Beschreibung:   Cyclin dependent kinase 5 (Cdk5) is a key regulator of cell cycle progression in neuronal differentiation that physically associates with and is activated by the neuron-specific protein p35. CDK5RAP1 (Cdk5 regulatory subunit-associated protein 1), also known as Cdk5 activator-binding protein C42, is a 601 amino acid protein that specifically inhibits Cdk5 activation by p35 through formation of a dimer that inhibits kinase activity. CDK5RAP1 contains one TRAM domain, which is thought to bind tRNA and deliver the RNA-modifying enzymatic domain to its target. There are 4 named isoforms of CDK5RAP1 that are produced as a result of alternative splicing events and are expressed at high levels in heart and skeletal muscle.

Beschreibung:   CD109 is a a novel member of the alpha2-macroglobulin (alpha2M)/complement superfamily. It is a glycosylphosphatidylinositol (GPI)-linked cell surface glycoprotein of approximately 170 kd found onendothelial cells, activated platelets and T-cells and on a subset of hematopoietic stem and progenitor cells. Although it has been suggested that T-cell CD109 may play a role in antibody-inducing T-helper function and it is known that platelet CD109 carries the Gov alloantigen system, the role of CD109 in hematopoietic cells remains largely unknown.CD109 has been identified as part of the TGF-beta receptor system in human keratinocytes and upregulation of CD109 expression has been observed in several different types of tumour.

Beschreibung:   Receptor for Wnt proteins. Most of frizzled receptors are coupled to the beta-catenin canonical signaling pathway, which leads to the activation of disheveled proteins, inhibition of GSK-3 kinase, nuclear accumulation of beta-catenin and activation of Wnt target genes. A second signaling pathway involving PKC and calcium fluxes has been seen for some family members, but it is not yet clear if it represents a distinct pathway or if it can be integrated in the canonical pathway, as PKC seems to be required for Wnt-mediated inactivation of GSK-3 kinase. Both pathways seem to involve interactions with G-proteins. May be involved in transduction and intercellular transmission of polarity information during tissue morphogenesis and/or in differentiated tissues.

Beschreibung:   EBP1 is a member of the peptidase M24C family and functions as an RNA-binding protein involved in cellular proliferation and differentiation processes. It is expressed in a variety of cell lines, including a wide range of tumor cell lines, and localizes to the cytoplasm. Upon treatment with Neuregulin-1 (heregulin), EBP1 translocates to the nucleus. EBP1 is a component of pre-ribosomal ribonucleoprotein complexes, participating in ribosome assembly and regulating the later steps of rRNA processing. In addition, EBP1 interacts with ErbB-3 and may function as a modulator of the ErbB-3-mediated signal transduction pathway by regulating the effects of Neuregulin-1 (heregulin). EBP1 also associates with histone deacetylases (HDACs), functioning as a transcriptional co-repressor of cell cycle regulatory genes.

Beschreibung:   Paroxon is an organophosphorus anticholinesterase compound, used topically in the treatment of glaucoma. It is produced in vivo in mammals by microsomal oxidation of the insecticide parathion. Parathion is inert until transformed to paroxon. Paroxonase or PON is an arylesterase that is capable of hydrolyzing paroxon to produce p-nitrophenol. PONs are nonspecific and their classification is based not only on substrate specificity but also on tissue distribution, inhibition properties and physicochemical characteristics such as electrophoretic mobility and molecular weight. In contrast to PON1, which is expressed mainly in the liver, PON2 is expressed in a variety of mouse tissues, including the pancreas. PON3 is a associated with the high density lipoprotein fraction of serum. The genes which encode PON1-3 are physically linked and map to human chromosome 7q21.3.

Beschreibung:   OTUB1 is a 271 amino acid protein that contains one OTU (ovarian tumour) domain and belongs to the OTU family of predicted cysteine proteases. Expressed as two isoforms (one of which is present throughout the body and the other of which is present only in lymphoid tissues), OTUB1 functions as a hydrolase that can remove ubiquitin residues from target proteins, thereby preventing protein degradation and playing an important role in protein turnover. OTUB1 interacts with GRAIL and, via this interaction, plays a role in the regulation and the induction of T-cell anergy, a phenomenon that occurs when T-cells are rendered unresponsive to their cognate antigens. Due to its interaction with GRAIL, OTUB1 is an important regulator of immune responses in secondary lymphoid organs.

Beschreibung:   CCDC11 is a 514 amino acid protein encode by a gene that maps to human chromosome 18q21.1. Encoding over 300 genes, chromosome 18 contains about 76 million bases. Trisomy 18, or Edwards syndrome, is the second most common trisomy after Downs syndrome. Symptoms of Edwards syndrome include low birth weight, a variety of physical development defects, heart deformations and breathing difficulty. Translocation between chromosome 18 and 14 is the most common translocation in cancers, and occurs in follicular lymphomas. Niemann-Pick disease, hereditary hemorrhagic telangiectasia and erythropoietic protoporphyria are associated with chromosome 18. The TGF∫ modulators, Smad2, Smad4 and Smad7 are encoded by chromosome 18.

