A unique family of Cysteine proteases has been described that differs in sequence, structure and substrate specificity from any previously described protease family. This family, termed CED-3/ICE, functions as key components of the apoptotic machinery and act to destroy specific target proteins which are critical to cellular longevity. Nuclear lamins are critical to maintaining the integrity of the nuclear envelope and cellular morphology as components of the nuclear lamina, a fibrous layer on the nucleoplasmic side of the inner nuclear membrane, which is thought to provide a framework for the nuclear envelope and may also interact with chromatin. B-type lamins undergo a series of modifications, such as farnesylation and phosphorylation. Increased phosphorylation of the lamins occurs before envelope disintegration and probably plays a role in regulating lamin associations. Nuclear Lamin B is fragmented as a consequence of apoptosis by an unidentified member of the ICE family.

APPBP1 is a member of the ubiquitin-activating E1 family. In fetal tissues APPBP1 is widely expressed and in adult tissues it is expressed throughout the brain. APPBP1 is a cell membrane associated protein and functions as the regulatory subunit in a heterodimer with UBA3. The APPBP1/UBA3 complex binds to and activates NEDD8, a ubiquitin-like protein involved in signal transduction, cell proliferation and development. This suggests that APPBP1 affects a variety of cellular functions. In addition, APPBP1 is essential for cell cycle progression through the S/M checkpoint. More specifically, it inhibits the entry into S phase and promotes entry into M phase.

Existing as thirteen alternatively spliced isoforms, Neuron navigator 2 displays 3' to 5' helicase activity and exonuclease activity and participates in the catalysis of ATP to ADP. Neuron navigator 2 is highly expressed in the nervous system of developing embryos and colon carcinomas, as well as in liver, brain and kidney. Lower levels of expression are found in lung, thyroid, spinal cord, heart, placenta and mammary gland. Neuron navigator 2 belongs to the Nav/unc-53 family and contains one CH (calponin-homology) domain.

These genes similarly contain sequences that encode for the HMG-box domain, which is responsible for the sequence-specific DNA-binding activity. Sox genes encode putative transcriptional regulators implicated in the decision of cell fates during development and the control of diverse developmental processes. The highly complex group of Sox genes cluster at least 40 different loci that rapidly diverged in various animal lineages. At present, 30 Sox genes have been identified. Members of this family have been shown to be conserved during evolution and to play key roles during animal development. Some are involved in human diseases, including sex reversal.

ADAM11 was first described as MDC (Metalloproteinase-like disintergin-like cysteine-rich protein) from analysis of human brain libraries, in search of brain-specific proteins. Two splice variants with different carboxyterminal ends were described. The message was found only in the brain in this publication. Another group identified ADAM11 in the human brain, where ADAM11 was thought to be involved in cell migration and spatial patterning. ADAM11 was mapped to 17q21.3, a region of interest for breast cancer, and mutations in ADAM11 are associated with some breast cancers. Retinoic acid caused a doubling in ADAM11 message levels over 24 hours in NT2/D1 cells, a human embryonic carcinoma cell line. ADAM11 null mutant mice have deficits in spatial learning and motor coordination, although they did have normal cell migration and differentiation during development. ADAM11 is a member of the ADAMs family (A Disintegrin And Metalloproteinase), but does not contain the canonical HExxHxxxxH zinc-binding metalloproteinase catalytic site. The domain structure of the full-length ADAM11 includes a signal sequence, propeptide domain, metalloproteinase-like domain, disintegrin-like domain, cys-rich domain, EGF-like domain, a spacer region, then the transmembrane domain and a short cytoplasmic domain.

ERp19, ERp29, ERp46, ERp57 and ERp72 may act as proteases, protein disulfide isomerases, thiol-disulfide oxidases, phospholipases or a combination of these. ERp19, also designated thioredoxin domain-containing protein 12 (TXNDC12), and ERp46, also designated thioredoxin domain containing 5 (TXNDC5), belong to the thioredoxin superfamily and contain a thioredoxin fold with a consensus active-site sequence (CxxC). Both ERp19 and ERp46 are widely expressed ER luminal proteins that are most abundant in the liver and are enriched in purified liver ER vesicles. ERp19 shows significant protein thiol-disulfide oxidase activity in vitro, which is dependent on the presence of both active-site cysteines.

FIG4 is a 907 amino acid protein that contains one SAC domain through which it is thought to function as a phosphoinositide phosphatase that may play an important role in signal transduction and vesicle trafficking. Defects in the gene encoding FIG4 are the cause of Charcot-Marie-Tooth disease type 4J (CMT4J) and amyotrophic lateral sclerosis type 11 (ALS11), both of which negatively affect the nervous system. CMT4J is a recessive demyelinating disorder of the peripheral nervous system and is characterized by reduced motor nerve conduction velocities and axonal degeneration. Unlike CMT4J, ALS11 is characterized by the degeneration of upper motor neurons in the brain and lower neurons in the spinal cord, causing paralysis and, ultimately, death.
