GPR120, a member of the rhodopsin family of G protein-coupled receptors (GPCRs), is a 377 amino acid protein which is expressed in the intestine. GPR120 is a receptor for unsaturated long-chain FFAs (free fatty acids). FFAs act as signaling molecules and are an important energy source. They also employ various physiological responses through their GPCRs. One such response occurs when dietary FFAs stimulate GPR120. This stimulation promotes the secretion of glucagon-like peptide 1 (GLP-1) <i>in vivo</i> and <i>in vitro</i>. GLP-1 belongs to the class of molecules known as the incretins, which are associated with insulin secreted from the pancreas as a result of food intake. GLP-1 also inhibits glucagon and gastric acid secretion and gastric emptying. Consequently, the role of GPR120 in the secretion of GLP-1 is critical in the treatment of diabetes.

The pantothenate kinase (PANK) family of proteins catalyzes the first step in coenzyme A (CoA) biosynthesis. Coenzyme A is an important coenzyme involved in the synthesis and oxidation of fatty acids, as well as the oxidation of pyruvate in the citric acid (Krebs) cycle. Pantothenate kinase 3 (PANK3) is a 370 amino acid member of the pantothenate kinase family that plays a role in the physiological regulation of the intracellular CoA concentration. Localised to the cytoplasm, PANK3 is regulated by feedback inhibition by CoA and its thioesters. PANK3 transfers a phosphate from ATP to pantothenate (Vitamin B5), resulting in formation of 4?-phosphopantothenate. Closely related to its family members, PANK1, PANK2 and PANK4, PANK3 is highly expressed in liver. Pantothenate kinase associated neurodegeneration (PKAN) results from mutations in the gene encoding PANK2, the only mitochondria targeted human PANK.

GOX is a 370 amino acid protein that is expressed in liver and pancreas. HAO1 is localized to peroxisomes and aids in organic acid metabolism via 2-hydroxyacid oxidase activity. 2-hydroxyacid oxidases, such as HAO1, are enzymes that require a flavin cofactor to oxidize 2-hydroxyacids to 2-ketoacids while reducing oxygen to hydrogen peroxide. HAO1 prefenentially oxidizes the substrate glycolate and also oxidizes other substrates, including 2-hydroxy fatty acids as well as L-?hydroxy acids of moderately short chain lengths. The oxidation of glycolate yields glyoxylate which is utilized for peroxisomal synthesis of glycine. HAO1 is also able to convert glyoxylate to oxalate. HAO1 is thought to play a role in the pathophysiology of hyperoxaluria type 1, which is caused by defects in AGXT, a peroxisomal enzyme, leading to accumulation of glyoxylate. Hyperoxaluria type 1 is characterized by an accumulation of oxalate that is thought to lead to precipitates of calcium oxalate in kidneys which can be fatal.

Predominantly localized to the nucleolus, BOP1 (Block of proliferation 1 protein) is a 746 amino acid highly conserved non-ribosomal protein that is involved in ribosome biogenesis. Truncation of the amino terminus of BOP1 leads to cell growth arrest in the G1 phase and specific inhibition of 28S and 5.8S rRNA synthesis, as well as a deficit in the cytosolic 60S ribosomal subunit. This suggests that BOP1 is involved in the formation of mature rRNAs and in the biogenesis of the 60S ribosomal subunit. BOP1 physically interacts with pescadillo (a protein involved in cell proliferation) and enables efficient incorporation of pescadillo into the nucleolar preribosomal complexes, thereby affecting rRNA maturation and the cell cycle. The BOP1-pescadillo complex is also necessary for biogenesis of 60S ribosomal subunits. Deregulation of BOP1 may lead to colorectal tumorigenesis.

HIC-2 is highly expressed in cerebellum and is localised to the nucleus in cells. HIC-2 contains a short amino acid sequence that is thought to interact with CtBP, a transcriptional repressor. The gene sequence associated with HIC-2 is thought to be a target for miRNAs (microRNAs) which are expressed in many cancers, suggesting that HIC-2 could possess tumor suppressor capabilities.
