The protein tyrosine phosphatase (PTP) family of proteins are Signalling molecules that regulate processes such as cell growth, cell differentiation, oncogenic transformation and the mitotic cycle. PCPTP1, also known as PTPRR (Receptor-type tyrosine-protein phosphatase R), ECPTP, PTPBR7 or PTPRQ, is a 657 amino acid protein that functions to sequester inactive mitogen-activated protein kinases (MAPKs) to the cytoplasm. Expressed primarily in the brain with weaker expression in other parts of the body, PCPTP1 is a receptor-like molecule that is able to dephosphorylate MAPKs, thereby rendering them inactive. Three isoforms of PCPTP1 exist and are designated alpha, beta and gamma. The alpha form is localised to the cell membrane, while the beta and gamma forms are localised to the perinuclear areas within the cytoplasm.

It binds to androgen and glucocorticoid receptors and up-regulates their transcriptional activity, thereby functioning as a nuclear receptor co-activator. Role of NRIP has been implicated in cell growth and also in cervical and prostrate cancer, thus indicating a potential therapeutic intervention. Northern Blot analysis detects a high expression of NRIP in skeletal muscle and testis and low expression in heart, prostrate and adrenal gland.

Predominantly localized to the nucleolus, BOP1 (Block of proliferation 1 protein) is a 746 amino acid highly conserved non-ribosomal protein that is involved in ribosome biogenesis. Truncation of the amino terminus of BOP1 leads to cell growth arrest in the G1 phase and specific inhibition of 28S and 5.8S rRNA synthesis, as well as a deficit in the cytosolic 60S ribosomal subunit. This suggests that BOP1 is involved in the formation of mature rRNAs and in the biogenesis of the 60S ribosomal subunit. BOP1 physically interacts with pescadillo (a protein involved in cell proliferation) and enables efficient incorporation of pescadillo into the nucleolar preribosomal complexes, thereby affecting rRNA maturation and the cell cycle. The BOP1-pescadillo complex is also necessary for biogenesis of 60S ribosomal subunits. Deregulation of BOP1 may lead to colorectal tumorigenesis.

Bloom’s syndrome is an autosomal recessive disorder characterized by pre- and post-natal growth deficiencies, sun sensitivity, immunodeficiency and a predisposition to various cancers. The gene responsible for Bloom’s syndrome, BLM, encodes a protein homologous to the RecQ helicase of E. coli and is mutated in most Bloom’s syndrome patients. One characteristic of Bloom’s syndrome is an increased frequency of sister chromatid exchange (SCE). BLM has been shown to unwind G4 DNA, and a failure of this function is thought to be responsible for the increased rate of SCE. BLM is known to be translocated to the nucleus, where its ATPase activity is stimulated by both single- and double-stranded DNA. Mutations in the yeast SGS1, a homolog of BLM, are known to cause mitotic hyperrecombination similiar to that observed in Bloom’s cells.

Constitutive expression of a VAV2 oncoprotein may result in morphological alterations including highly enlarged cells in which karyokinesis and cytokinesis frequently are uncoupled.

MYBPC1 is a 1141 amino acid protein that contains three fibronectin type-III domains and seven Ig-like C2-type domains. Existing as a member of the immunoglobulin superfamily, MYBPC1 functions as a thick filament-associated protein that localizes to striated muscle bands in vertebrae and is thought to modify the activity of select ATPases. Additionally, MYBPC1 may play a role in the modulation of muscle contraction and in the overall structural integrity of the cell. The gene encoding MYBPC1 maps to human chromosome 12, which encodes over 1100 genes and comprises approximately 4.5% of the human genome. Chromosome 12 is associated with a variety of diseases and afflictions, including hypochondrogenesis, achondrogenesis, Kniest dysplasia, Noonan syndrome and Trisomy 12p, which causes facial developmental defects and seizure disorders.

Characteristics of the BTB domain suggest that ZBTB44 functions as a transcription regulator. Four isoforms of ZBTB44 have been identified.

The GLP2 receptor (GLP2R) is a G protein-coupled receptor superfamily member closely related to the glucagon receptor ans GLP1 receptor. Glucagon-like peptide-2 (GLP2) is a 33-amino acid proglucagon-derived peptide produced by intestinal enteroendocrine cells. FRAT1 and FRAT2 were originally characterised as proteins frequently rearranged in advanced T cell lymphoma, and they have since been identified as proto-oncogenes involved in tumourigenesis. These proteins share significant homology with the Xenopus glycogen synthase kinase-3 (xGSK-3) binding protein, which is designated GBP and is essential for the formation of the dorsal-ventral axis during embryonic development. Establishment of these embryonic axes is mediated by the Wnt intracellular Signalling pathway. Wnt Signalling is regulated in part by the activity of GSK-3, which phosphorylates and thereby facilitates the degradation of catenin. GBP binds to GSK-3 and inhibits this phosphorylation, resulting in the accumulation of catenin and the subsequent transcription of Wnt target genes. Like GBP, FRAT2 has been shown to bind xGSK-3, suggesting that FRAT1 and FRAT2 may be GSK-3 regulatory proteins.

Dual specificity phosphatases (DSPs) are a subclass of the protein tyrosine phosphatase (PTP) gene superfamily, which are selective for dephosphorylating critical phosphothreonine and phosphotyrosine residues within MAP kinases. DSP gene expression is induced by a host of growth factors and/or cellular stresses, thereby negatively regulating MAP kinase superfamily members including MAPK/ERK, SAPK/JNK and p38. MKP-5 preferentially binds to p38, but also to SAPK/JNK. It is ubiquitously expressed and localizes to both the cytoplasm and the nucleus. MKP-5 has been implicated in cell proliferation and apoptosis, tumor invasion and immune responses.
