Complete remission of acute promyelocytic leukemia can be achieved by treating patients with retinoic acid, and PML-RAR-a (promyelocytic leukemia-retinoic acid receptor alpha fusion protein) plays a major role in mediating retinoic acid effects in leukemia cells. The retinoic acid-induced gene, PRAM-1 (PML-RAR-a target gene encoding an adaptor molecule 1) encodes an adaptor protein which is expressed and modulated during normal human myelopoiesis. PRAM-1 expression is hindered by expression of PML-RAR-a. The 718 amino acid PRAM-1 protein contains eight N-terminal proline-rich repeats and several proline residues that are clustered as type I or type II SH3 recognition motifs. PRAM-1 demonstrates expression in hematopoietic tissues and lung.

Hyperpolarisation-activated, cyclic nucleotide-binding channels (HCN) are voltage-gated cation channels that are activated by direct binding of intracellular cyclic nucleotides. The HCN family consists of four members (HCN1-4), each with a core transmembrane segment domain and a C-terminal 120 amino-acid cyclic nucleotide-binding domain motif. HCN channels are expressed in the brain, heart, thalamus and testis. The pacemaker properties of HCN channels contribute to spontaneous rhythmic activity in the brain and heart. HCN3 contains a segment characterised by a series of positively charged amino acids at every third position. This region designated S4 is likely to be the voltage sensor of the protein. In the brain, HCN3 and HCN4 exhibit subcortical distribution mainly concentrated in the hypothalamus and thalamus, respectively.

Myotrophin (V-1 protein) is a ubiquitously expressed cytoplasmic protein that can translocate to the nucleus during sustained NFkB activation. The gene encoding for this protein localizes to chromosome 7q33. Myotrophin may be involved in cerebellar morphogenesis and contains an acetylated N-terminus and 2.5 internal 33 amino acid ankyrin repeats. It is important in the differentiation of cerebellar neurons, particularly of granule cells. The 117 amino acid protein has been associated with, and able to induce, cardiac hypertrophy. Myotrophin increases protooncogene, ANF and Beta-Myosin heavy chain transcript levels. Myotrophin is upregulated when myocytes undergo cyclic stretch or are treated with tumor necrosis factor Alpha (TNF Alpha) or interleukin-1Beta. Highest levels of Myotrophin are detected in brain and lowest levels in skeletal muscle.

RILPL2 is a 211 amino acid protein that belongs to the RILPL family. RILPL2 does not regulate lysosomal morphology or distribution. RILPL2 shares 32% and 18% amino acid identity with RILPL1 and RILP, respectively. RILPL2 as a novel interacting partner for the actin-based molecular motor MyoVa, and has a novel role for RILPL2 in controlling neuronal morphogenesis. It has been suggested that there is also a novel role for RILPL2 in the regulation of cellular shape and dendritic-spine morphogenesis, probably via the Rac1-Pak pathway. PCR analysis of human tissues detects highest RILPL2 expression in lung, followed by placenta. Lower expression is detected in liver, kidney, pancreas, heart and brain, but no expression is detected in skeletal muscle. The RILPL2 gene is conserved in chimpanzee, dog, cow, mouse, rat, chicken and zebrafish, and maps to human chromosome 12q24.31.

A gene of chromosome 6q23 encodes the 640 amino acid protein, EYA4 (eyes absent) (1). EYA is one of four members of the eyes absent family (1). A 271 amino acid domain at the carboxyl terminal is highly conserved amongst the members of the eyes absent family (1). EYA4 is expressed in the craniofacial mesenchyme, the dermamyotome, and the limb (1). The conserved region in other EYA proteins interacts with SIX, DACH, and G-proteins, which regulate transcription in early embryonic development (1,2,3,4). SIX translocates EYA1-3 to the nucleus, and G-proteins can stop this interaction (3,4). Premature stop codon mutations in EYA4 cause postlingual, progressive autosomal dominant hearing loss in humans (2). This shows that EYA4 is also vital to the mature organ of Corti (2). EYA4 may cause oculo-dento-digital syndrome, based on its expression pattern and map postion (1).
