The charged multivesicular body proteins, commonly designated CHMPs, belong to the vacuolar sorting protein family and function as chromatin-modifying proteins. CHMP1-6 are all components of ESCRT (endosomal sorting complex required for transport) I, II or III complexes. These complexes are crucial for sorting endosomal articles into multivesicular bodies (MVBs), and are also required for the formation of these bodies. CHMP2B, also known as CHMP2.5 or vacuolar protein-sorting-associated protein 2-2, is a 213 amino acid cytosolic protein. Widely expressed in brain, heart, skeletal muscle, small intestine, pancreas, lung, placenta and leukocytes, CHMP2B associates directly with CHMP2A and vps4 for the disassembly of the ESCRT-III complex. Defects in the gene encoding CHMP2B have been shown to cause chromosome 3-linked frontotemporal dementia (FTD3).

The pantothenate kinase (PANK) family of proteins catalyzes the first step in coenzyme A (CoA) biosynthesis. Coenzyme A is an important coenzyme involved in the synthesis and oxidation of fatty acids, as well as the oxidation of pyruvate in the citric acid (Krebs) cycle. Pantothenate kinase 3 (PANK3) is a 370 amino acid member of the pantothenate kinase family that plays a role in the physiological regulation of the intracellular CoA concentration. Localized to the cytoplasm, PANK3 is regulated by feedback inhibition by CoA and its thioesters. PANK3 transfers a phosphate from ATP to pantothenate (Vitamin B5), resulting in formation of 4’-phosphopantothenate. Closely related to its family members, PANK1, PANK2 and PANK4, PANK3 is highly expressed in liver. Pantothenate kinase associated neurodegeneration (PKAN) results from mutations in the gene encoding PANK2, the only mitochondria targeted human PANK.

FRAT1 and FRAT2 were originally characterized as proteins frequently rearranged in advanced T cell lymphoma, and they have since been identified as proto-oncogenes involved in tumorigenesis. These proteins share significant homology with the Xenopus glycogen synthase kinase-3 (xGSK-3) binding protein, which is designated GBP and is essential for the formation of the dorsal-ventral axis during embryonic development. Establishment of these embryonic axes is mediated by the Wnt intracellular signaling pathway. Wnt signaling is regulated in part by the activity of GSK-3, which phosphorylates and thereby facilitates the degradation of ?catenin. GBP binds to GSK-3 and inhibits this phosphorylation, resulting in the accumulation of ?catenin and the subsequent transcription of Wnt target genes. Like GBP, FRAT2 has been shown to bind xGSK-3, suggesting that FRAT1 and FRAT2 may be GSK-3 regulatory proteins.
