Beschreibung:   This MAb reacts with both SUMO-2 and SUMO-3. The small ubiquitin-related modifier (SUMO) proteins, which include SUMO-1, 2 and 3, belong to the ubiquitin-like protein family. Like ubiquitin, the SUMO proteins are synthesized as precursor proteins that undergo processing before conjugation to target proteins. Also, both utilize the E1, E2 and E3 cascade enzymes for conjugation. However, SUMO and ubiquitin differ with respect to targeting. Ubiquitination predominantly targets proteins for degradation, whereas sumoylation targets proteins to a variety of cellular processing, including nuclear transport, transcriptional regulation, apoptosis and protein stability. The unconjugated SUMO-1, 2 and 3 proteins localize to the nuclear membrane, nuclear bodies and cytoplasm, respectively. SUMO-1 utilizes Ubc9 for conjugation to several target proteins, which include MDM2, p53, PML and RanGap1. SUMO-2 and 3 contribute to a greater percentage of protein modification than does SUMO-1 and unlike SUMO-1, they can form polymeric chains. In addition, SUMO-3 regulates beta-Amyloid generation and may be critical in the onset or progression of Alzheimer’s disease.

Beschreibung:   WASP (for Wiskott-Aldrich syndrome protein) and N-WASP are downstream effectors of Cdc42 that are implicated in Actin polymerization and cytoskeletal organization. The WASP family also includes VASP (vasodilator-stimulated phosphoprotein) and Mena (for mammalian enabled protein), which accumulate at focal adhesions and are also involved in the regulation of the Actin cytoskeleton. The WAVE proteins are related to the WASP family proteins and are likewise involved in mediating Actin reorganization downstream of the Rho family of small GTPases. The protein homologs WAVE1 and WAVE2 regulate membrane ruffling by inducing the formation of Actin filament clusters in response to GTP binding and by activating Rac. They mediate Actin polymerization by cooperating with the Arp2/3 complex, thereby promoting the formation of Actin filaments. WAVE1, which is also designated SCAR (suppressor of cAR), is expressed primarily in the brain, while WAVE2 is widely expressed, with the expression highest in peripheral blood leukocytes. WAVE3 forms a multiprotein complex that links receptor kinases with Actin and plays a role in the transduction of signals involving changes in cell shape, function or motility.

Beschreibung:   The Ras superfamily of GTPases can be subdivided into the Ras, Rho/Rac, Sar, Rab, Arf, Rap and Ran subfamilies, all of which control multiple aspects of cell function, including cytoskeletal rearrangement, nuclear signaling and cell growth. The Ras superfamily of GTPases function as regulated switches that toggle between a biologically active GTP-bound and an inactive GDP-bound form. This activation is catalyzed by guanine nucleotide exchange factors (GEFs). RIC-8A (resistance to inhibitors of cholinesterase 8 homolog A), also known as RIC8 or Synembryn-A, is a 530 amino acid cytoplasmic protein that can activate several G-alpha proteins, including G?i-1, G?q and G?o. Functioning as a guanine nucleotide exchange factor, RIC-8A binds to GDP-associated substrates and exchanges bound GDP for free GTP. Via its ability to stimulate protein function, RIC-8A plays a role in regulating mitotic movement and may be involved in receptor-mediated ERK activation. RIC-8A is expressed as three isoforms due to alternative splicing events.

Beschreibung:   This MAb reacts with MUC1. The dominant epitope of this MAb has not yet been determined. MUC1 is a large cell surface mucin glycoprotein expressed by most glandular and ductal epithelial cells and some hematopoietic cell lineages. It is expressed on most secretory epithelium, including mammary gland and some hematopoietic cells. It is expressed abundantly in lactating mammary glands and over expressed abundantly in >90% breast carcinomas and metastases. Transgenic MUC1 has been shown to associate with all four c-erbB receptors and localize with c-erbB1 (EGFR) in lactating glands. The MUC1 gene contains seven exons and produces several different alternatively spliced variants. The major expressed form of MUC1 uses all seven exons and is a type 1 transmembrane protein with a large extracellular tandem repeat domain. The tandem repeat domain is highly O glycosylated and alterations in glycosylation have been shown in epithelial cancer cells. Antibody to EMA is useful as a pan-epithelial marker for detecting early metastatic loci of carcinoma in bone marrow or liver.