Beschreibung:   The Thrombospondin proteins, Thrombospondins 1-4 and Thrombospondin 5 (also designated COMP), compose a family of glycoproteins that are involved in cell-to-cell and cell-to-matrix signaling. These extracellular, cell-surface proteins form complexes of both homo- and heteromultimers. Spondin-2, or Mindin, is also designated DIL-1 for its differential expression in cancerous and non-cancerous lung cells. Full-length SPON2 cDNA encodes a 331 amino acid protein with a domain arrangement similar to zebrafish F-Spondin and Mindin-1/Mindin-2: an FS1 domain, an FS2 domain, a hydrophobic signal sequence in the N-terminus and a Thrombospondin type I repeat. Immunoblot analysis demonstrates expression of dimers and oligomers in a concentration-dependent manner under nonreducing conditions.

Beschreibung:   Increase in fetal surfactant synthesis and lung maturity is caused by the glucocorticoidal induction of enzymes required for phosphatidylcholine synthesis towards the end of gestation (1). The regulation of gestational age-dependent induction of phosphatidylcholine synthesis by glucocorticoids is still unclear (1). The rate-controlling enzyme in the phosphatidylcholine biosynthetic pathway is CTP-phosphocholine cytidylyltransferase A (CCT A) (2–4). In cultured eukaryotic cells, this enzyme is essential for survival (3). The alpha isoform is located in the nucleus and is regulated by reversible phosphorylation and membrane association (3). There is significant identity between the alpha-helical membrane-binding domains of CCT A and soybean oleosin (2). Expressed CCT A has lipid-dependent cytidylyltransferase activity (5). The gene which encodes CCT A maps to human chromosome 3q (4).

Beschreibung:   IRGM belongs to a family of interferon-gamma inducible GTPases, belonging to the GTPase superfamily, and are selectively induced by IFN-gamma or bacterial lipopolysaccharide (LPS) stimulation. IRGM is primarily expressed in all cells derived from B-cell lineages, and is highly expressed in macrophages following IFN-gamma stimulation. IRGM has been shown to be required for host defences against a broad range of intracellular pathogens. Specifically, IRGM deficient mice show defects with intracellular microbial killing, phagosome maturation and autophagy. More recently IRGM has been shown to inhibit baseline hematopoietic proliferation and is required for a normal hematopoietic stem cell response to chemical and infectious stimuli.

Beschreibung:   Insulin receptor substrates (IRS) are responsible for several insulin related activities, such as glucose homeostasis, cell growth, cell transformation, apoptosis and insulin signal transduction. Serine/threonine phosphorylation of IRS1 has been demonstrated to be a negative regulator of insulin signaling and is responsible for its degradation, although IRS1 degradation pathways are not well understood. IRS1 has also been shown to be constitutively activated in cancers such as breast cancer, Wilm's tumors, and adrenal cortical carcinomas, thus making IRS1 phosphorylation and subsequent degradation an attractive therapeutic target. To date there have been four subtypes identified: IRS1, 2, 3 and 4, with IRS1 being widely expressed.

Beschreibung:   NFKB1 or NFKB2 is bound to REL, RELA, or RELB to form the NFKB complex. The NFKB complex is inhibited by I-kappa-B proteins (NFKBIA, or NFKBIB), which inactivate NF-kappa-B by trapping it in the cytoplasm. Phosphorylation of serine residues on the I-kappa-B proteins by kinases (IKBKA, or IKBKB) marks them for destruction via the ubiquitination pathway, thereby allowing activation of the NF-kappa-B complex. Activated NFKB complex translocates into the nucleus and binds DNA at kappa-B-binding motifs such as 5-prime GGGRNNYYCC 3-prime or 5-prime HGGARNYYCC 3-prime (where H is A, C, or T; R is an A or G purine; and Y is a C or T pyrimidine). For some genes, activation requires NFKB interaction with other transcription factors, such as STAT, AP1 (JUN), and NFAT.

Beschreibung:   G protein-coupled receptors (GPRs), also known as seven transmembrane receptors, heptahelical receptors or 7TM receptors, comprise a superfamily of proteins that play a role in many different stimulus-response pathways. G protein coupled receptors translate extracellular signals into intracellular signals (G protein activation) and they respond to a variety of signaling molecules, such as hormones and neurotransmitters. GPR146 (G protein-coupled receptor 146), also known as PGR8, is a 333 amino acid multi-pass transmembrane protein that belongs to the G-protein coupled receptor 1 family. Characterized as an orphan receptor for which its endogenous ligand has yet to be identified, GPR146 is thought to play a role in signaling events throughout the cell.
