A gene of chromosome 6q23 encodes the 640 amino acid protein, EYA4 (eyes absent) (1). EYA is one of four members of the eyes absent family (1). A 271 amino acid domain at the carboxyl terminal is highly conserved amongst the members of the eyes absent family (1). EYA4 is expressed in the craniofacial mesenchyme, the dermamyotome, and the limb (1). The conserved region in other EYA proteins interacts with SIX, DACH, and G-proteins, which regulate transcription in early embryonic development (1,2,3,4). SIX translocates EYA1-3 to the nucleus, and G-proteins can stop this interaction (3,4). Premature stop codon mutations in EYA4 cause postlingual, progressive autosomal dominant hearing loss in humans (2). This shows that EYA4 is also vital to the mature organ of Corti (2). EYA4 may cause oculo-dento-digital syndrome, based on its expression pattern and map postion (1).

Calmodulin consists of two glycoproteins, 34 and 39 kDa, sometimes designated epithelial antigen, epithelial specific antigen, and epithelial glycoprotein. The glycoproteins are located on the cell membrane surface and in the cytoplasm of virtually all epithelial cells with the exception of most squamous epithelia, hepatocytes, renal proximal tubular cells, gastric parietal cells and myoepithelial cells. Epithelial Calmodulin is found in the large majority of adenocarcinomas of most sites (50-100% in various studies; as well as neuroendocrine tumours, including small cell carcinoma. Renal cell carcinoma and hepatocellular carcinoma stain in about 30% of the cases. Calmodulin mediates the control of a large number of enzymes and other proteins by Ca(2+). Among the enzymes to be stimulated by the calmodulin Ca(2+) complex are a number of protein kinases and phosphatases. Calmodulin has four functional calcium binding sites.

This antibody recognises a single protein of 33-34 kDa, identified as the Prostate specific antigen. This PSA antibody is highly specific and stains prostatic secretory and ductal epithelium in both normal and neoplastic tissues. Prostate specific antigen is a chymotrypsin-like serine protease (kallikrein family) exclusively produced by the prostate epithelium, and abundant in seminal fluid. PSA can be detected in the sera of patients with prostatic carcinoma. It is predominantly complexed to a liver-derived serine protease inhibitor, alpha-1-antichymotrypsin (ACT). A higher proportion of serum PSA is complexed to ACT in prostate cancer than in benign hyperplasia.

The MEL-14 monoclonal antibody specifically reacts with L- selectin (CD62L), a receptor with lectin-like and Epidermal Growth Factor-like domains. The weight of the CD62L molecules depend on their origin: 74 kDa (on lymphocytes) or 95 kDa (on neutrophils).In the Mouseorganism, CD62L can be expressed by most thymocytes, on subsets of B and T lymphocytes, monocytes, eosinophils, and neutrophils. The L-selectin binds sulfated, fucosylated, and glycosylated glycoproteins (MadCAM-1, GLYCAM-1, and CD 34). It mediates the migration of lymphocytes to the site of inflammation and their return to the peripheral lymphoid tissues and to the HEV (high endothelial venules). In vitro, L-selectin inhibits this binding and the lymphocyte extravasation into peripheral lymph nodes.
