PLEKHM3 Antibody: PLEKHM3, also known as DAPR, is a member of the M family of Pleckstrin homology domain-containing proteins. PLEKHM3 was initially identified through chromatin immunoprecipitation and CpG microarray analysis examining proteins regulated by myocyte-enhancing factor 2. In C2C12 myoblast cells, PLEKHM3 binds to the PI3K signaling member protein kinase B in the cytosol prior to differentiation into myotubes. Following the initiation of differentiation, PLEKHM3 was also found in membrane fractions. Knockdown of PLEKHM3 expression by RNAi resulted in the inhibition of myotube formation, suggesting that PLEKHM3 is a key component required by myoblasts for orchestrating their differentiation during myogenesis.

WIPI2 Antibody: WD repeat proteins play a role in many essential biologic functions, regulating the assembly of multiprotein complexes by presenting a beta-propeller platform for simultaneous and reversible protein-protein interactions. WIPI2, also known as ATG18B or ATG21, is a human homolog to yeast ATG18 and contains three WD repeats and has a 7-bladed propeller structure with a conserved motif that facilitates its interaction with other proteins. It is recruited to early autophagosomal structures along with Atg16L and ULK1 and is required for the formation of LC3-positive autophagosomes. Along with the highly related WIPI1, WIPI2 is found at the plasma membrane in addition to autophagosomal membranes.

PRTFDC1 Antibody: Phosphoribosyl transferase domain containing 1 (PRTFDC1) is highly homologous to the hypoxanthine phosphoribosyltransferase (HPRT1) and may have arisen from a gene duplication event of a common ancestor gene. Recently, it was shown that CpG islands in the PRTFDC1 promoter could be hypermethylated in ovarian cancers and oral squamous-cell carcinomas (OSCC), leading to gene silencing. Restoration of PRTFDC1 expression in OSCC inhibited cell growth in colony-formation assays, while knockdown of PRTFDC1 expression in OSCC that expressed the gene promoted cell growth. These results suggest that PRTFDC1 can act as a tumor-suppressor gene. At least three isoforms of PRTFDC1 are known to exist.

This reaction is mediated by a class of crosslinkers known as carbodiimides and results in the formation of an amide bond. The reaction with EDC, the most common carbodiimide crosslinker, is generally performed in an MES buffer at pH 4,5 to 5 and requires just minutes to complete.

Insulin-like growth factors (IGFs) comprise a family of endocrine, paracrine and autocrine polypeptides consisting of the ligands IGF1 and IGF2, two receptors (IGF1R, IGF2R), at least 6 IGF-binding proteins (IGFBPs) and IGFBP proteases. Among the binding proteins, IGFBP6 is unique because of its N-terminal disulphide linkages and its marked binding preference for IGF2. It is a potent inhibitor of the interaction between IGF2 and its receptor IGF1R, thus preventing major functions of IGF2, such as induction of proliferation, differentiation, cell adhesion, or colony formation. In particular, IGFBP-6 inhibited the growth of neuroblastoma and rhabdomyosarcoma xenografts. GFBP-6 is expressed in many tissues, including lung, liver, gut and the central nervous system.

This antibody reacts with the Ig domain of CD47 protein. It has been shown to inhibit polymorphonuclear neutrophil (PMN) transmigration across cell monolayers and matrix. CD47, originally named integrin-associated protein (IAP), is a 50 kDa protein containing five membrane-spanning sequences and a short cytoplasmic tail. CD47 plays a role in both cell adhesion by acting as an adhesion receptor for THBS1 on platelets, and in the modulation of integrins. It is important in memory formation and synaptic plasticity in the hippocampus. CD47 may play a role in membrane transport and/or integrin dependent signal transduction.

Plexin Domain-Containing Protein 1 (PLXDC1) is a single-pass type I membrane protein that belongs to the plexin family. Secreted PLXDC1 is localized predominantly at the tight junctions of vascular endothelial cells and to a lesser extent at the luminal surface of vascular endothelial cells. PLXDC1 is expressed in fibrovascular membrane with increased expression in individuals with proliferative diabetic retinopathy. It can detect in endothelial cells from colorectal cancer, and in endothelial cells from primary cancers of the lung, liver, pancreas, breast and brain. PLXDC1 interacts with NID1 and may also interact with CTTN. It plays a important role in endothelial cell capillary morphogenesis, the proliferation and maintenance of neovascular endothelial cells in the formation of fibrovascular membranes (FVMs).

MCM7 encodes a protein that is one of the highly conserved mini-chromosome maintenance proteins (MCM) that are essential for the initiation of eukaryotic genome replication. The hexameric protein complex formed by the MCM proteins is a key component of the pre-replication complex (pre_RC) and may be involved in the formation of replication forks and in the recruitment of other DNA replication related proteins. The MCM complex consisting of this protein and MCM2, 4 and 6 proteins possesses DNA helicase activity, and may act as a DNA unwinding enzyme. Cyclin D1-dependent kinase, CDK4, is found to associate with this protein, and may regulate the binding of this protein with the tumorsuppressor protein RB1/RB.

Caveolin proteins are proposed to be scaffolding proteins for organizing and concentrating certain caveolin-interacting molecules. Mutations identified in this gene lead to interference with protein oligomerization or intra-cellular routing, disrupting caveolae formation and resulting in Limb-Girdle muscular dystrophy type-1C (LGMD-1C), hyperCKemia or rippling muscle disease (RMD). Alternative splicing has been identified for this locus, with inclusion or exclusion of a differentially spliced intron. In addition, transcripts utilize multiple polyA sites and contain two potential translation initiation sites.
