Sox genes comprise a family of genes that are related to the mammalian sex determining gene SRY. These genes similarly contain sequences that encode for the HMG-box domain, which is responsible for the sequence-specific DNA-binding activity. Sox genes encode putative transcriptional regulators implicated in the decision of cell fates during development and the control of diverse developmental processes. The highly complex group of Sox genes cluster at least 40 different loci that rapidly diverged in various animal lineages. At present, 30 Sox genes have been identified. Members of this family have been shown to be conserved during evolution and to play key roles during animal development. Some are involved in human diseases, including sex reversal.

The protein binds to and is activated by neuregulins and other factors and induces a variety of cellular responses including mitogenesis and differentiation. Activity of c-Abl protein is negatively regulated by its SH3 domain, and deletion of the SH3 domain turns ABL1 into an oncogene. The t(9;22) translocation results in the head-to-tail fusion of the BCR (MIM:151410) and ABL1 genes present in many cases of chronic myelogeneous leukemia. The DNA-binding activity of the ubiquitously expressed ABL1 tyrosine kinase is regulated by CDC2-mediated phosphorylation, suggesting a cell cycle function for ABL1. Members of the leucine-rich repeat family includes LRCH1, LRCH2, LRCH3 and LRCH4. All family members contain one calponin-homology domain and nine leucine-rich repeats. The best characterised leucine-rich repeat family member is LRCH4, which is suggested to be involved in ligand binding in the brain, with expression observed primarily in the hippocampus. As a cell adhesion molecule and signal receptor, LRCH4 may play an important role in maintenance of hippocampus-dependent memories, with defects in the gene possibly contributing to a loss of long-term memory. The gene encoding LRCH3 maps to human chromosome 3, which spans 200 million base pairs and encodes between 1,100 and 1,500 genes. There are three isoforms of LRCH3 that are produced as a result of alternative splicing events.

Beta-arrestin 2, a receptor-regulated MAP kinase scaffold protein, is found to interact with and stimulate the phosphorylation of this kinase by MAP kinase kinase 4 (MKK4). Cyclin-dependent kianse 5 (CDK5) can phosphorylate and inhibit the activity of this kinase, which may be important in preventing neuronal apoptosis. Four alternatively spliced transcript variants encoding distinct isoforms have been reported.

MIB1 is a 1006 amino acid E3 ubiquitin ligase that activates the Notch ligand, Delta. MIB1 ubiquinates Delta by binding to its intracellular domain, leading to the endocytosis and eventual degradation of the Delta receptor, which, paradoxically, results in the up-regulation of receptor activity and enhances Notch signaling. MIB1 also interacts with DAPK, a protein that plays an important role in the regulation of apoptosis. Ubiquination of DAPK leads to inhibition of caspase-dependent apoptosis, therefore it is likely that overexpression of MIB1 can lead to tumor growth. Although it seems to be widely expressed at low levels, MIB1 is expressed at highest concentrations in the CNS and ovary. Both DAPK and MIB1 are overexpressed in epileptic brain tissue, suggesting that they probably cooperate as regulators of neuronal death in epilepsy.

Limd1 (LIM domain-containing protein 1) is a ubiquitously expressed tumor suppressor containing 3 LIM zinc-binding domains. LIM domains consist of a cysteine-rich consensus sequence containing two distinct zinc-binding subdomains, which mediate protein-protein interactions. Limd1 interacts with the proteins SQSTM1, Rb, p62 and TRAF6. Limd1 was first identified when the deletion of its gene was noted in some cervical cancers. Limd1 blocks <i>in vitro</i> and <i>in vivo</i> tumor growth and is down-regulated in lung cancer. Limd1 may regulate osteoclast development under stressful conditions via its interactions with TRAF6 and p62.

Leupaxin is highly expressed in lymphoid tissues such as spleen, lymph node, thymus and appendix, with low expression in bone marrow and fetal liver. Consisting of four leucine-rich LD-motifs at the N-terminus and four LIM domains at the C-terminus, leupaxin associates with a member of the focal adhesion kinase family, PYK2, in lymphoid cells. The leupaxin and PYK2 complex is involved in cell type-specific signaling in which it regulates signaling at sites of adhesion. Leupaxin is a substrate for tyrosine kinase in lymphoid cells and is suggested to participate in and be regulated by tyrosine kinase activity. Leupaxin may be a potential progression marker for a subset of prostate cancer and may act as a novel coactivator of the androgen receptor.

Amisyn is a mostly cytosolic protein related to Tomosyn which plays an important role in SNARE complex assembly. Amisyn contains a v-SNARE coiled coil homology domain that binds to Syntaxin 1A and weakly to Syntaxin 4. Three isoforms exist for Amisyn. Isoform 1 is the full length protein, isoform 2 has a different amino acid sequence between residues 204-210 and isoform 3 is missing amino acids 1-102 and contains a different sequence for amino acids 103-150. Amisyn lacks a transmembrane domain and therefore is unable to assemble into a functional, membrane-anchored SNARE complex. This suggests that Amisyn may instead be acting to maintain SNARE conformation and facilitate the binding of VAMP-2. Amisyn can inhibit exocytosis independent of Syntaxin binding.