Beschreibung:   Polyadenylation of mRNA precursors is a two-step reaction that requires multiple protein factors. The first step, endonucleolytic cleavage of polyadenylation substrates, requires CstF (cleavage stimulation factor), a heterotrimer that is composed of three distinct subunits. CstF-64 contains an RNA binding domain and is responsible for the RNA binding activity of CstF. CstF-64 is expressed in all somatic cells and in pre- and postmeiotic, but not meiotic, germ cells. However, a large variant of CstF-64, called t CstF-64, is abundantly expressed in meiotic and postmeiotic cells in the testis and to a lesser extent in the brain, and promotes the germ cell pattern of polyadenylation. The gene encoding CstF-64 (designated CSTF2) maps to the X chromosome, whereas t CstF-64 is encoded by an autosomal gene. The increase in CstF-64 concentration during B cell activation switches IgM heavy chain mRNA expression from membrane-bound to secreted forms, suggesting that CstF-64 plays a key role in regulating IgM heavy chain expression during B cell differentiation.

Beschreibung:   Serine/threonine-protein kinase that plays a critical role in initiating innate immune response against foreign pathogens. Involved in Toll-like receptor (TLR) and IL-1R signaling pathways. Is rapidly recruited by MYD88 to the receptor-signaling complex upon TLR activation to form the Myddosome together with IRAK2. Phosphorylates initially IRAK1, thus stimulating the kinase activity and intensive autophosphorylation of IRAK1. Phosphorylates E3 ubiquitin ligases Pellino proteins (PELI1, PELI2 and PELI3) to promote pellino-mediated polyubiquitination of IRAK1. Then, the ubiquitin-binding domain of IKBKG/NEMO binds to polyubiquitinated IRAK1 bringing together the IRAK1-MAP3K7/TAK1-TRAF6 complex and the NEMO-IKKA-IKKB complex. In turn, MAP3K7/TAK1 activates IKKs (CHUK/IKKA and IKBKB/IKKB) leading to NF-kappa-B nuclear translocation and activation. Alternatively, phosphorylates TIRAP to promote its ubiquitination and subsequent degradation. Phosphorylates NCF1 and regulates NADPH oxidase activation after LPS stimulation suggesting a similar mechanism during microbial infections.

Beschreibung:   Fibrinogen is the main protein of blood coagulation system. It is a large protein and it consists of two identical subunits that contain three polypeptide chains: alpha, beta and gamma. All chains are connected with each other by a number of disulfide bonds. Fibrinopeptides A (1 to 16 amino acids) and B (1 to 17 amino acids) are released by thrombin from the N terminal parts of alpha and beta chains, respectively. In this way fibrinogen is converted into fibrin, which by means of polymerisation forms a fibrin clot. Fibrinogen clotting underlies pathogenesis of MI, thromboembolism and thromboses of arteries and veins, Zince fibrin is the main substrate for thrombus formation. Fibrinogen activation is also involved in pathogenesis of inflammation, tumor growth and many other diseases. The normal fibrinogen concentration in plasma is about 3 mg/ml. The elevated level of fibrinogen in patient's blood is regarded as an independent risk factor for cardiovascular diseases. An increase in blood fibrinogen concentration was shown to be a strong predictor of coronary heart disease. All these facts make fibrinogen an important parameter in the diagnosis of cardiovascular diseases.

Beschreibung:   Pbx 1, 2, 3 and 4 are members of the TALE (three amino acid loop extension) family of homeodomain-containing proteins. Human pre-B cell acute leukemias are frequently associated with a t(1;19)(q23;p13.3) chromosomal rearrangement, which creates a chimeric gene encoding a fusion between the E2A and Pbx 1 gene products. Pbx 2 and Pbx 3 share 92% and 94% respective identities with Pbx 1 over a 266 amino acid region flanking their homeobox domains, while all three proteins are quite divergent at their amino- and carboxy-termini. Two forms of Pbx 1 and Pbx 3 each differ primarily in their carboxy-termini and result from alternative mRNA splicing. Unlike other homeotic selector genes which are expressed transiently during development and differentiation, Pbx gene transcripts are ubiquitously expressed in both fetal and adult tissues and cell lines. Additionally, Pbx 2 and Pbx 3 transcripts are detected in lymphoid cells, which do not express Pbx 1. Pbx 4 expression is confined to the testis, especially to spermatocytes in the pachytene stage of the first meiotic prophase.

