MCM3 is one of the highly conserved mini-chromosome maintenance proteins that are involved in the initiation of eukaryotic genome replication. The hexameric protein complex formed by MCM proteins is a key component of the pre-replication complex (pre_RC) and may be involved in the formation of replication forks and in the recruitment of other DNA replication related proteins. This protein is a subunit of the protein complex that consists of MCM2-7. It has been shown to interact directly with MCM5/CDC46. This protein also interacts with, and thus is acetlyated by MCM3AP, a chromatin-associated acetyltransferase. The acetylation of this protein inhibits the initiation of DNA replication and cell cycle progression.

The enzyme encoded is responsible for catalyzing the production of gamma-aminobutyric acid from L-glutamic acid. A pathogenic role for this enzyme has been identified in the human pancreas since it has been identified as an autoantibody and an autoreactive T cell target in insulin-dependent diabetes. CLN8, a 286 amino acid transmembrane protein, localizes mainly to the endoplasmic reticulum, but also partially to the ER-Golgi intermediate compartment (ERGIC). Mutations in the CLN8 gene cause neuronal ceroid lipofuscinosis 8 and progressive epilepsy with mental retardation (EPMR). Both disorders are forms of neuronal ceroid-lipofuscinose (NCL), a group of progressive neurodegenerative diseases found in children, characterized by failure of psychomotor development, impaired vision, seizures and premature death. The CLN8 protein is one of eight proteins in the CLN family, including CLN1-CLN7, which are associated with NCL.

The PSD-95/SAP 90 family of proteins, which are known to bind to and cluster various membrane proteins, are involved in the organization of synaptic structure. SynGAP (for synaptic GTPase activating protein), a novel synaptic RasGAP, is a brain-specific protein abundant in the cortex, hippocampus and olfactory bulb. SynGAP interacts with all three PDZ domains within PSD-95/ SAP 90 proteins. It represents one of three classes of mammalian RasGAPs and is specifically localized to excitatory synapses possessing NMDA receptors. SynGAP may be involved in the regulation of BDNF as well as Ras signaling. Its activity is inhibited by phosphorylation by CaMKII, which could result in the activation of the MAP kinase pathway.

mTOR is one of a family of proteins involved in cell cycle progression, DNA recombination, and DNA damage detection. In rat, it is a 289-kDa protein (symbolized RAFT1) with significant homology to the Saccharomyces cerevisiae protein TOR1 and has been shown to associate with the immunophilin FKBP12 in a rapamycin dependent fashion. The FKBP12-rapamycin complex is known to inhibit progression through the G1 cell cycle stage by interfering with mitogenic signaling pathways involved in G1 progression in several cell types, as well as in yeast. The binding of FRAP to FKBP12-rapamycin correlated with the ability of these ligands to inhibit cell cycle progression.

PTCHD2 is a 1,392 amino acid multi-pass membrane protein that contains one SSD (sterol-sensing) domain and belongs to the patched family. Expressed in retina, brain and testis, PTCHD2 localizes to endoplasmic reticulum and colocalizes with cholesterol. PTCHD2 overexpression leads to increased cholesterol levels, suggesting that PTCHD2 may play a role in cholesterol homeostasis. PTCHD2 is further hypothesized to act as a link between thyroid hormone and cholesterol metabolism. Existing as two alternatively spliced isoforms, PTCHD2 is thought to assist in the release of lipid-anchored secreted proteins and is encoded by a gene that maps to human chromosome 1p36.22.

DYX1C1 encodes a nuclear tetratricopeptide repeat domain protein that is dynamically regulated in brain. In human brain, DYX1C1 protein localizes to a fraction of cortical neurons and white matter glial cells. It is also expressed in lung, kidney and testis.
