JMJD2B is a 1,064 amino acid protein encoded by the human gene JMJD2B. JMJD2B belongs to the JMJD2B histone demethylase family and contains one JmjC domain, one JmjN domain, two PHD-type zinc fingers and two Tudor domains. The two Tudor domains recognize and bind methylated histones and have an interdigitated structure; the unusual fold is required for its ability to bind methylated histone tails. JMJD2B is a histone demethylase that specifically demethylates Lys 9 residues of Histone H3, thereby playing a role in histone code. It does not demethylate Histone H3 Lys 4, H3 Lys 27, H3 Lys 36 or H4 Lys 20, however, and is only able to demethylate trimethylated H3 Lys-9 and has weaker activity than JMJD2A, JMJD2C and JMJD2D. JMJD2B demethyl-ation of Lysine residues will generate formaldehyde and succinate. JMJD2B is a ubiquitously expressed nuclear protein.

Interleukin-36 gamma (IL-36 gamma ) is a member of the interleukin 1 cytokine family that includes three closely related genes, IL-36 alpha, beta , and gamma , formerly known as IL-1F6, F8, and F9 respectively. IL-36 alpha has been detected in both neuronal and synovial tissue, whereas IL-36 beta and IL-36 gamma are expressed in both cutaneous and mucosal epithelial cells, including the respiratory tract. IL-36 beta and IL-36 gamma stimulate proliferation, maturation and/or cytokine expression by innate immune cells (such as keratinocytes and dendritic cells), and adaptive immune cells (neutrophils and T-cells) in both humans and mice. The activity of IL-36 alpha is mediated by interleukin 1 receptor-like 2 (IL1RL2/IL1R-rp2), and is specifically inhibited by interleukin 1 family, member 5 (IL1F5/IL-1 delta). IL-36 gamma plays an important role in communicating the cell death to surrounding cells.

Reacts with a common epitope of human major histocompatibility (MHC) class II antigens, HLA-DR and DP. Human MHC class II antigens are transmembrane glycoproteins composed of an chain (36 kDa) and a chain (27 kDa). They are expressed primarily on antigen presenting cells such as B lymphocytes, monocytes, macrophages, and thymic epithelial cells and are also present on activated T lymphocytes. Human MHC class II genes are located in the HLA-D region that encodes at least six and ten chain genes. Three loci, DR, DQ and DP, encode the major expressed products of the human class II region. The human MHC class II molecules bind intracellularly processed peptides and present them to T-helper cells. They, therefore, have a critical role in the initiation of the immune response. It has been shown that some autoimmune diseases are associated with certain class II alleles.

This MAb reacts with the beta-chain of HLA-DRB1 antigen, a member of MHC class II molecules. It does not cross react with HLA-DP and HLA-DQ. HLA-DR is a heterodimeric cell surface glycoprotein comprised of a 36 kDa alpha (heavy) chain and a 28 kDa beta (light) chain. It is expressed on B-cells, activated T-cells, monocytes/macrophages, dendritic cells and other non-professional APCs. In conjunction with the CD3/TCR complex and CD4 molecules, HLA-DR is critical for efficient peptide presentation to CD4 T cells. It is an excellent histiocytic marker in paraffin sections producing intense cytoplasmic staining. True histiocytic neoplasms are similarly positive. HLA-DR antigens also occur on a variety of epithelial cells and their corresponding neoplastic counterparts. Loss of HLA-DR expression is related to tumor microenvironment and predicts adverse outcome in diffuse large B-cell lymphoma.

FBL11, also known as FBXL11 (F-box and leucine-rich repeat protein 11), CXXC8, KDM2A, JHDM1A (JmjC domain-containing histone demethylation protein 1A) or LILINA, is a 1,162 amino acid member of the F-box protein family that contains one F-box domain and localizes to the nucleus. Expressed ubiquitously with highest expression in testis, ovary and brain, FBL11 functions to demethylate the Lys-36 residue of histone H3, thereby modulating the histone code. Additionally, FBL11 is thought to promote the ubiquitination and subsequent degradation of various phosphorylated proteins. Three isoforms of FBL11 exist due to alternative splicing events.

The transcription factor NFkB is retained in the cytoplasm in an inactive form by the inhibitory protein IkB. Activation of NFkB requires that IkB be phosphorylated on specific serine residues, which results in the targeted degradation of IkB (1). IkB kinase alpha (IKK alpha), previously designated CHUK (2), interacts with IkB-alpha and specifically phosphorylates IkB-alpha on the sites that trigger its degradation, serines 32 and 36 (3). IKKalpha appears to be critical for NFkB activation in response to proinflammatory cytokines (4). Phosphorylation of the IkB by IKK alpha is stimulated by the NFkB inducing kinase (NIK), which itself is a central regulator for NFkB activation in response to TNF and IL-1 (5). The functional IKK complex contains three subunits, IKK alpha, IKK beta and IKK gamma (also designated NEMO), and each appears to make essential contributions to IkB phosphorylation (6). IKAP (IKK-complex-associated protein) is a protein that acts as a scaffold, interacting with NIK, IKK alpha and IKK beta and assembling them into an active kinase complex (7,8)