Beschreibung:   C12orf23 (chromosome 12 open reading frame 23), also known as FLJ11721, FLJ13959 or MGC17943, is a 116 amino acid multi-pass membrane protein belonging to the UPF0444 family. C12orf23 is encoded by a gene located on human chromosome 12, which encodes over 1,100 genes and comprises approximately 4.5% of the human genome. Chromosome 12 is associated with a number of skeletal deformities, including hypochondrogenesis, achondrogenesis and Kniest dysplasia. Noonan syndrome, which includes heart and facial developmental defects among the primary symptoms, is caused by a mutant form of PTPN11 gene product, SH-PTP2. Chromosome 12 is also home to a homeobox gene cluster which encodes crucial transcription factors for morphogenesis, and the natural killer complex gene cluster encoding C-type lectin proteins which mediate the NK cell response to MHC I interaction. Trisomy 12p leads to facial development defects, seizure disorders and a host of other symptoms varying in severity depending on the extent of mosaicism and is most severe in cases of complete trisomy.

Beschreibung:   This MAb is specific to heavy chain of IgA and shows minimal cross-reaction with heavy chains of other immunoglobulins. It is reactive with all subclasses of Alpha heavy chain. Immunoglobulins are four-chain, Y-shaped, monomeric structures comprised of two identical heavy chains and two identical light chains held together through inter-chain disulfide bonds. The chains form two domains, the Fab (antigen binding) fragment and the Fc (constant) fragment. Immunoglobulin A (IgA) is the main protein of the mucosal immune system. It is generated by B-cells in gut-associated lymphoid tissues. Daily production of IgA exceeds that of any of the other immunoglobulins.IgA exists mainly in dimers but can also exist as polymers or as monomers. Dimers and polymers contain a joining (J) chain that can be bound by the polymeric immunoglobulin receptor (pIgR) for transportation of the molecule to mucosal surfaces. The most common feature of plasmacytomas, and certain non-Hodgkin's lymphomas is the restricted expression of a single heavy chain class. Demonstration of clonality in lymphoid infiltrates indicates that the infiltrate is clonal and therefore malignant.

Beschreibung:   Sox-30 encodes a member of the Sox (SRY-related HMG-box) family of transcription factors involved in the regulation of embryonic development and in the determination of the cell fate. Sox-30 is expressed in testis and may act as a transcriptional regulator after forming a protein complex with other proteins. Sox-30 may be involved in the differentiation of developing male germ cells. Two transcript variants encoding distinct isoforms have been identified for the human Sox-30 gene. Sox family transcription factors influence cell differentiation, development and sex determination. Sox-30 contains a unique DNA binding domain, known as the high mobility group (HMG) box, that is related to that of the testis determining gene, SRY. The highly complex group of Sox genes cluster at a minimum of 40 different loci that rapidly diverged in various animal lineages. Several Sox genes have been identified, and members of this family have been shown to be conserved during evolution and to play key roles during animal development.

Beschreibung:   Component of a heterodimeric G-protein coupled receptor for GABA, formed by GABBR1 and GABBR2. Within the heterodimeric GABA receptor, only GABBR1 seems to bind agonists, while GABBR2 mediates coupling to G proteins. Ligand binding causes a conformation change that triggers signaling via guanine nucleotide-binding proteins (G proteins) and modulates the activity of down-stream effectors, such as adenylate cyclase. Signaling inhibits adenylate cyclase, stimulates phospholipase A2, activates potassium channels, inactivates voltage-dependent calcium-channels and modulates inositol phospholipid hydrolysis. Plays a critical role in the fine-tuning of inhibitory synaptic transmission. Pre-synaptic GABA receptor inhibits neurotransmitter release by down-regulating high-voltage activated calcium channels, whereas postsynaptic GABA receptor decreases neuronal excitability by activating a prominent inwardly rectifying potassium (Kir) conductance that underlies the late inhibitory postsynaptic potentials. Not only implicated in synaptic inhibition but also in hippocampal long-term potentiation, slow wave sleep, muscle relaxation and antinociception.
