Pro inflammatory cytokines activate the transcription factor NF kappa B by stimulating the activity of a protein kinase that phosphorylates Ikappa B, an inhibitor of NF kappa B, at sites that trigger its ubiquitination and degradation. A large, cytokine responsive Ikappa B kinase (IKK) complex has been purified and the genes encoding 2 of its subunits have been cloned. These subunits, IKK alpha and Ikk beta, are protein kinases whose function is needed for NF kappa B activation by pro inflammatory stimuli. IKK is composed of similar amounts of IKK alpha, Ikk beta, which are differentially processed forms of a third subunit, IKK gamma. IKK gamma interacts preferentially with IKK beta and is required for the activation of the IKK complex.

Heterotrimeric G proteins function to relay information from cell surface receptors to intracellular effectors. Each of a very broad range of receptors specifically detects an extracellular stimulus (i.e. a photon, pheromone, odorant, hormone or neurotransmitter), while the effectors (e.g. adenyl cyclase), which act to generate one or more intracellular messengers, are less numerous. In mammals, G protein alhfa, beta and gamma polypeptides are encoded by at least 16, 4 and 7 genes, respectively. Most interest in G proteins has been focused on their a subunits, since these proteins bind and hydrolyze GTP and most obviously regulate the activity of the best studied effectors. Evidence, however, has established an important regulatory role for the beta gamma subunits. It is becoming increasingly clear that different G protein complexes expressed in different tissues carry structurally distinct members of the gamma as well as the alhfa and beta subunits, and that preferential associations between members of subunit families increase G protein functional diversity.

PCDHB6 (Protocadherin beta-6) is a 794 amino acid single pass transmembrane protein that is one of 16 proteins in the protocadherin beta cluster. Unlike the alpha and gamma gene clusters whose genes are spliced to downstream constant region exons during transcription, members of the beta cluster (such as PCDHB6) do not use constant-region exons to produce mRNAs. As a result, each protocadherin beta gene encodes the transmembrane, extracellular and short cytoplasmic domains of the protein. PCDHB6 is likely a calcium-dependent cell adhesion protein that is involved in the maintenance of neural connections in the brain. Unlike most protocadherin-beta proteins, PCDHB6 has not one but two PXXP motifs within its cytoplasmic domain, suggesting a role in signal transduction cascade events.

IKK beta (I-Kappa-B kinase-beta) is a member of the IKK complex which is composed of IKK alpha, IKK beta, IKK gamma and IKAP. Phosphorylation of I-Kappa-B on a serine residue by the IKK complex frees NF-kB from I-Kappa-B and marks it for degradation via ubiquination. IKK beta has been shown to activate NF-kB and phosphorylate IKB alpha and beta. Phosphorylation of 2 sites at the activation loop of IKK beta is essential for activation of IKK by TNF and IL1. Once activated, IKK beta autophosphorylates which in turn decreases IKK activity and prevents prolonged activation of the inflammatory response.
