3- (4,5-Dimethyl-2-thiazolyl-2)-2,5-diphenyl-tetrazolium bromide is used as a colorimetric metabolic activity indicator in cell viability assays. Thiazolyl Blue Tetrazolium Blue (MTT) is a dye used in measurement of cell proliferation. MTT produces a yellowish solution that is converted to dark blue, water-insoluble MTT formazan by mitochondrial dehydrogenases of living cells. The blue crystals are solubilized with acidified isopropanol and the intensity is measured colorimetrically at 570 nm. MTT has been used as a histochemical/cytochemical reagent and for the detection of NAD. ADP-linked enzyme systems in tissue cannot be detected with MTT, due to binding of the cation by the cyanide trap used. MTT is rapidly reduced to the formazan, which chelates with nickel, copper, and cobalt; the cobalt chelate has been used in oxidative systems.

The matrix metalloproteinases (MMPs) are a family of at least eighteen secreted and membrane bound zincendopeptidases. Collectively, these enzymes can degrade all the components of the extracellular matrix, including fibrillar and non fibrillar collagens, fibronectin, laminin and basement membrane glycoproteins. In general, a signal peptide, a propeptide, and a catalytic domain containing the highly conserved zinc binding site characterizes the structure of the MMPs. In addition, fibronectin like repeats, a hinge region, and a C terminal hemopexin like domain allow categorization of MMPs into the collagenase, gelatinase, stomelysin and membrane type MMP subfamilies. All MMPs are synthesised as proenzymes, and most of them are secreted from the cells as proenzymes. Thus, the activation of these proenzymes is a critical step that leads to extracellular matrix breakdown. MMPs are considered to play an important role in wound healing, apoptosis, bone elongation, embryo development, uterine involution, angiogenesis and tissue remodelling, and in diseases such as multiple sclerosis, Alzheimer's, malignant gliomas, lupus, arthritis, periodontis, glumerulonephritis, atherosclerosis, tissue ulceration, and in cancer cell invasion and metastasis.MMP17 has been reported to be elevated in several tumor cell lines, and is constituitively produced by some normal cell lines. Treatment of cells with Concanavolin A or the phorbol ester TPA stimulates production of MMP17 in some cell types, and the enzyme can be recovered in cell lysates. Shed forms of MMP17 have also been reported.

JMJD2A Antibody: Members of the Jumonji domain 2 (JMJD2) family contain a JmjN domain, a JmjC domain, a JD2H domain, two TUDOR domains, and two PHD-type zinc fingers. The first member of this group, JMJD2A, is widely expressed in human tissues and cell lines and functions as a trimethylation-specific demethylase, converting specific trimethylated histone residues to the dimethylated form, and as a transcriptional repressor. JMJD2A can also form a complex with the androgen receptor (AR), a transcription factor that is pivotal for the development of prostate cancer. Overexpression of JMJD2A stimulates AR function and this stimulation is dependent on JMJD2A catalytic activity, suggesting that JMJD2A might be a critical protein with roles in cell proliferation and oncogenesis.

Butyrylcholine Esterase (BCHE) is a secreted protein that belongs to the type-B carboxylesterase/lipase family. BCHE is a major acetylcholine hydrolyzing enzyme in the circulation. It is detected in blood plasma and present in most cells except erythrocytes. BCHE is an esterase with broad substrate specificity. BCHE can contribute to the inactivation of the neurotransmitter acetylcholine. BCHE can degrade a large number of neurotoxic organophosphate esters. Thus, it plays important pharmacological and toxicological roles and is thought to be involved in the pathological progression. Defects in BCHE are the cause of butyrylcholinesterase deficiency (BChE deficiency) which is a metabolic disorder characterised by prolonged apnoea after the use of certain anesthetic drugs, including the muscle relaxants succinylcholine and other ester local anesthetics.
