Photoreceptor-specific nuclear receptor, also known as NR2E3 or PNR, belongs to a large family of nuclear hormone receptor transcription factors. The proteins belonging to this family are characterized by discrete domains functioning in DNA and ligand binding. NR2E3 has a role in regulating the signaling pathway elemental to the photoreceptor cell function and in regulating pathways involved in embryonic development. NR2E3 is an eye specific nuclear protein found in the outer nuclear layer of the adult retina (where the nuclei of cone and rod photoreceptors are located). Defects in this gene encoding for the protein, which localizes to chromosome 15q22.32, cause enhanced S cone syndrome.

Presenilins are postulated to regulate APP processing through their effects on gamma-secretase, an enzyme that cleaves APP. Also, it is thought that the presenilins are involved in the cleavage of the Notch receptor, such that they either directly regulate gamma-secretase activity or themselves are protease enzymes. Three alternative splice variants of PSEN1 have been identified.

The product of this gene is localized to the Z-line and M-line lattices of myofibrils, where titin's N-terminal and C-terminal regions respectively bind to the sarcomere. ABL2 (or ARG) is a nonreceptor cytoplasmic tyrosine kinase which is closely related to but distinct from ABL1. The similarity of ABL1 and ABL2 includes the tyrosine kinase domains and extends amino-terminal to include the SH2 and SH3 domains. ABL2 is expressed in both normal and tumor cells. It is involved in translocation with the ETV6 gene in human leukemia and has an altered expression in several human carcinomas. Two isoforms of ABL2 with different N-termini (1A and 1B) have been identified. The C-terminal domain of ABL2 contains two F-actin-binding sequences that perform a number of actions related to cell morphology and motility by interacting with actin filaments.

BAMBI transcription regulation is under the influence of beta-catenin, BMP, smad3 and smad4. BAMBI expression can increase in colorectal and hepatocellular carcinomas relative to non-cancerous tissues. BAMBI is expressed at high levels during odontogenesis. BAMBI is coexpressed with Bmp-4 during early Xenopus embryogenesis and can be detected in poorly metastatic human melanoma cell lines.

GNL2 is a nucleolar guanasine-triphosphate binding protein that is ubiquitously expressed at low levels in almost all tissues. GNL2 is involved in the crucial process of trafficking proteins out of the nucleus. Specifically, it is a GTPase that interacts with the 60s preribosomal subunit in the nucleus and facilitates export of the subunit into the cytoplasm. GTPases are responsible for the hydrolysis of GTP by way of a protein region dubbed the G domain. GTPases are often involved in the translocating proteins through membranes gleaning energy for the activity by hydrolizing GTP. GNL2 shares G domain homology and some functionality with nucleostemin (GNL3), another nuclear GTPase. Highest expression of GNL2 is found in testis.
