The 2.43 antibody reacts with the 32-34 kDa alpha subunit of mouse CD8, known as CD8a or CD8 alpha. CD8a can form a homodimer (CD8 alpha-alpha), but is more commonly expressed as a heterodimer with a second chain known as CD8b or CD8 beta. CD8 acts as a co-receptor in antigen recognition and subsequent T cell activation induced by binding of the T cell receptor (TCR) to antigen-bearing MHC Class I molecules. The tetratricopeptide repeat (TPR) motif is a degenerate, 34 amino acid sequence found in many proteins and acts to mediate protein-protein interactions in various pathways. At the sequence level, there can be up to 16 tandem TPR repeats, each of which has a helix-turn-helix shape that stacks on other TPR repeats to achieve ligand binding specificity. TTC21B (tetratricopeptide repeat domain 21B), also known as THM1, is a 1,316 amino acid protein that contains 19 TPR repeats and belongs to the TTC21 family. Localizing to cytoplasm and cytoskeleton, TTC21B exists as two alternatively spliced isoforms and is thought to negatively regulate Shh signal transduction. TTC21B may also be involved in retrograde intraflagellar transport in cilia, and is encoded by a gene that maps to human chromosome 2q24.3.

The human ataxin-7 gene, also known as spinocerebellar ataxia 7 or SCA7, maps to chromosome 3p13-p12, has a 2,727-bp open reading frame, and encodes a 892 amino acid protein containing a nuclear localization signal and a polyglutamine tract (1,2). SCA7 is an autosomal dominant neurodegenerative disorder characterized by ataxia and selective neuronal cell loss caused by the expansion of a translated CAG repeat encoding a polyglutamine tract in ataxin-7, which is the SCA7 gene product (3,4). Ataxin-7 is expressed within neurons both affected and unaffected in SCA7 pathology with subcellular localization being variable depending upon the neuronal subtype (5). Polyglutamine expanded in ataxin-7 may carry out its pathogenic effects in the nucleus by altering the matrix-associated nuclear structure and/or by disrupting nucleolar function (6).

Calmodulin consists of two glycoproteins, 34 and 39 kDa, sometimes designated epithelial antigen, epithelial specific antigen, and epithelial glycoprotein. The glycoproteins are located on the cell membrane surface and in the cytoplasm of virtually all epithelial cells with the exception of most squamous epithelia, hepatocytes, renal proximal tubular cells, gastric parietal cells and myoepithelial cells. Epithelial Calmodulin is found in the large majority of adenocarcinomas of most sites (50-100% in various studies; as well as neuroendocrine tumours, including small cell carcinoma. Renal cell carcinoma and hepatocellular carcinoma stain in about 30% of the cases. Calmodulin mediates the control of a large number of enzymes and other proteins by Ca(2+). Among the enzymes to be stimulated by the calmodulin Ca(2+) complex are a number of protein kinases and phosphatases. Calmodulin has four functional calcium binding sites.

IL-34 Antibody: Interleukin-34 (IL-34) was identified through a functional screening of a library of secreted proteins after transfection of an embryonic kidney cell line with recombinant cDNAs. Flow cytometry and monocyte viability assays demonstrated increased survival and/or proliferation of monocytes induced by IL-34 and specific binding of IL-34 to CD14+ monocytes. IL-34 is a second ligand to the colony stimulating factor (CSF)-1 receptor in addition to CSF1. Like CSF1, IL-34 stimulates phosphorylation of ERK1 and ERK2 in human monocytes and induces differentiation of bone marrow cells into macrophages.
