PDZD9 (PDZ domain containing 9) is a 264 amino acid protein that contains one PDZ (DHR) domain and participates in protein binding. Conserved in chimpanzee, dog, cow, mouse and rat, PDZD9 exists as two alternatively spliced isoforms and is encoded by a gene that maps to human chromosome 16p12.1. Chromosome 16 encodes over 900 genes, approximately 90 million base pairs, makes up nearly 3% of human cellular DNA and is associated with a variety of genetic disorders. Giant axonal neuropathy, a nervous system disorder characterized by increasing malfunction with growth, and the rare disorder Rubinstein-Taybi syndrome, characterized by mental retardation and predisposition to tumor growth and white blood cell neoplasias, are associated with chromosome 16. Crohn's disease, a gastrointestinal inflammatory condition, and systemic lupus erythematosis are also associated with chromosome 16.

A gene of chromosome 6q23 encodes the 640 amino acid protein, EYA4 (eyes absent) (1). EYA is one of four members of the eyes absent family (1). A 271 amino acid domain at the carboxyl terminal is highly conserved amongst the members of the eyes absent family (1). EYA4 is expressed in the craniofacial mesenchyme, the dermamyotome, and the limb (1). The conserved region in other EYA proteins interacts with SIX, DACH, and G-proteins, which regulate transcription in early embryonic development (1,2,3,4). SIX translocates EYA1-3 to the nucleus, and G-proteins can stop this interaction (3,4). Premature stop codon mutations in EYA4 cause postlingual, progressive autosomal dominant hearing loss in humans (2). This shows that EYA4 is also vital to the mature organ of Corti (2). EYA4 may cause oculo-dento-digital syndrome, based on its expression pattern and map postion (1).

One well characterized family of helix-loop-helix (HLH) proteins known to play an important role in the regulation of muscle cell development includes Myo D, myogenin, Myf-5 and Myf-6 (also designated MRF-4 or herculin). Myo D transcription factors form heterodimers with products of a more widely expressed family of bHLH genes, the E family, which consists of at least three distinct genes: E2A, IF2 and HEB. Myo D-E heterodimers bind avidly to consensus (CANNTG) E box target sites that are functionally important elements in the upstream regulatory sequences of many muscle-specific terminal differentiation genes. It acts through at least two types of receptors. This gene encodes the type 1 receptor which is thought to mediate the major cardiovascular effects of angiotensin II. This gene may play a role in the generation of reperfusion arrhythmias following restoration of blood flow to ischemic or infarcted myocardium. It was previously thought that a related gene, denoted as AGTR1B, existed; however, it is now believed that there is only one type 1 receptor gene in humans. At least five transcript variants have been described for this gene. Additional variants have been described but their full-length nature has not been determined.
