Caspase-4 Antibody: Caspases are a family of cysteine proteases that can be divided into the apoptotic and inflammatory caspase subfamilies. Unlike the apoptotic caspases, members of the inflammatory subfamily are generally not involved in cell death but are associated with the immune response to microbial pathogens. Members of this subfamily include caspase-1, -4, -5, and -12. Activation of these caspases results in the cleavage and activation of proinflammatory cytokines such as IL-1 beta and IL-18. Caspase-4 was initially identified as a homologous protein to Caspase-1 and the C. elegans Ced-3 which could induce apoptosis in transfected cells. More recent studies have shown that it can be activated by ER stress and has been suggested to be involved in multiple neuronal pathologies such as Alzheimer's disease.

SPP1 Antibody: The secreted protein 1 (SPP1), also known as osteopontin, is a major noncollagenous protein of bone, but is also found in the extracellular matrix of other mineralized tissues and in bodily fluids. In bone, SPP1 is produced by osteoblasts, osteocytes, macrophages, and osteoclasts (1,2). SPP1 binds to cells through integrin and non-integrin receptors, as well as the adhesion receptor CD44 in an RGD-independent manner, enabling SPP1 to induce a number of functional effects including macrophage chemotaxis, cytoprotection, and regulation of T helper type 1 cells. SPP1 can regulate biomineralization by inhibiting the formation of hydroxyapatite (3) and the growth of calcium oxalate crystals.

Members of this superfamily are widely represented throughout the animal kingdom and have been implicated in a variety of developmental processes. Proteins of the TGF beta superfamily are disulfide-linked dimers composed of two 12-15 kDa polypeptide chains. As implied by their name, BMPs initiate, promote and regulate bone development, growth, remodeling and repair. Smad1 translocation to the nucleus is observed after the addition of BMP4 (also designated BMP2B), suggesting that BMP4 may play a role in activation of the Smad pathway. BMP is secreted into the extracellular matrix.

FAM19A4 is a secreted, 12 kDa member of the FAM19/TAFA family of chemokine-like proteins. Like other members of the FAM19/TAFA family, with the exception of TAFA5, mature FAM19A4 contains 10 regularly spaced cysteine residues. The FAM19A4 proteins are predominantly expressed in specific regions of the brain and the biological functions of FAM19A4 family members remain to be determined, but there are a few tentative hypotheses. First, FAM19A4 may modulate immune responses in the CNS by functioning as brain specific chemokines, and may act with other chemokines to optimize the recruitment and activity of immune cells in the CNS. Second, FAM19A4 may represent a novel class of neurokines that act as regulators of immune nervous cells. And third, FAM19A4 may control axonal sprouting following brain injury.
