This gene, a member of the cadherin superfamily, encodes one of the receptor tyrosine kinases, which are cell-surface molecules that transduce signals for cell growth and differentiation. This gene plays a crucial role in neural crest development, and it can undergo oncogenic activation <i>in vivo</i> and <i>in vitro</i> by cytogenetic rearrangement. Mutations in this gene are associated with the disorders multiple endocrine neoplasia, type IIA, multiple endocrine neoplasia, type IIB, Hirschsprung disease, and medullary thyroid carcinoma. Two transcript variants encoding different isoforms have been found for this gene. Additional transcript variants have been described but their biological validity has not been confirmed.

This form is a late adult onset form of the disease (after age 20 years). The patients show a benign course, most of them remaining ambulatory 10 to 40 years after clinical onset.

KCNT2 is a 1,135 amino acid multi-pass transmembrane protein belonging to the potassium channel family (calcium-activated subfamily) of proteins. KCNT2 produces rapidly activating outward rectifier potassium currents in reponse to high intracellular sodium and chloride levels. Its channel activity is inhibited by ATP, inhalation anesthetics, such as isoflourane, and upon stimulation of G-protein coupled receptors, such as mAChR M1 and GluR-1. There are four isoforms of KCNT2 that are produced as a result of alternative splicing events.

Eukaryotic elongation factor 2 kinase (EEF2k) previously known as Ca2+/calmodulin dependent protein kinase III, is an abundant cytoplasmic protein highly specific for elongation factor 2 (eEf2). Phosphorylation of eEF2 by eEF2 kinase on specific threonine residues results in the inactivation of eEF-2 and in termination of mRNA translation. The activity of eEF2 kinase is not only dependent upon Ca2+ ions, calmodulin (CaM) and insulin, but is also regulated both negatively and positively via phosphorylation by different protein kinases (AMPK, S6K1, p90 RSK). There is also evidence that eEF-2 phosphorylation is involved in the regulation of cell cycle progression, cellular differentiation, oogensis and malignant tumors.
