Interleukin-12 (IL-12), also known as Natural Killer Cell Stimulatory Factor (NKSF) or Cytotoxic Lymphocyte Maturation Factor (CLMF), is a heterodimeric pleiotropic cytokine made up of a 40 kDa (p40) subunit and a 35 kDa (p35) subunit. IL-12 is produced by macrophages and B lymphocytes and has been shown to have multiple effects on T cells and Natural Killer (NK) cells. Some of these IL-12 activities include the induction of IFN-gamma and TNF in resting and activated T and NK cells; the enhancement of cytotoxic activity of resting NK and T cells, the stimulation of resting T cell proliferation in the presence of a comitogen; and the enhancement of NK cell proliferation. Current evidence indicates that IL-12 is a key mediator of cellular immunity and induces the differentiation of Th1 cells from precursor T helper cells. Based on its activities, it has been suggested that IL-12 may have therapeutic potential as a vaccine adjuvant that promotes cellular immunity and as an antitumour and anti viral agent.

Recognises a DQ antigen, which is a dimer of 60 kDa. The class II molecule is a heterodimer consisting of an alpha (DQA) and a beta chain (DQB), both anchored in the membrane. It plays a central role in the immune system by presenting peptides derived from extracellular proteins. Class II molecules are expressed in antigen presenting cells (APC: B Lymphocytes, dendritic cells, macrophages). The alpha chain is approximately 33-35 kDa. It is encoded by 5 exons; exon 1 encodes the leader peptide, exons 2 and 3 encode the two extracellular domains, and exon 4 encodes the transmembrane domain and the cytoplasmic tail. Within the DQ molecule both the alpha chain and the beta chain contain the polymorphisms specifying the peptide binding specificities, resulting in up to four different molecules. Typing for these polymorphisms is routinely done for bone marrow transplantation.

EC Blue ist eine einfache, schnelle und zuverlässige Testmethode zur qualitativen und quantitativen Bestimmung von Coliformen und <i>E.coli</i> in Wasser. Geben Sie das zu testende Wasser in einen vorbereiteten, sterilen, nicht fluoreszierenden EC Blue 100-Behälter. Anschließend inkubieren Sie die Mischung bei 35 ±2 °C. Das Medium erfordert keine weiteren Vorbereitungsarbeiten. Dadurch kann der Test ohne großen Aufwand überall durchgeführt werden. Herkömmliche Inkubationsmethoden benötigen etwa drei Tage für den Nachweis von Coliformen oder <i>E.coli</i>. EC Blue hingegen liefert bereits nach 24 Stunden verlässliche Testergebnisse. Der Nachweis basiert auf einer Farbreaktion und ist somit einfach zu interpretieren: Sind coliforme Bakterien vorhanden, färbt sich das Wasser durch die Spaltung des chromogenen Substrats X-Gal blau. Bei einer fäkalen Kontamination durch <i>E.coli</i> können diese Bakterien nur unter UV-Licht (Reaktion mit einem Fluoreszenzfarbstoff) eindeutig nachgewiesen werden. Schwebstoffe, trübe, gelbliche oder gechlorte Wasserproben stören im Allgemeinen nicht den Nachweis. EC Blue ist bei Raumtemperatur bis zu zwei Jahre haltbar, wenn es vor Licht geschützt ist.

3- (4,5-Dimethyl-2-thiazolyl-2)-2,5-diphenyl-tetrazolium bromide is used as a colorimetric metabolic activity indicator in cell viability assays. Thiazolyl Blue Tetrazolium Blue (MTT) is a dye used in measurement of cell proliferation. MTT produces a yellowish solution that is converted to dark blue, water-insoluble MTT formazan by mitochondrial dehydrogenases of living cells. The blue crystals are solubilized with acidified isopropanol and the intensity is measured colorimetrically at 570 nm. MTT has been used as a histochemical/cytochemical reagent and for the detection of NAD. ADP-linked enzyme systems in tissue cannot be detected with MTT, due to binding of the cation by the cyanide trap used. MTT is rapidly reduced to the formazan, which chelates with nickel, copper, and cobalt; the cobalt chelate has been used in oxidative systems.

This antibody recognises proteins of 33, 35 and 41 kDa, which are identified as various isoforms of CD74. The CD74 antibody epitope is localized in the extracellular domain of CD74. CD74 is a type II transmembrane protein which binds to the peptide binding groove of newly synthesised MHC class II alpha/beta heterodimers and prevents their premature association with endogenous polypeptides. The CD74 molecule plays a critical role in the presentation of peptides, by the MHC class II antigens, to CD4 positive lymphocytes. CD74 is expressed on MHC class II positive cells including B cells, a subset of activated T cells, monocytes, dendritic cells and by various types of carcinomas. CD74 is expressed primarily by antigen presenting cells, such as B-lymphocytes (from before the pre-B cell stage to before the plasma cell stage), macrophages, and monocytes, and many epithelial cells. CD74 antibody stains predominantly germinal center lymphocytes and B-cell lymphomas, but rarely T-cell lymphomas. CD74 antibody has been shown to be useful in differentiating atypical fibroxanthoma (-) from malignant fibrous histiocytoma (+).

It recognises a protein of 35 kDa, which is identified as tartrate-resistant acid phosphatase (TRAcP). It exists as two isoforms (5a and 5b). This mAb reacts with both the isoforms. Serum TRAcP 5a is secreted by macrophages and dendritic cells and increased in many patients of rheumatoid arthritis. Serum TRAcP 5b is produced from osteoclasts and elevated during bone resorption. TRAcP is an iron containing glycoprotein, which catalyses the conversion of orthophosphoric monoester to alcohol and orthophosphate. It is the most basic of the acid phosphatases and is the only form not inhibited by L(+)-tartrate. TRAcP is synthesised as a latent proenzyme and is activated by proteolytic cleavage and reduction. Normally, TRAcP is highly expressed by osteoclasts, activated macrophages, neurons and endometrium during pregnancy. Expression of TRAcP is increased in certain pathological conditions such as Leukemic Reticuloendotheliosis (Hairy Cell Leukemia), Gaucher's Disease, HIV-induced Encephalopathy, Osteoclastoma and in osteoporosis and metabolic bone diseases. Anti-TRAcP antibody labels the cells of Hairy Cell Leukemia (HCL) with a high degree of sensitivity and specificity. Other cells stained with this antibody are tissue macrophages and osteoclasts.

This gene encodes tumor protein p73, which is a member of the p53 family of transcription factors involved in cellular responses to stress and development. The family members include p53, p63, and p73 and have high sequence similarity to one another, which allows p63 and p73 to transactivate p53-responsive genes causing cell cycle arrest and apoptosis. The family members can interact with each other in many ways involving direct or indirect protein interactions, resulting in regulation of the same target gene promoters or regulation of each other's promoters. The p73 protein is expressed at very low levels in normal tissues and is differentially expressed in a number of tumors. The p73 gene expresses at least 35 mRNA variants due to the use of alternate promoters, alternate translation initiation sites, and multiple splice variations. Theoretically this can account for 29 different p73 isoforms; however, the biological validity and the full-length nature of most variants have not been determined.
