The retinal pigment epithelium (RPE) is a monolayer simple epithelium in proximity to the outer surface of the retinal photoreceptor cells. Retinal pigment epithelium-specific protein (RPE65) is a 65 kDa protein belonging to the carotene dioxygenase family. This protein is important in 11-cis retinal production as well as in visual pigment regeneration. RPE65 is attached to the membrane by a lipid anchor when palmitoylated (membrane form) and soluble when unpalmitoylated. The soluble form of the protein binds vitamin A. Defects in RPE65 causes autosomal dominant retinitis pigmentosa and/or Leber congenital amaurosis type 2.

The exosome is a multisubunit complex of 3? to 5? exoribonucleases. It is involved in a variety of cellular processes and is responsible for degrading unstable mRNAs that contain AU-rich elements in their untranslated 3? region. EXOSC7 (exosome component 7), also known as p8, EAP1, RRP42 (Ribosomal RNA-processing protein 42), Rrp42p or hRrp42p, is a component of the exosome multienzyme ribonuclease complex. It belongs to the RNase PH family and localizes to the nucleolus. EXOSC7 is one of the six RNase-PH domain subunits of the exosome. Together, these six subunits form a PNPase-like ring. EXOSC7 is required for the processing of the 7S pre-RNA.

DACH1 is similar to the Drosophila melanogaster dachshund gene, which encodes a nuclear factor essential for determining cell fates in the eye, leg, and nervous system of the fly. It is a member of the Ski gene family and is thought to be involved in eye development. Four alternatively spliced transcripts encoding different isoforms have been described for this gene. (from Entrez gene). DACH1 has also been found to inhibit oncogene-mediated breast tumor growth via downregulation of cyclin D1. Expression of DACH1 has been found to inversely correlate with metastasis.

Derlin proteins are multi-pass membrane proteins localizing to the ER. Derlins are involved in transferring misfolded proteins from the ER to the cytosol, where the misfolded proteins are destroyed in an ubiquitin-dependent manner by the proteasome. In the case of cytomegalovirus infection, Derlin-1, as opposed to Derlins-2 and -3, is involved in the export of MHC class I heavy chains from the ER via its interaction with the viral protein US11. Derlins may also be important for cell proliferation.

LIN-28 is a highly conserved, RNA-binding, cytoplasmic protein. It consists of a cold shock domain and retroviral-type (CCHC) zinc finger motifs that were first identified in Caenorhabditis elegans. LIN-28 controls the timing of events during embryonic development and is readily expressed in embryos, embryonic stem cells and embryonal carcinoma cells. The presence of LIN-28 persists in some adult tissues including cardiac and skeletal muscle. In differentiating myoblasts, LIN-28 increases protein synthesis efficiency and binds to the growth and differentiation factor IGF-II.

P2Y11 is a multi-pass cell membrane protein that acts as a receptor for both ATP and ATD coupled to G proteins. Due to these interactions, P2Y11 is involved in phosphatidylinositol-calcium and adenylyl cyclase pathways. Induced by DMSO and retinoic acid, P2Y11 is highly expressed in spleen tissue. A putative trans-splicing event involving the gene that encodes P2Y11 and an upstream gene encoding PPAN has been found to result in a fusion protein, designated PPAN-P2RY11.

SUMF1 is a 374 amino acid alternatively spliced protein that localizes to the lumen of the endoplasmic reticulum and belongs to the sulfatase-modifying factor family. Expressed ubiquitously with highest expression in liver, kidney and pancreas, SUMF1 exists as either a monomer, a homodimer or a heterodimer (with SUMF2) and functions to oxidize sulfatase cysteine residues to an active FGIy residue, thereby playing an important role in sulfatase activity. Defects in the gene encoding SUMF1 are the cause of multiple sulfatase deficiency (MSD), a heterogeneous disorder characterized by metachromatic leukodystrophy, mucopolysaccharidosis, chondrodysplasia punctata, hydrocephalus, ichthyosis, neurologic deterioration and developmental delay.

It is characterized by a conserved core domain, which binds to phospholipids in a Ca2+-dependent manner, and a unique amino terminal region, which may confer binding specificity. Two forms of Annexin XI, designated A and B, have been identified. Transfection of COS-7 cells with Annexin XI-A, but not Annexin XI-B, causes formation of Annexin XI-associated vesicles.

It shuttles electrons between primary flavoprotein dehydrogenases and the membrane-bound electron transfer flavoprotein ubiquinone oxidoreductase. Defects in electron-transfer-flavoprotein have been implicated in type II glutaricaciduria in which multiple acyl-CoA dehydrogenase deficiencies result in large excretion of glutaric, lactic, ethylmalonic, butyric, isobutyric, 2-methyl-butyric, and isovaleric acids.
