Prosapip2 Antibody: Prosapip2 (TBKBP1) is essential for signal transduction during viral infection thus plays a major role in the TNF/NF-kappa B pathway. It is an adaptor protein that constitutively binds TBK1 (TANK-binding kinase) and IKBKE and may play a role in antiviral innate immunity. Prosapip2 is a 615 amino acid adaptor protein belonging to the fibrillar collagen family, consisting of trimers of identical alpha 1 chains which are linked to each other by interchain disulfide bonds. It has a ubiquitous expression with highest levels in ovary, followed by the neuronal system. Prosapip2 binds to TBK1 and helps in the activation of IRF3 which controls the expression of antiviral genes during infection. Recent studies show that Prosapip2 is an interaction partner of ProSAP2/Shank3 and actin, suggesting a role as a linker molecule between postsynaptic density and the cytoskeleton.

The amine-reactive portion of SPDP reagents is the N-hydroxysuccinimide (NHS) ester. Reactions are most commonly performed in phosphate, carbonate/bicarbonate, or borate buffers at pH 7-8. Other buffers can be used provided they do not contain primary amines (or thiols or disulfide reducing reagents – see discussion of the sulfhydryl reactivity). The rate of reaction and degradation by hydrolysis increases with increasing pH; for example, the half-life of the NHS ester is several hours at pH 7 and less than 10 minutes at pH 9. NHS-ester reagents like SPDP and LC-SPDP have limited aqueous solubility and must be dissolved in organic solvent before adding them to a reaction mixture. Sulfo-NHS-ester reagents like Sulfo-LC-SPDP are water-soluble and can be added directly to aqueous reaction mixtures.

beta -Defensin 4A is a membrane-active cationic peptide that functions in inflammation and innate immune responses. There are at least 30 beta -Defensins, which are distinguished from alpha-Defensins by the connectivity pattern of their three intermolecular disulfide bonds. Members of the Defensin family are highly similar in protein sequence. This gene encodes Defensin, DEFB4;, which has broad-spectrum antimicrobial activity and may play an important role in innate epithelial defense. They are highly expressed in skin and tonsils, and to a lesser extent in trachea, uterus, kidney, thymus, adenoid, pharynx and tongue. beta -Defensin 4A has low expression in salivary gland, bone marrow, colon, stomach, polyp and larynx. No expression in small intestine. The 45 amino acid mature human BD3 shares 38% and 33% amino acid sequence identity with mouse and rat BD3, respectively.

The PDPH is a heterobifunctional crosslinker containing sulfhydryl-reactive pyridyldithiol and carbonyl-reactive hydrazide moieties. Pyridyldithiols react with free sulfhydryls (-SH) to form disulfide bonds. Hydrazide groups react with carbonyls (aldehydes and ketones) to form stable hydrazone bonds. Aldehyde groups can be created by periodate-oxidation of sialic acid and other sugar components of glycoprotein polysaccharides. Thus, PDPH is useful for conjugating glycoproteins and sulfhydryl-containing peptides or proteins. Likewise, PDPH is useful as a sulfhydryl-addition reagent for glycoproteins and other carbohydrates; after reaction of the hydrazide to an oxidized carbohydrate, the pyridyldithiol group can be cleaved by a reducing agent to expose a sulfhydryl group. Yet another application for PDPH is to react the primary amine of the hydrazide moiety to a carboxyl group using the crosslinker EDC.

Serine Protease Inhibitor Kazal-Type 4 (SPINK4) is a secreted protein containing one Kazal-like domain. SPINK4 is a member of the SPINK protein family. The gene family of serine protease inhibitors of the Kazal type (SPINK) are functional and positional candidate genes for celiac disease (CD). SPINK1 plays an important role in protecting the pancreas against excessive trypsinogen activation. It is a potent natural inhibitor of pancreatic trypsin activity. SPINK1 mutations are associated with the development of acute and chronic pancreatitis and have been detected in all forms of chronic pancreatitis. SPINK2 functions as a trypsin/acrosin inhibitor and is synthesized mainly in the testis and seminal vesicle where its activity is engaged in fertility. The SPINK2 protein contains a typical Kazal domain composed by six cysteine residues forming three disulfide bridges. SPINK9 was identified in human skin. Its expression was strong in palmar epidermis, but not detectable or very low in non palmoplantar skin.

Interleukin 23 (IL-23) is a heterodimeric cytokine composed of two disulfide-linked subunits, a p19 subunit that is unique to IL-23, and a p40 subunit that is shared with IL-12. The p19 subunit has homology to the p35 subunit of IL-12, as well as to other single chain cytokines such as IL-6 and IL-11. The p40 subunit is homologous to the extracellular domains of the hematopoietic cytokine receptors. Although p19 is expressed by activated macrophages, dendritic cells, T cells, and endothelial cells, only activated macrophages and dendritic cells express p40 concurrently to produce IL-23. IL-23 has biological activities that are similar to, but distinct from IL-12. Both IL-12 and IL-23 induce proliferation and IFN-gamma production by human T cells. IgM is the first antibody generated in an immune response to an antigen. It is generally a pentamer with each of the five immunoglobulins linked together with disulfide bonds. In its pentamer form, it has a molecular mass of 970 kDa and 10 antigen binding sites (due to the large size of most antigens, not all binding sites can be filled simultaneously. IgM antibodies account for approximately 5%-10% of all the antibody in the body.<p>This mAb is specific for the human IgM heavy chain. It does not cross-react with other immunoglobulin heavy chains, T-cells, monocytes, granulocytes, or erythrocytes. IgM antibody is useful in the identification of certain cancer types. ITGA4 encodes integrin alpha chain 4. Integrins are heterodimeric integral membrane proteins composed of an alpha chain and a beta chain. Unlike other integrin alpha chains, alpha 4 neither contains an I-domain, nor undergoes disulfide-linked cleavage. Although alpha 4 is variably cleaved to yield two unlinked chains, this cleavage is unnecessary for integrin assembly and function. Alpha 4 combines with beta 1 (ITGB1) on T-cells to form the integrin very late (activation) antigen 4 (VLA-4) that can bind to the extracellular matrix molecules fibronectin or thrombospondin, and is also a ligand for the cell surface molecule vascular cell adhesion molecule 1 (VCAM-1). In addition, alpha 4 combines with beta 7 to form the lymphocyte homing receptor known as LPAM-1 (lymphocyte Peyer Patch adhesion molecule 1). Integrins are also known to participate in cell-surface mediated signalling.
