DNA repair enzyme that can remove a variety of covalent adducts from DNA through hydrolysis of a 5'-phosphodiester bond, giving rise to DNA with a free 5' phosphate. Catalyzes the hydrolysis of dead-end complexes between DNA and the topoisomerase 2 (TOP2) active site tyrosine residue. Hydrolyzes 5'-phosphoglycolates on protruding 5' ends on DNA double-strand breaks (DSBs) due to DNA damage by radiation and free radicals. The 5'-tyrosyl DNA phosphodiesterase activity can enable the repair of TOP2-induced DSBs without the need for nuclease activity, creating a 'clean' DSB with 5'-phosphate termini that are ready for ligation. Has also 3'-tyrosyl DNA phosphodiesterase activity, but less efficiently and much slower than TDP1. May also act as a negative regulator of ETS1 and may inhibit nuclear factor-kappa-B activation.

Stanniocalcin 1 (STC1) and stanniocalcin 2 (STC2) are mammalian peptide hormones that were previously considered to be present only in bony fish, where they are involved in calcium homeostasis. STC1 plays a role in calcium and phosphate homoeostasis and is phosphorylated in vitro by protein kinase C, and STC2 is phosphorylated in vitro by casein kinase II (CK2). A human fibrosarcoma cell line, HT1080, expresses both STC1 and STC2 as secreted phosphoproteins in vivo, with STC2 being phosphorylated by an ecto-CK2-like enzyme. STC1 and STC2 have opposite effects on calcium and phosphate homeostasis, namely anti-hypercalcemic and anti-hypocalcemic actions, respectively. STC1 and STC2 are detected in human adrenal tumors, such as pheochromocytoma, differentiated neuroblastoma aldosterone-producing adenoma, and in cultured adrenal tumor cells (rat pheochromocytoma PC-12 cells and human neuroblastoma NB-1 cells).

PIPKH, also known as PIP5KL1 (phosphatidylinositol-4-phosphate 5-kinase-like 1), is a 394 amino acid phosphoinositide kinase-like protein that contains one PIPK domain. Although PIPKH lacks intrinsic lipid kinase activity, it associates with type I PIPKs and may play a role in localization of PIPK activity. Encoded by a gene that maps to human chromosome 9q34.11, PIPKH localizes to cytoplasm, specifically to large cytoplasmic vesicular structures, and exists as two alternatively spliced isoforms. Highly expressed in brain and testis, PIPKH is also expressed at very low levels in heart, spleen, lung, liver, skeletal muscle and kidney. PIPKH heterodimerizes with other type I phosphatidylinositol-4-phosphate 5-kinases, and may function as a scaffold to localize and regulate kinases to specific cell compartments. Overexpression of PIPKH may suppress cell proliferation and migration in human gastric cancer cells and may also inhibit cervical cancer formation.
