety of human cancer-related gene loci.

WD-repeats are motifs that are found in a variety of proteins and are characterized by a conserved core of 40-60 amino acids that commonly form a tertiary propeller structure. While proteins that contain WD-repeats participate in a wide range of cellular functions, they are generally involved in regulatory mechanisms concerning chromatin assembly, cell cycle control, signal transduction, RNA processing, apoptosis and vesicular trafficking. WDR40A (WD-repeat-containing protein 40A), also known as DDB1- CUL4-associated factor 12, is a 453 amino acid cytoplasmic protein that contains four WD-repeats. WDR40A is highly expressed in some cancer cell lines, lung cancer tissues and normal testis. A probable substrate receptor for CUL4-DDB1 E3 ubiquitin-protein ligase complex, WRD40A is suggested to interact with DDB1. WDR40A is encoded by a gene mapping to human chromosome 9p13.3. Human chromosome 9 houses over 900 genes and comprises nearly 4% of the human genome.

All LRR repeats contain a variable segment and a highly conserved segment, the latter of which accounts for 11 or 12 residues of the entire LRR motif. The primary function of these motifs is to provide a versatile structural framework to mediate the formation of protein-protein interactions. LRRs are present in a variety of proteins with diverse structure and function, including innate immunity and nervous system development. Several human diseases are associated with mutations in genes encoding LRR-containing proteins. LRRC23 (leucine-rich repeat-containing protein 23), also known as leucine-rich protein B7, is a 343 amino acid protein that contains eight LRR (leucine-rich) repeasts and one LRRCT domain. LRRC23 exists as two alternatively spliced isoforms and is encoded by a gene mapping to chromosome 12.
