This gene encodes a DNA topoisomerase, an enzyme that controls and alters the topologic states of DNA during transcription. This nuclear enzyme is involved in processes such as chromosome condensation, chromatid separation, and the relief of torsional stress that occurs during DNA transcription and replication. It catalyzes the transient breaking and rejoining of two strands of duplex DNA which allows the strands to pass through one another, thus altering the topology of DNA. Two forms of this enzyme exist as likely products of a gene duplication event. The gene encoding this form, alpha, is localized to chromsome 17 and the beta gene is localized to chromosome 3. The gene encoding this enzyme functions as the target for several anticancer agents and a variety of mutations in this gene have been associated with the development of drug resistance. Reduced activity of this enzyme may also play a role in ataxia-telangiectasia.

JMJC domain-containing histone demethylases are also likely involved in development via their ability to regulate gene expression. JMJD7 contains one JMJC domain and was originally thought to be an alternatively spliced isoform of PLA2G4B.

The combination of an X and Y chromosome lead to normal male development while two copies of X lead to normal female development. More than one copy of the X chromosome with a Y chromosome causes Klinefelter's syndrome. A single copy of X alone leads to Turner's syndrome. More than 2 copies of the X chromosome, in the absence of a Y chromosome, is known as Triple X syndrome. Color blindness, hemophilia, and Duchenne muscular dystrophy are well known X chromosome-linked conditions which affect males more frequently as males carry a single X chromosome. Some F box proteins have been shown to be critical for the ubiquitin mediated degradation of cellular regulatory proteins. In fact, F box proteins are one of the four subunits of ubiquitin protein ligases, called SCFs. SCF ligases bring ubiquitin conjugating enzymes to substrates that are specifically recruited by the different F box proteins. A large family of mammalian F box proteins has recently been identified and classified into three groups based on the presence of either the WD 40 repeats, the leucine rich repeats, or the presence or absence of other protein protein interacting domains. The FBXW2 gene product, the second identified member of the F box gene family, contains multiple WD 40 repeats.

Fe65L is a 758 amino acid protein that contains one WW domain and two PID domains. Binding to the intracellular domain of the ∫-Amyloid precursor protein, Fe65L is thought to modulate the internalization and, therefore, the accessibility and function of ∫-Amyloid. Multiple isoforms of Fe65L exist due to alternative splicing events.
