Recognizes the L1 or Calprotectin molecule, an intra-cytoplasmic antigen comprising of a 12 kDa alpha chain and a 14 kDa beta chain expressed by granulocytes, monocytes and by tissue macrophages. Macrophages usually arise from hematopoietic stem cells in the bone marrow. Under migration into tissues, the monocytes undergo further differentiation to become multifunctional tissue macrophages. They are classified into normal and inflammatory macrophages. Normal macrophages include macrophages in connective tissue (histiocytes), liver (Kupffer's cells), lung (alveolar macrophages), lymph nodes (free and fixed macrophages), spleen (free and fixed macrophages), bone marrow (fixed macrophages), serous fluids (pleural and peritoneal macrophages), skin (histiocytes, Langerhans's cell) and in other tissues. Inflammatory macrophages are present in various exudates. Macrophages are part of the innate immune system, recognizing, engulfing and destroying many potential pathogens including bacteria, pathogenic protozoa, fungi and helminthes. This MAb reacts with neutrophils, monocytes, macrophages, and squamous mucosal epithelia and has been shown as an important marker for identifying macrophages in tissue sections.

Protein kinase C (PKC) is a family of serine- and threonine-specific protein kinases that can be activated by calcium and second messenger diacylglycerol. PKC family members phosphorylate a wide variety of protein targets and are known to be involved in diverse cellular signaling pathways. PKC also serve as major receptors for phorbol esters, a class of tumor promoters. Each member of the PKC family has a specific expression profile and is believed to play distinct roles in cells. The protein encoded by this gene is one of the PKC family members. There have been at least 12 different PKC isoforms identified in humans to date including alpha, beta I, beta II, gamma, delta, epsilon, zeta, eta, theta, iota, lambda, and mu. PKC gamma is expressed solely in the brain and spinal cord and its localization is restricted to neurons. It has been demonstrated that several neuronal functions, including long term potentiation (LTP) and long term depression (LTD), specifically require this kinase. Knockout studies in mice also suggest that this kinase may be involved in neuropathic pain development. Defects in this protein have been associated with neurodegenerative disorder spinocerebellar ataxia-14 (SCA14).

There are seven 14-3-3 proteins in most mammals: beta, gamma epsilon, zeta, eta, sigma and tau. 14-3-3 sigma, also called Stratifin, is a product of the SFN gene. It is a tumor suppressor gene inactivated via DNA methylation. Reduced or no expression of 14-3-3 sigma is seen in various carcinoma including skin, breast and prostate, suggesting that 14-3-3 gene silencing may be involved in tumor progression.

Calprotectin comprises 60% of the cytoplasmic protein fraction of circulating polymorphonuclear granulocytes and is also found in monocytes, macrophages and ileal tissue eosinophils. Peripheral blood monocytes carry the antigen extra- and intracellularly, neutrophils only intracellularly. Calprotectin has antibacterial, antifungal, immunomodulating and antiproliferative effects. Besides this it is a potent chemotactic factor for neutrophils. Plasma concentrations are elevated in diseases associated with increased neutrophil activity, like inflammatory bowel disease. Granulocytes terminate their existence after transmigration through the intestinal wall. Therefore calprotectin is also detectable in feces. Elevated levels of calprotectin have been observed in body fluids such as plasma, saliva, gingival crevicular fluid, stools, and synovial fluid during infection and inflammatory conditions.This MAb reacts with neutrophils, monocytes, macrophages, and squamous mucosal epithelia and is important for identifying macrophages in tissue sections.
